采用荧光光谱法和吸收光谱法研究了1-(2-吡啶偶氮)-2-萘酚-磺酸(PAN-S)与牛血清白蛋白(BSA)的结合反应.研究表明,PAN-S对BSA的荧光猝灭机理为静态猝灭.测定了反应的结合常数(K=5.32×105~8.53×105L·mol-1)、荧光猝灭常数(Ksv=7.08×105L·mol-1)及结合位点数(n=2.88).进一步通过同步荧光法研究了PAN-S对BSA构象的影响,认为PAN-S主要结合在BSA的色氨酸残基附近.研究发现,PAN-S与BSA之间存在着类似于其同阳离子表面活性剂的静电作用.  相似文献

解析3种黄酮类小分子桑色素(MOR)、槲皮素(QUE)、芦丁(RUT)与牛血清白蛋白(BSA)相互作用后的紫外-可见光谱和荧光猝灭光谱,研究在同一实验条件下黄酮类小分子与BSA的相互作用及其构效关系。结果显示,黄酮母核B环上的羟基处于相邻位置时,比处于其他位置时表现出的BSA猝灭作用和结合作用更强,而有较大取代基团存在时,由于位阻较大,黄酮类小分子与BSA间的猝灭作用及结合作用则明显变弱。  相似文献

合成了二氢杨梅素铜配合物(DMY-Cu),通过红外光谱、原子吸收法对其进行表征,并采用荧光光谱法研究了二氢杨梅素铜配合物与牛血清白蛋白(BSA)的相互作用.结果表明,DMY-Cu的组成为[C15H10O8Cu·H2O]n.激发波长为288nm时,BSA的发射峰位于345nm,DMY-Cu对BSA有较强的荧光猝灭作用,由温度对DMY-Cu与BSA体系荧光猝灭速率的影响和动态猝灭常数(KSU)以及静态猝灭结合常数(KA)的计算得出,DMY-Cu对BSA内源荧光的猝灭机制属于形成BSA-DMY-Cu复合物的静态猝灭,荧光猝灭常数为1.33×105L·mol-1;由反应前后热力学函数△Hθ<0,△Sθ>0推出DMY-Cu与BSA之间的作用力主要是静电作用力,在BSA分子上荧光敏感部位有1个结合位点,结合常数为4.95×105.  相似文献

应用荧光光谱法研究了水溶液体系中,苯酚磺酞类酸性染料苯酚红、甲酚红、氯酚红、溴酚红、甲酚紫与牛血清白蛋白(BSA)之间的相互作用.实验表明:苯酚磺酞类酸性染料对牛血清白蛋白的荧光有较强的猝灭作用,其荧光猝灭主要为静态猝灭,从荧光猝灭结果中求得不同温度下各种染料与BSA的结合常数K,发现染料取代基的引入使K值增大,且随反应温度上升K值下降.由染料与BSA反应焓变、熵变,确定染料与BSA的结合主要是静电引力.依据非辐射能量转移机理,探讨了不同温度下该类染料与BSA相互结合时,其给体-受体间距离和能量转移效率,进一步证实了该类反应为单一静态猝灭过程,且阐明了其猝灭机理是通过能量转移产生的.  相似文献

应用荧光法研究了姜黄素钐配合物与牛血清白蛋白(BSA)的结合反应.根据配合物对BSA的荧光猝灭效应,利用变温试验,求得相互作用的结合常数、结合位点数及热力学参数,由此推测猝灭机理和结合力类型;依据Forster能量转移理论计算结合距离;并利用探针进行结合点定位;用同步荧光光谱确定配合物对牛血清白蛋白结构的影响.结果表明,姜黄素钐配合物对BSA的内源荧光猝灭主要以静态方式,同时发生非辐射能量转移;由△Hθ和△sθ〈0推测范德华力与氢键是主要结合力;配合物在BSA分子的SiteⅡ位结合,结合距离为2.4nm;配合物改变了BSA分子构像,对色氨酸残基影响更大.  相似文献

利用紫外光谱法、 荧光光谱法研究了美他环素(METC)与牛血清白蛋白(BSA)相互作用的机理; 考察了温度、 溶液酸碱度、 金属离子对METC与BSA相互作用的影响. 实验结果表明, METC对BSA的猝灭属于静态猝灭过程; 它们的结合主要以静电引力为主, 根据Frster非辐射能量转移原理计算了METC与BSA的结合距离; 金属配合物对BSA的猝灭属于静态猝灭过程, 但结合常数相对于在常温时METC直接猝灭BSA的结合常数有很大差异, 而且结合位点数也有相应的改变, 表明金属离子的介入对METC的结构及疏水性能都有不同程度的影响.  相似文献

利用荧光光谱法、紫外光谱法、荧光寿命和圆二色光谱法等方法研究了6-糠氨基嘌呤(KT)与牛血清白蛋白(BSA)之间相互作用机理.结果表明,KT可以显著猝灭BSA的内源荧光,其猝灭机制为静态猝灭,猝灭常数Ksv在288 K和303 K时分别为1.45×104和1.42×104L/mol,Stern-Volmer曲线是两段相交于cKT=8.0×10-5mol/L的回归曲线,说明KT与BSA之间存在两类结合位点.数据处理及热力学参数计算表明:低浓度KT-BSA(cKT8.0×10-5mol/L)的结合主要是疏水作用,结合位点数约为1;较高浓度KT(cKT8.0×10-5mol/L)主要以氢键、范德华力与BSA结合,结合位点数为3.7.紫外光谱和圆二色光谱实验结果表明,KT的存在使BSA二级结构发生了改变.  相似文献

应用荧光光谱法研究了生理条件下吡罗昔康对牛血清白蛋白,Cu(Ⅱ)对牛血清白蛋白以及Cu(Ⅱ)对吡罗昔康和牛血清白蛋白荧光光谱特性的影响.结果表明:Cu(Ⅱ)和吡罗昔康均可使牛血清白蛋白的荧光强度发生静态猝灭,并且在Cu(Ⅱ)存在下,吡罗昔康对牛血清白蛋白的荧光猝灭作用显著增强.根据荧光猝灭双倒数图计算吡罗昔康和牛血清白蛋白的结合常数为3.18×103 L/mol,结合位点数为0.75;二元配合物Cu(Ⅱ)与牛血清白蛋白之间的结合常数为1.13×103 L/mol,结合位点数为0.74.  相似文献

目的:运用荧光猝灭法研究不同温度下穗花杉双黄酮(AMF)与牛血清白蛋白(BSA)的相互作用.方法:采用荧光光谱研究AMF与BSA之间的猝灭类型,计算二者之间的结合常数及结合位点数等.结果:穗花衫双黄酮能使BSA发生内源性荧光猝灭,属静态猝灭机制.在298,308,318 K下,AMF与BSA结合常数分别为6.73,6.38,6.17 L·mol-1,结合位点数n近似为1;热力学分析表明AMF与BSA之间结合力为静电力作用;同步荧光光谱表明AMF使BSA构象发生改变,且色氨酸所处微环境的疏水性增强.结论:荧光猝灭法可用于AMF与BSA之间结合反应的研究,方法简便,灵敏度高.  相似文献

应用荧光光谱法研究了三苯甲烷类染料邻氯酚红与牛血清白蛋白(BSA)的相互作用.研究表明,邻氯酚红能与牛血清白蛋白形成稳定的络合物从而引起BSA的荧光猝灭,疏水作用是结合反应的主要作用力,二者的结合常数为1.2002×106L.mol-1,结合位点数n=1.2586(17℃时),能量转移效率为E=0.3086,染料距色氨酸残基最短距离为r=4.92nm.同时考察了邻氯酚红的加入对BSA构象的影响.  相似文献

Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
