由猪胰岛素酶促半合成人胰岛素的研究进展

本文通过查阅化学文献,收摘了1972年以来的有关由猪胰岛素促半合成人胰岛素的大量文献,这里引用了21篇,在总结文献的基础上综述了由猪胰岛素促半合成人胰岛素方法中酶的选择,反应条件的影响,苏氨酸酯的选择,以及反应历程和反应动力学的研究,认为由猪胰岛素促半合成人胰岛素的方法操作简单,生产成本相对较低,是生产人胰岛素的一种不可缺少的方法。     

酶促半合成人胰岛素的副反应及纯化

研究酶促半合成人胰岛素的副反应．采用DEAE－SephadexA－25阴离子交换层析纯化合成人胰岛素中间体衍生物,去除脱八肽胰岛素以及人胰岛素类似物,精制酶促半合成人胰岛素．     

蛋白水解酶转肽使猪胰岛素转成人胰岛素

利用赖氨酸专一性的蛋白水解酶进行了酶促人胰岛素半合成反应，猪胰岛素经该酶切去Ｂ链羧基端丙氨酸形成去丙氨酸胰岛素素ＤＡＩ。进一步在水／有机溶剂介质中该酶的转肽反应，使Ｂ链生成苏氨酸叔丁酯羧基末端，水解去酯后产生人胰岛素。研究了转肽反应的最佳条件。转肽反应生成的人胰岛素具有较强的生物活性。     

一个简便、经济的转化胰岛素基因表达产物为重组胰岛素的方法及其应用

建立了直接用胰蛋白酶酶解重组胰岛素基因表达产物的方法,并用以加工猪胰岛素前体(PIP),[B16Ala]PIP和[B26Ala]PIP,分别获得保留了各自亲本全部生物活力的(去B30)胰岛素、(去B30)[B16Ala]胰岛素和(去B30)[B26Ala]胰岛素.进一步证明胰岛素的B链C末端残基不是胰岛素生物活力所必需的.该方法比转肽方法简便、产品得率高、避免了使用有机溶剂;比胰蛋白酶/羧肽酶方法,省去了羧肽酶制剂的使用.(去B30)[B16Ala]胰岛素和(去B30)[B26Ala]胰岛素还保留了其亲本的无自身聚合能力的性质,是单体胰岛素,在临床应用中可代替原单体胰岛素,[B16Ala]胰岛素和[B26Ala]胰岛素.     

产人胰岛素毕赤酵母工程菌的构建

根据人胰岛素的氨基酸序列和酵母偏爱的密码子,采用基因半合成技术合成人胰岛素基因.将合成的胰岛素基因克隆到pP IK 9质粒,构建了毕赤(P ich ia)酵母重组表达载体pP IN S319,用B g lⅡ线性化后用电转化法导入毕赤酵母G S ll5菌株,经过营养筛选和PCR复筛,得到含人胰岛素基因的毕赤酵母工程菌株.经SDS-PAGR和密度扫描分析表明,合成的人胰岛素基因能够在毕赤酵母中高效分泌性表达,表达量可达0.563 m g/mL,表达产物无需转肽过程,只经过胰蛋白酶和羧肽酶B简单处理即得到优质重组人胰岛素,其体内活力约为30 IU/m g.     

固定化HRP海藻酸钙/壳聚糖微球催化水溶性导电聚苯胺的合成与表征

以海藻酸钙/壳聚糖微球为载体、用戊二醛共价交联固定于微球表面的辣根过氧化物酶(HRP)催化合成水溶性导电聚苯胺。考察了酶的固定化效果、负载率、比活力、贮存稳定性及酶促聚合特性,并对酶促聚合产物进行了表征。结果表明:共价交联微球固定化HRP可有效地用于水溶性导电聚苯胺的模板导向合成。     

人胰激肽原酶在毕赤酵母中的分泌表达

人胰激肽原酶(hPK)编码基因由本实验室设计、合成,并重组构建成表达质粒pPIC9k-hPK。经电穿孔将该质粒转化毕赤酵母GS115 (His^-Mut⁺) ,筛选His⁺Mut^s 型高拷贝菌株,摇瓶诱导表达重组hPK。经SDS-PAGE凝胶电泳分析确定表达重组蛋白相对分子质量为37500,产量达40mg/L,质量分数占总蛋白的30%以上。初步浓缩的重组hPK经测定具有酶活性,表明其在毕赤酵母中得到了正确的表达,每mL酶活达0.717nmol/s。     

通过酶促拆分制备S(+)3-甲基-1-(2-(1-哌啶基)苯基)丁胺

以脂肪酶Novozym435为催化剂,乙酸乙酯作溶剂和酰基供体,酶促合成瑞格列奈关键中间体S(+)3-甲基-1-(2-(1-哌啶基)苯基)丁胺,总收率为17%.该酶促拆分方法较化学拆分法具有高度的对映选择性及底物的专一性,副反应少,反应条件温和,合成路线易于操作,酶经活化后可重复利用.     

耐热DNA聚合酶介导的DNA酶促自发合成

构建了无模板引物的“类ＰＣＲ体系”。该体系在适当温度保温一段时间之后，能检测到产物出现，利用热变性分析等手段，初步研究了产物的性质，发现这些产物是随机合成的具有一些特殊构型的ＤＮＡ。本文报道三种构型，两类碱基组成比例，并通过改变反应条件观察对酶促自发合成的影响，探讨耐热ＤＮＡ聚合酶的非特异聚合活性以及介导的酶促自发合成的机理和意义。     

基于猪链球菌2型烯醇化酶建立的筛查并监控细菌侵袭性感染的ELISA方法

目的探索猪链球菌2型(S.suis2)重组烯醇化酶(Enolase)蛋白的免疫学特性,建立调查S.suis2侵袭性感染的标记和方法,并用于人群及猪群中S.suis 2感染的监控和筛选有效的疫苗抗原。方法 PCR法检测enolase在各血清型菌株中的分布情况,免疫印记检测Enolase的免疫反应性,建立筛查S.suis 2感染的酶联免疫吸附试验(ELISA)方法。结果 PCR检测显示enolase在多种血清型中广泛存在,Western-blot表明重组Enolase蛋白具有较好的免疫反应性。基于Enolase建立的ELISA结果显示,感染S.suis 2的猪血清中Enolase抗体效价较高。结论基于Enolase建立的ELISA方法能有效地用于S.suis 2感染的筛查和监控。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
