Synthese von 22,26-Imino-cholestan-Derivaten

Summary 3, 16-diacetoxy-5-pregnane-20-one and the analogue pregn-5-ene derivative have been converted into (22R, 25R)-22, 26-imino-5-cholestane-3, 16-diol. The configuration at C-22 and C-25 was determined by transformation into the known (22R, 25R)-5-solanidane-3-one. Application of the Ruschig method led to (25R)-22,26-imino-5-cholest-22(N)-ene-3, 16-diol.

Synthese der Spiroaminoketal-Gruppierung von Solanum-Alkaloiden

Summary The ring-E opened dihydro derivatives of tomatidine and soladulcidine give the corresponding N-chloroamines with N-chlorosuccinimide. Treatment of these compounds with sodium methoxide yields the spiroaminoketal alkaloids, tomatidine and soladulcidine respectively, in high yields, probably via instable C=N-unsaturated intermediates which undergo spontaneous stereospecific cyclisation.

Versuche zur Synthese von Verbindungen der Steroidreihe

Summary Reduction of the ketal II of 7-keto-1,4-dimethoxy-4b-methyl-5, 6, 7, 9, 10-hexahydrophenanthrene with lithium in liquid ammonia furnishes a corresponding dihydro derivative, from which a tricyclic triketone, m.p. 121°, is obtained upon acid hydrolysis. This triketone is identical with the recently described antitrans-4b-methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10-dodecahydro-phenanthrene-1, 4, 7-trione III and thus conforms to the stereochemistry of the natural steroids.

Summary After application of cholesterol-³H in ligated abdomina of 5th larva stages of the caterpillarMamestra brassicae ecdyson-³H and ecdysteron-³H was demonstrated in the abdomina 6 days after injection. The results show that in this case the synthesis of the moulting hormones from cholesterol occurs without participation of the prothoracic glands.

Chemische Kopplung von Hexokinase aus Hefe an Sephadex

Summary Yeast hexokinase was coupled to Sephadex by means of cyanogen bromide. The activity of the fixed enzyme was 2% of that before coupling. The activity of the noncoupled part of the enzyme was also decreased to the same degree. This decrease cannot be caused by the coupling reaction itself. The Michaelis-constants and the pH-activity curve of the coupled enzyme do not differ significantly from that of the free enzyme.

Summary By use of bentonite and sodium dodecylsulfate, which bind or inhibit ribonucleases, pure RNA preparations were obtained from human bone marrow. Phenol extraction at 4 °C yielded cytoplasmic RNA, which could be separated into 3 main peaks by sucrose density gradient centrifugation. Sedimentation coefficients of these peaks were about 30S, 19S, and 5S, respectively. Re-extraction of the phenolic interphase at 65° gave a polydisperse ribonucleic acid, sedimenting between 2S and 19S. This RNA is characterized by template activity and is supposed to be biologically active m-RNA.

Summary Cobra venom contains an anaphylatoxin-forming principle. This component has been purified by gel filtration and ion exchange chromatography. It has been obtained free from proteolytic or hemolytic activity as well as from phospholipase A. It seems to be an enzyme that splits the anaphylatoxin from its inactive precursor.

Freisetzung von Kalium aus denerviertem und innerviertem Skelettmuskel durch Acetylcholin

Summary Both hind limbs of cats (one being normally innervated, the other 8–12 days after denervation) were perfused and the quantities of potassium liberated from the two limbs in response to intraarterial injections of varying amounts of acetylcholine were compared by collecting the venous outflow from both sides separately; considerably larger quantities of potassium were found to be liberated from the denervated than from the innervated muscle after equal doses of ACH; the threshold dose of ACH for eliciting a measurable liberation of potassium being much lower on the denervated side. The dose response relation between injected ACH and potassium-liberation is of a different nature for innervated and denervated muscle. The results obtained are compatible with expectations derived from the short circuit theory of the transmitter action on the neuromuscular endplate (Fatt andKatz). The reasons of the difference in the dose relations for the innervated and denervated muscle are discussed.     

Die Isolierung von Cortisol-21-sulfat aus dem Urin von Cushingpatienten vor und nach totaler Adrenalektomie

Summary Cortisol-21-sulphate has been identified in the urine of three patients with Cushing's syndrome before and after total adrenalectomy (substitution with 200 mg F orally). The identification was based upon the following data: Identical behaviour of the standard cortisol-21-sulphate and the unknowns in two different paper chromatographic systems; positive methylene blue reaction and UV-contact-photogram; identical absorption spectra in concentrated sulphuric acid; correspondence with free cortisol of the steroid moiety liberated by dioxan solvolysis and formation of adrenosterone following treatment with chromic acid. The data support evidence that cortisol-21-sulphate may originate not only from the human adrenal gland but also from extra-adrenal sources, presumably the liver.

Eine Methode zur fortlaufenden Blutentnahme aus der Vena portae von Hunden

Summary A method is described for the introduction of a PVC.-Catheter into the portal vein of dogs. The catheter is introduced into one of the mesenteric veins of the ileum and pushed through the vena mesenterica communis until the portal vein is reached. The catheter, the end of which is fixed to the skin on the outside, can be removed without any injury to the animal by just drawing it out through the skin. The method can be applied for taking blood at various intervals during experiments on absorption, measurement of blood pressure in the portal vein and its branches, for the injection of substances directly into the liver, etc., on unanaesthetized animals.
