目的:本文报告了重组人骨形态发生蛋白-2在小鼠肌肉内诱导异位成骨的生物学作用。结果:植入重组人骨形态发生蛋白-2后1天有间充质细胞向植入区内聚集;5天出现典型的软骨细胞;7天钙化开始;10天形成雏形的骨髓腔;14天骨髓腔内出现幼稚的造血细胞;21 ̄30天形成大量网状骨小梁、骨单位,在骨小梁间隙出现骨髓细胞。结论:大肠杆菌表达的重组人骨形态发生蛋白-2具有较强的诱导成骨活性。  相似文献

珊瑚/胶原/rhBMP-2复合人工骨异位诱导成骨的实验研究   总被引:1,自引:0,他引:1  
目的:弥补单纯珊瑚无骨诱导活性、骨修复能力弱等缺陷,给临床提供一种良好的骨移植替代材料。方法:将珊瑚与胶原和rhBMP-2复合,制备出珊瑚/胶原/rhBMP-2复合人工骨,将其植入大鼠背部肌肉陷窝内,以珊瑚/胶原和珊瑚/rhBMP-2复合人工骨植入作对照;取材后通过组织学方法和图像分析评价其骨诱导活性。结果:珊瑚/胶原/rhBMP-2植入后1周,在局部诱导出软骨细胞分化和软骨基质形成;4周形成含骨髓的板层骨;诱导成骨的量有明显的rhBMP-2剂量依赖性(P<0.01);而珊瑚/胶原和珊瑚/rhBMP-2植入区均无骨或软骨形成。结论:珊瑚/胶原/rhBMP-2复合人工骨具有良好的异位诱导成骨活性,是一种较理想的骨移植替代材料。  相似文献

研制MPC/CPC骨水泥纤维蛋白胶复合BMP新型人工骨的制备及人工骨修复骨缺损的性能.通过对成年山羊双侧股骨髁部分别制备1处直径10mm、深15mm松质骨缺损,制备MPC/CPC/FG/BMP(A组)、MPC/CPC/FG(B组)、CPC/FG(C组)人工骨支架材料,分别植入各组实验观察材料,分别于6周、12周进行影像学、组织学、形态计量学观察骨缺损修复情况.说明新型MPC骨水泥纤维蛋白胶复合BMP新型生物活性人工骨支架材料在促进成骨、加快磷酸钙骨水泥降解速度上优于MPC/CPC/FG、CPC/FG的人工骨,同时新型MPC骨水泥复合纤维蛋白胶的成骨和促骨水泥降解作用优于单纯CPC复合FG的人工骨.  相似文献

为了观察Wnt联合BMP诱导骨髓间充质干细胞向成骨方向分化是否具有协同作用,本研究以密度梯度离心联合贴壁法分离培养骨髓间充质干细胞;实验分组为:对照组、Wnt组、BMP-7组、Wnt和BMP-7联合诱导组;以CCK-8法检测细胞的增殖活性;定量测定碱性磷酸酶(ALP)活性;以RT-PCR技术检测特异成骨转录因子的表达情况;使Von Kossa染色法观察细胞内矿化沉积程度。结果显示:以CKK-8法测培养第7天时联合组吸光度值为0.847,Wnt组为0.739,均明显高于对照组0.408,差异具有显著性(P0.05);联合诱导组的ALP活性比值在培养第6天和第12天时分别为63.9和144.4,BMP-7组分别为40.9和104.2,均明显高于对照组的7.2和18.8,差异具有统计学意义(P0.05);培养14 d后,OCN、β-Catenin的相对灰度值联合组高于对照组,差异具有统计学意义(P0.05),而Runx2及Osterix的表达量各组间差异无统计学意义;培养3周后,以Von Kossa染色后观察到钙结节在大小和数量上联合组均大于对照组。由此可知,Wnt3a因子和BMP-7联合可促进骨髓间充质干细胞的成骨分化,且两者具有协同作用。  相似文献

为评价珊瑚/ 胶原/ 重组人骨形成蛋白2 复合人工骨( 简称复合骨) 的骨修复能力,将复合骨植入兔颅骨直径15m m 的圆形缺损,以单纯珊瑚植入作对照;术后6 周、12 周取材,通过组织学观察和计算机图像分析,评价其骨修复能力.发现复合骨植入区形成的骨组织量明显多于同期的珊瑚植入区( P< 0 .01) ;术后12 周,复合骨完全被成熟的骨组织取代,其骨修复效果明显优于单纯的珊瑚.说明复合骨具有骨传导和骨诱导活性,骨修复能力较强,是一种较为理想的新型生物性植骨材料  相似文献

目的:探讨重组骨形成蛋白-2(recombinant human bone morphogenetic protein rhBMP-2)/珊瑚人工骨复合物(复合骨)与珊瑚人工骨(珊瑚骨)在拔牙窝修复中的作用,方法:12只成年狗作为实验动物,拔除两侧上颌 第2及第3切牙,并去除牙槽窝之间的牙槽间 ,一侧随即植入复合骨,对侧植入珊瑚内作为对照,于并植骨后4,8,。12周取材,采用组织学观察及计算机图像分析方法,观察比较两种植入材料在拔牙窝内的骨修复能力及修复效果,结果:复合骨具有较强的骨修复作用,植入牙槽窝后,材料被逐渐降解吸收,新骨不断形成,12周后,植入材料完全被成熟的骨组织取代,图像分析结果显示复合骨组新生骨形成的比值明显高于珊瑚骨组,两组比较均有显著性差异(P<0.05),结论:复合骨在拔牙窝中的骨修复能力和修复效果明显优于珊瑚骨。  相似文献

成骨细胞诱导骨髓基质细胞体外成骨的初步研究   总被引:3,自引:0,他引:3  
目的:探讨在不使用细胞因子或化学药物的情况下,成骨细胞(Osteoblast,OB)与骨髓基质细胞(Bone Marrow Stromal Cells,BMSCs)混合培养时,成骨细胞提供的成骨微环境能否在体外诱导BMSCs向成骨细胞分化,并复合支架形成成熟的骨组织.研究成骨细胞诱导BMSCs有效成骨的最小比值(指成骨细胞与骨髓基质干细胞数量的比值).方法:SY别培养SD乳鼠的成骨细胞与SD大鼠的BMSCs,将成骨细胞和BMSCs以1:9、2:8、3:7、1:0的不同比例进行混合培养,通过测定第3、6、9天培养液上清中的碱性磷酸酶(ALP)的含量,研究成骨细胞促BMSCs有效成骨的最小比值.将两种细胞以该最小浓度比混匀接种于涂附Ⅰ型胶原壳聚糖材料支架上(直径9 mm,高3mm)作为混合培养组,相同终浓度的单纯成骨细胞和单纯BMSCs分别接种于相同支架作为阳性对照及阴性对照.另设置低比值成骨细胞对照组(仅含有共培养组中相同的成骨细胞数,但不含有共培养组中的BMSCs).全部标本均于体外培养8周后取材,通过大体观察、组织学及免疫组织化学等相关检测对新生骨进行评价.结果:成骨细胞和BMSCs以3:7的比例进行混合培养时已可实现有效成骨.3:7比例的混合培养组及阳性对照组(成骨细胞组)体外培养8周后大体观察和苏木素-伊红染色(HE)、ALP染色基本相同,均表达骨特异性细胞外基质Ⅰ型胶原,形成了较成熟的骨组织.阴性对照组(单纯BMSCs组)和低比值成骨细胞组,原细胞支架复合物变小、变形.低比值成骨细胞组在局部形成了少量的骨组织,阴性对照组(单纯BMSCs组)未能发现骨样组织形成.结论:在不使用细胞因子或化学药物的情况下,成骨细胞提供的成骨微环境能够在体外诱导BMSCs向成骨细胞分化并形成成熟的骨组织.混合细胞中成骨细胞与BMSCs的比例为3:7时是有效成骨的最小比值.  相似文献

目的:检测富血小板纤维蛋白提取液(PRFe)对人牙周膜细胞(h PDLCs)增殖及成骨分化能力的影响.方法:组织块法分离培养h PDLCs,免疫组化鉴定来源;Choukroun法制取PRFe;MTT法检测不同体积分数的PRFe对h PDLCs增殖活性的影响以确定最适宜体积分数的PRFe.实验分为空白对照组和PRFe组.碱性磷酸酶试剂盒检测ALP活性;茜素红染色观察细胞矿化功能;Western Blotting检测细胞中成骨蛋白BMP2和Runx2的表达.结果:体积分数为50%的PRFe吸光度值高于其他实验组(P0.05).选择体积分数为50%的PRFe用于后续实验,ALP活性检测、茜素红染色结果显示PRFe组的检测指标随着时间延长上升幅度均显著大于空白对照组,具有统计学差异(P0.05),PRFe组ALP活性与矿化结节积分光密度值于各时间点均高于空白对照组,具有统计学差异(P0.05).Western Blotting结果显示PRFe组成骨蛋白表达强于对照组,具有统计学差异(P0.05).结论:PRFe具有促进体外h PDLCs增殖以及成骨分化的作用.  相似文献

骨形态发生蛋白15(Bone Morphogenetic Portein-15,BMP15)由卵母细胞特异表达,在卵母细胞发育与成熟过程中起重要调控作用.本文以长白猪为研究模型,克隆BMP15基因cDNA全长并探究该基因组织表达图谱.研究显示,家猪BMP15基因cDNA全长1298bp,编码具402个氨基酸的前体蛋白.依赖RT-PCR技术,家猪Bmp15基因被发现特征性表达于家猪卵巢中.在本研究中,还分离到家猪BMP15基因潜在启动子区(3279bp),针对该启动子区,部分转录因子结合位点获得预测.这些研究结果为探究BMP15基因的表达调控机制及其对家猪卵泡发育影响奠定基础.  相似文献

目的:本研究初步探讨脱钙牙基质(DDM)在牙槽嵴窝中植入应用的可能性。方法:杂种犬6只,随机分为3组拔除所有的前磨牙,在一侧上颌前磨牙区植入DDM,另一侧作为对照组。在术后4、9、14周依次处死2只动物,所有标本都经大体观察、放射线片连续检查、组织学观察、电镜观察。结果:DDM植入前后牙槽骨高度和宽度与非植入DDM组分别减少0.35mm,0.40mm和1.60mm,1.54mm,差异具有统计学意义。14周后,DDM已基本吸收形成新骨。结论:DDM具有良好的骨诱导活性,能够促进牙槽窝的愈合。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献
