Fluorescence microscopy (Falck and Hillarp technique) of 11th day old chick embryo shows aminergic neurons and processes in the paraventricular organ area. Administration of tritiated amines permits us to observe uptake and storage of serotonin only as early as the 10th day; this suggests a very early hypothalamic serotoninergic nervous system differentiation.  相似文献

Summary The DNA synthesis in glial cells during experimental brain edema following extradural compression was studied autoradiographically with H3-thymidine. During the spontaneous differentiation (day 5 to day 15 of the culture) of Caco-2 cells, the sulfation of cell layer glycosaminoglycans increased, whereas protein kinase C activity was concomitantly redistributed from the membrane to the cytosol. The protein kinase C activators, 4 beta-phorbol 12 beta-myristate, 13 alpha-acetate and 1,2-dioctanoylglycerol inhibited glycosaminoglycan sulfation. By contrast, 4 alpha-phorbol 12, 13 didecanoate was ineffective. These results suggest that membrane-bound PKC may exert a modulatory effect on glycosaminoglycan sulfation, and this effect is gradually attenuated as Caco-2 cell differentiation progresses.  相似文献

Responsiveness of free-running human circadian rhythms to a single pulse of bright light was examined in a temporal isolation unit. Bright light (5000 lx) of either 3 or 6 h duration, applied during the early subjective day, produced phase-advance shifts in both the sleep-wake cycle and the rhythm of rectal temperature; the light pulse had essentially no effect on the phase of the circadian rhythms, when it was introduced during the late subjective day or the early subjective night. The results indicate that bright light can reset the human circadian pacemaker.  相似文献

Summary The effect of a juvenile hormone mimetic, fenoxycarb, Ro 13-5223, was tested on the larval instars of the European corn borer,Ostrinia nubilalis, by dipping or topical application. When larvae were treated in instars 2, 3 or 4, the duration of the fifth instar was modified. More permanent and fewer supernumerary larvae were obtained when treatment occurred in the early instars. This non-neurotoxic compound exhibited a strong dose-dependent juvenile hormone type of activity when it was applied to last instar larvae. Fenoxycarb prevented the onset of pupation and produced supernumerary larvae and intermediates. Permanent larvae were obtained if fenoxycarb was applied on day 0 or day 1 of the last instar. The use of such a JH mimetic in the understanding of endocrine control of diapause is discussed.  相似文献

Pregnant Swiss albino mice were exposed to low doses of X-rays (approximately 9 mGy) in the range used for diagnostic exposure, on day 3.5 of gestation (preimplantation period), day 6.5 (early organogenesis period) or day 11.5 (late organogenesis period). The fetuses were examined on the 18th day of gestation. Exposure at 3.5 days post coitus (d.p.c.) resulted in a significant increase in prenatal mortality, and an increased incidence of retarded fetuses was observed after exposure at 3.5 and 6.5 d.p.c. The major effect of exposure at 11.5 d.p.c. was a significant decrease in the fetal head size and brain weight.  相似文献

Summary In the brain of the honeybee (Apis mellifica), it was found that the rRNA content decreases rapidly during the first 2 days of adult life. An increase is observed from days 3 to 5, followed by another decrease to a level inferior to that of day 2 to 3. Except for a rise on the 5th day, 4s RNA remains constant. An unidentified RNA fraction of low molecular weight (D) sharply decreases on day 5. The possibility is discussed that these results reflect changes in RNA metabolism that can be related to the sequences of activity observed in the bees during the first 9 days after hatching.  相似文献

Plasma thyroxine (T4) levels were separately ascertained in male and female embryo and young chicken from the 12th day of incubation till the 4th day after hatching by the thyroxine binding globulin technique. In both sexes, plasma T4 reach a peak the 20th day of incubation, but values are significantly higher in females. Occurrence of ovine Chorionic Gonadotropin (oCG) is demonstrated in placenta and amniotic fluid with the use of a radioreceptor assay (corpus luteum membranes) in ewes. Identification of oCG is possible as early as 15th day of pregnancy. It should be secreted at a constant rate, and its maximum concentration is recorded on the 130th day. This hormonal factor might be one of the major components accountable for high progesteronemia observed during the 100 last days of pregnancy.  相似文献

Use of enkephalinase inhibitor phosphoramidon in the in vitro radiochemical assay for juvenile hormone biosynthesis enhanced allatostatin-mediated inhibition of hormone production by corpora allata of the cockroach, Diploptera punctata. Significant increases in inhibition in day 2 virgin female CA by AST 1 (at 10(-7) M) and AST 4 (10(-8)-10(-7) M) were observed in the presence of phosphoramidon (10(-5) M or greater). No significant increase in inhibition were seen in CA from day 6 mated females with AST 4 (10(-9)-10(-7) M) and phosphoramidon combined. Phosphoramidon alone had no effect on JH biosynthesis. Analysis of allatostatin content of the CA, as determined by ELISA, revealed that addition of phosphoramidon to the medium increased the endogenous allatostatin content in CA of virgin and mated females. The similarity in primary structure between allatostatins and enkephalin-like peptides and their similar distribution makes it probable that phosphoramidon acts by preventing breakdown of allatostatins within the CA.  相似文献

The changes in the labeling index (LI) with time after a single injection of tritiated thymidine (3HTdR) at each of 4 different times of the day have been studied. Slight differences occur in the shape of these LI curves, (e.g. in the timing of the peaks) depending on the time of day when the initial injection was given. Thus, the time of day influences not only the number of cells in DNA synthesis but also determines the subsequent behavior of the labeled cells. The curves show 3 distinct peaks from which estimates of the cell cycle time can be made. The technique permits the cell cycle time to be estimated. From the data as a whole a minimum cell cycle time of 90 h for basal cells in the epidermis on the back of a mouse is obtained. The technique also provides estimates for the duration of S + G2 + M which varies depending on the time of day that the label is given. The LI curves can best be understood if the basal layer is assumed to contain 2 cell populations with differing cell cycle times; one having a long cell cycle (about 180 h) but short S-phase and containing the stem cells, the other having a short cell cycle (about 90 h) and a long S-phase duration and consisting of transit cells.  相似文献

The patterns of Glut1 and Glut3 glucose transporter protein and mRNA expression were assessed during embryogenesis of chicken brain and skeletal muscle, Glut4 protein levels were also evaluated in skeletal muscle and heart, and Glut1 was examined in the developing heart and liver. Glut1 protein expression was detectable throughout brain ontogeny but was highest during early development. Glut1 mRNA levels in the brain remained very high throughout development. Glut3 protein was highest very early and very late and mRNA was highest during the last half of development. In embryonic skeletal muscle, the levels of Glut1and Glut3 proteins and mRNA were highest very early, and declined severely by mid-development. Glut1 protein and mRNA in the heart also peaked early and then decreased steadily. Although Glut1 mRNA levels were consistently high in the embryonic liver, Glut1 protein expression was not detected. These results suggest that (1) Glut1 is developmentally regulated in chick brain, skeletal muscle, and heart, (2) Glut1 mRNA is present in liver but does not appear to be translated, (3) Glut3 in brain increases developmentally but is virtually absent in muscle, and (4) Glut4 protein and mRNA appear to be absent from chick heart and skeletal muscle.
