泥蚶(TegillarcagranosaLinnaeus)凝集素能凝集所测试的14种人或动物的红细胞,其中对兔的红细跑的凝集活性最高.泥蚶凝集素也能凝集大鼠和小鼠的肺、脾脏、肝脏、骨髓细胞以及小鼠精子细胞.该凝集素对兔的红细胞的凝集活性不被测试的10种浓度为200mmol·L-1的糖所抑制.  相似文献

日本对虾血清和肌肉提取液凝集活力初步研究   总被引:7,自引:0,他引:7  
用12种天然或经酶修饰的动物及人红细胞测定日本对虾的血清和肌肉提取液的凝集活力,并且进行糖抑制、热稳定性、pH及Ca^2 影响试验。结果显示:血清和肌肉提取液均能凝集12种红细胞,但两者对每种红细胞的凝集效价各不相同。血清对鹤鹑红细胞的凝集作用可被蔗糖抑制,最小抑制浓度为200mmol/L,其凝集活性依赖于Ca^2 ,在pH4.0-7.0范围内较稳定,经90℃保温10min保持最高活性;肌肉提取液的凝集活性不被7种糖抑制,在pH6.0-7.0,温度为4-80℃范围内均具有最高活性,其凝集活性不依赖于Ca^2 。由以上结果推测:血清与肌肉提取液均含有凝集素,且种类可能不同。  相似文献

研究结果表明,杂色鲍血清中自然存在凝集素,该凝集素能够识别和凝集多种不同的细菌和人及动物的红细胞,其中对人体O型和B型血细胞的凝集效价显著高于A型和AB型血细胞(P<0.05);Ca2+能促进其凝集活性,而Mg2+则抑制其凝集活性;凝集作用不同程度地受反应介质的pH、离子强度及反应时间等因素的影响;该凝集素不被透析,在95℃、pH2.0~10.0时仍保持凝集活性,抗RNase、DNase、SDS和乙醚,对蛋白酶K、糖氧化剂NaIO4敏感;凝集素活性能被D-树胶醛糖、L-鼠李糖特异性抑制,D-果糖、D-木糖对其也有显著的抑制作用.  相似文献

对同型巴蜗牛(Bradybaena similaris)蛋白腺提取液凝集活性进行测定,初步提取其凝集素并进行部分性质鉴定.结果表明:在同型巴蜗牛的蛋白腺中存在凝集素,具有血球凝集活性,能够凝集9种动物红细胞,其中,对家兔红细胞的凝集活性最高,可达28,不过,其对家兔红细胞的凝集活性明显依赖于Ca2+;在pH值为7.0~8.0的范围内其凝集活性较稳定,保持100%的凝集活力;温度为40℃持续作用10 min后就失去活性;向同型巴蜗牛蛋白腺提取液加入45%的固体硫酸铵使凝集素沉淀,再用不同饱和度的硫酸铵溶液依次对沉淀物进行抽提,抽提物的比活力提高了约2.56倍,总活力回收为20.00%.  相似文献

利用血凝试验测定西施舌(Coelomactra antiquata)血清和肌肉提取液对8种红细胞的凝集活性,同时进行热稳定性、糖抑制和pH影响试验.结果表明,西施舌的血清和肌肉提取液均能凝集试验所用的8种红细胞,但肌肉提取液的凝集效价高于血清(人O型血除外).二者的热稳定性均较高.血清对草鱼红细胞的凝集作用可被试验用的7种糖抑制,而肌肉提取液则不被7种糖抑制.二者在pH 4.0-10.0的范围内均具有凝集活性.试验结果表明,西施舌血清和肌肉提取液均含有凝集素,但种类不同.  相似文献

文昌鱼体表粘液经卵粘蛋白 -Sepharose 6B亲和层析法分离出一种凝集素 (AmphioxusMucusLectin ,简称AML) .该凝集素能凝集兔及人的红血细胞 ,无血型专一性 ;其凝血活性不依赖于Ca2 和Mg2 ,在 pH6-1 0范围较稳定 ,经 60℃保温 1 0min活性则完全消失 ;SDS PAGE测得其亚基分子量为93 0 0 0 ;含糖量为 2 6% .文昌鱼凝集素在文昌鱼的免疫防卫中可能起着重要作用 .  相似文献

通过液体摇瓶培养蒙古口蘑菌丝体,用磷酸盐缓冲液浸提,得到菌丝体的凝集素粗提物.分析表明:蒙古口蘑菌丝体凝集素对牛血和羊血都能凝集;D-果糖、β-葡萄糖、半乳糖和木糖对菌丝体凝集素均具有抑制作用;弱酸或弱碱性浸提液有利于凝集素的提取;菌丝体凝集素具有一定的热稳定性,直到60℃以后凝集红细胞的活力才丧失;其凝集活性对Ca2+、Mg2+、Mn2+和Fe3+这四种离子有不同程度的依赖.  相似文献

在两种淡水育珠蚌(三角帆蚌和背角无齿蚌)血清中发现有凝集素存在,三角帆蚌的血清凝集素能凝集鸡、鸭和兔的红细胞,在pH5~9的范围内保持活性, 凝集活性能被甘露糖抑制,对Ca^2+没有表现出明显的依赖性,背角无齿蚌的血清凝集素能凝集鸡和鸭的红细胞,但不能凝集兔的红细胞,在pH6~9的范围内保持活性,其凝集活性对Ca^2+有明显的依赖性,而且可被N-乙酰葡萄糖胺强烈抑制,两者的血清经60℃加热处理30  相似文献

将分别从狗脊蕨、扇叶铁线蕨叶片组织中提取得到的凝集素进行细胞凝集和抑菌活性比较.采用倍比稀释法在"V"型微量血凝板上进行凝集活性测定,采用琼脂平板法测定抑菌效果.结果发现二种凝集素均能有选择地凝集不同来源的动物血红细胞.狗脊蕨凝集素对鲫鱼红细胞的凝集活性最高达到29,扇叶铁线蕨凝集素对鸡红细胞的凝集活性最高达到210,对微藻细胞、细菌、酵母菌和肿瘤细胞的凝集具有特异性,对二种植物病原菌及细菌的抑制活性存在特异性.  相似文献

17种蕨类植物叶组织经磷酸缓冲液抽提,硫酸铵沉淀,得到蛋白质提取液,用不同类型的红细胞检测凝集活性,实验结果表明,有12种蕨类植物的提取物中含有凝集素,能凝集一种或多种类型的红细胞,不同种类对红细胞的凝集效应不同,产生凝集作用所需的最低蛋白质浓度相差很大,从0.8μg/ml到1100.0μg/ml,在供试的红细胞中,鸽子红细胞对蕨类植物集素提取物最敏感,某些种类提取物与兔红细胞的凝集活性可分别被1种或多种类型糖类所抑制,多数种类凝集素提取物能够耐高温处理(90-100℃),这可能与蕨类植物凝集素的某些化学性质有关。  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
