抗人红细胞单链抗体基因的构建及表达

从分泌抗人红细胞单克隆抗体的杂交瘤细胞３Ｆ５中提取总ＲＮＡ,逆转录成ｃＤＮＡ,用合成的寡核苷酸引物通过ＰＣＲ扩增和克隆单抗的轻、重链可变区基因,用双脱氧核苷酸链终止法分析核苷酸序列,采用ＰＣＲ拼接法构建单链抗体基因,并插入原核表达载体ＰＥｔ－２８ａ,转化大砀杆菌ＢＬ２１（ＤＥ３）,经ＩＰＴＧ诱导后表达,序 分析表明所克隆的ＶＬ、ＶＨ分别属于鼠ＩｇＫａｐｐａ轻链ＩＶ亚组和Ｉｇ重链Ⅱ亚组,ＶＨ、ＶＬ基     

简便分离鉴定酵母细胞总RNA的方法

针对酵母具有细胞壁的特点,建立了一种简便,快速,有效分离鉴定酵母细胞总ＲＮＡ的方法,并对三种便于提取酵母细胞总ＲＮＡ的方法进行分析,所得取的总ＲＮＡ质量高,可用于进行分离ｍＲＮＡ,ＰＣＲ扩增以及ｄｏｔ或Ｎｏｒｔｈｅｒｎ杂交。     

人锰超氧化物歧化酶cDNA基因的克隆

利用人胚胎肝组织提取总ＲＮＡ,从总ＲＮＡ 中分离纯化出ｍ ＲＮＡ,经逆转录得到ｃＤＮＡ 第一条链,并以之为模板和相应寡聚脱氧核苷酸为引物,进行ＰＣＲ 合成人锰超氧化物歧化酶ｃＤＮＡ 基因,将所得ｃＤＮＡ 克隆到质粒ｐＢｌｕｅｓｃｒｉｐｔＳＫ 上,转化大肠杆菌ＤＨ５α细胞,进行诱导表达筛选,将筛选的克隆进行ｃＤＮＡ 全序列测定     

BMP－3及BMP－5在不同组织和细胞中的表达

报道ＢＭＰ－３及ＢＭＰ－５在不同组织细胞中的表达．将人的神经母细胞瘤ＳＫ细胞的总ＲＮＡ及人的脑、肝、胸腺、脾、胎盘及睾丸的总ＲＮＡ反转录成ｃＤＮＡ作为模板，利用设计的编码ＢＭＰ－３和ＢＭＰ－５成熟蛋白的专一性引物分别扩增出相应的片段．ＰＣＲ产物的琼脂糖凝胶电泳结果表明，ＢＭＰ－３及ＢＭＰ－５在不同组织细胞中表达类型不同．     

一种简易的植物核酸提取方法─—从干叶和新鲜叶中快速提取RNA和DNA

本文介绍了一种植物核酸提取的简易方法．该法只需在常温下按常规的分子生物学操作条件从已干燥的和新鲜的植物叶中快速提取总ＲＮＡ和ＤＮＡ．所获得的ＲＮＡ和ＤＮＡ能顺利用于下一步的ＰＣＲ、测序及小分子ＲＮＡ研究等分子生物学实验．     

人源性抗HBc单链噬菌体抗体库的构建

利用RT－PCR和噬菌体表面展示技术，直接从乙肝病毒核心抗体（抗HBc）阳性患淋巴细胞中提取总RNA，反转录成cDNA；合成全套人抗体可变区引物扩增抗体可变区基因，并将重、轻链可变区基因进行拼接装配成单链抗体（ScFv）基因，重组于噬菌粒载体pHEN1，转化抑制型大肠杆菌E.coliTG1，以辅助噬菌体援救后，构建成人源性单链噬菌体库。库容量达106。     

小鼠4.5S RNA逆转座子cDNA的克隆及其表达载体的构建

利用ＲＴ－ＰＣＲ方法,从小鼠肝脏组织总ＲＮＡ中扩增出４．５Ｓ ＲＮＡ的ｃＤＮＡ．此ｃＤＮＡ被克隆到ｐＧＥＭ３Ｚｆ（＋）质粒,酶切鉴定并测序。然后将该序列插入以虫荧光素酶基因作了报导基因的表达载体ｐＳＶｌｕｃ２０的ＰｖｕⅡ位点,构建;了４．５Ｓ ＲＮＡ逆转座子的表达载体ｐＳＶｌｕｃ２０－４．５Ｓ,并对４．５Ｓ ＲＮＡ逆转座子的生物学功能进行了研究。     

抗牙周病厌氧菌单链抗体基因的构建与表达

从牙周病厌氧菌单克隆抗体的杂交瘤细胞株出发，通过基因工程方法－ＰＣＲ技术，调出其抗体的重链和轻链基因可变区序列，体外重组后得到单链抗体片段，克隆到了ｐＣＡＮＴＡＢ５载体上，在ＴＧ１中得到了成功表达。     

桂北五步蛇纤溶酶金属蛋白酶原基因的初步研究

从桂北五步蛇毒腺中抽提蛇毒总ＲＮＡ,采用一步法（ＲＴ－ＰＣＲ和ＰＣＲ在同一试管内进行）扩增纤溶酶金属蛋白酶原基因,并进行电泳检测,可见３条特异的ＤＮＡ条带,分别约为１．５Ｋｂ、１．３Ｋｂ和８００ｂｐ,利用平端连接的方法将其中的８００ｂｐＰＣＲ产物克隆至ｐＧＥＭ－Ｔ Ｅａｓｙ载体,挑选白色菌落,用酶切和ＰＣＲ法对其进行鉴定。     

PCR用于孕牛外周血中胎儿SRY序列检测和奶牛早期胚胎性别鉴定

根据文献报道的人，鼠，兔ＳＲＹ同源序列设计引物，从牛基因组中扩增出一段牛特异的ＳＲＹ序列，对该序列进行克隆测序后设计出一对雄牛特异的ＰＣＲ引物，利用此引物对奶牛早期胚胎细胞和妊娠晚期母牛外周血ＤＮＡ样品进行ＰＣＲ扩增，经分娩牛犊性别验证，本引物能够准确鉴别孕牛外周血中雄性胎儿的ＳＲＹ序列，并且对早期胚胎性别签定的准确率为８０％．     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
