Similarities between metallothionein and low molecular weight testicular cadmium-binding protein

The incorporation of 35S-cysteine and 3H-glutamic acid was studied in mouse hepatic and renal metallothionein and in testicular cadmium-binding protein of similar molecular weight. Preferential incorporation of 35S-cysteine over 3H-glutamic acid was observed not only in hepatic and renal metallothionein, but also in testicular cadmium-binding protein. When the antigenic reactivity of these proteins was compared, all three proteins reacted with the metallothionein antibody. These similarities suggest that the low molecular weight testicular cadmium-binding protein is apparently metallothionein.     

Metallothionein induced in the earthworm

Summary One of the 3 different molecular weight cadmium-binding proteins induced in the earthworm was characterized as a metallothionein; this characterization was based on a high cysteine and cadmium content, low molecular weight, heatstability, and mercaptide bonding.M.Y. and K.T.S. thank Dr K. The enhanced induction of metallothionein by zinc,its half-life in the marine fishPleuronectes platessa,and the influence of stress factors on metallothionein levels

Summary Intraperitoneal injection of zinc raised levels of a hepatic metallothionein-like species. Assuming that this species was metallothionein (MT) then levels were raised from approximately 20 g/g to 300 g/g in 7 days, and levels thereafter remained high for the next 4 weeks. The half-lives of the protein in liver and kidney from starved fish, measured using in vivo incorporation of³⁵S cysteine at 11°C, were approximately 27 days and 32 days respectively. The following agents failed to stimulate synthesis of MT in plaice: stress (due to catching), endotoxin, dexamethasone, cortisol and turpentine.     

Incorporation of32P into renal phospholipids of mice during postnatal growth

Summary During the first 40 days of life the rate of incorporation of³²P into total phospholipids and into phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, sphingomyelin, diphosphatidylglycerol, phosphatidic acid and phosphatidylinositol of mouse kidney was by some 25–35% higher than in older animals. Results suggest a different involvement of cellular membranes during of normal and compensatory renal growth.     

The site of synthesis of hemocyanin in the crayfish,Astacus leptodactylus

Summary The in vitro incorporation of³⁵S-methionine into hemocyanin was tested in the crayfish,Astacus leptodactylus. All organs assayed (hemocytes, heart, hypodermis, midgut gland, muscle and stomach) were able to synthesize proteins under our experimental conditions, but only midgut gland (=hepatopancreas) incorporated labeled methionine into a protein which was precipitated by an antibody againstAstacus hemocyanin, and which comigrated with authentic hemocyanin on SDS gels.     

In vitro effects of methionine-enkephalin,somatostatin and insulin on cultured gonadal cells of the snailHelix aspersa

Isolated snail gonadal cells were cultured in the presence of synthetic neuropeptides in order to determine the subsequent effect of these substances on gonadal synthetic activities. Gonadal cells were incubated for 24 h in concentrations of methionine-enkephalin, somatostatin and insulin ranging from 10^–4 M to 10^–9 M, in medium 199 supplemented with 6% Ultroser G. Synthesis of DNA and protein by the cultured cells were simultaneously estimated by measuring incorporation of³H thymidine and³⁵S methionine. The rate of labelled precursor incorporation was measured using the liquid scintillation technique. All substances tested exerted a dose-dependent effect. The synthetic activity of the cultured cells was highest when the concentration of the peptides added to the medium approximated the physiological levels. Methionine-enkephalin, somatostatin and insulin at 2×10^–8 M significantly increased³H thymidine incorporation, by 62%, 69% and 69% respectively, and protein synthesis by 42%, 57% and 57%, respectively. In the case of juvenile gonadal cultured cells, a similar increase in³H and³⁵S incorporation was registered for a 10^–7 M peptide concentration. Both lower and higher peptide concentrations inhibited³H thymidine and³⁵S methionine incorporation. Pharmacological studies suggest the existence of methionine-enkephalin and somatostatin-like receptors on snail gonadal cells. These results indicate that our gonadal cell culture model provides a useful tool for the study of the neuroendocrinological control of the activity of snail gonadal cells.     

35S-Sulphate incorporation into myelin sulphated mucopolysaccharides during rat brain development

Summary Rat brain myelin acid mucopolysaccharides (AMPS) incorporate 15%, 8%, 5.5% and 4% of tojtal associated³⁵S-sulphate, 14, 21, 30 and 75 days after birth, respectively. The course of³⁵S-sulphate incorporation into total rat brain mucopolysaccharides, as well in those from myelin, had a similar feature with peak on the 2nd week and a significant decrease on the 3rd and 4th week postnatally.     

Stimulation of the incorporation of3H-leucine into proteins by oestradiol in the foetal uterus of the guinea-pig

Summary Oestradiol treatment increases the incorporation of³H-leucine into acid insoluble proteins in the foetal guineapig uterus (50–65 days of gestation) 10 times above control values by 8 h and 20 times by 24 h after administration of oestradiol to the mother.     

Influence of progesterone on protein and RNA synthesis in cultured chick embryo liver cells

Summary Chick embryo liver primary cultures, when added with progesterone, exhibit, in comparison with the controls, a normal growth, a decline of both³H-uridine uptake and incorporation into total RNA, a decline of³H-leucine and¹⁴C serine incorporation into the proteins. Progesterone is not able to stimulate phosvitin synthesis induction.These studies were supported in part by the Italian CNR grants.     

A differential effect of L-serine on the incorporation of3H-deoxycytidine and3H-thymidine into DNA of rat thymocytes

Summary The addition of L-serine to short-term cultures of rat thymocytes stimulated the incorporation of³H-deoxycytidine into DNA, but simultaneously depressed the incorporation of³H-thymidine into DNA.This investigation was supported by NIH grants No. CA-10291 and T12CA8021 from the National Cancer Institute.     

Untersuchungen über den Wirkungsmechanismus von Procarbazin (Natulan®)

Summary The cytotoxic methylhydrazine derivative procarbazine or Natulan® has a marked influence on the synthesis of deoxyribonucleic acid in Ehrlich ascites carcinoma cells. The incorporation of³H-thymidine into DNA — measured by autoradiographic methods — is inhibited.     

Effect of estradiol on aminoacid incorporation into proteins of different hypothalamic areas in prepuberal rats

Summary Estradiol in vitro produces a significant increase in the incorporation of³H-leucine into proteins of the anterior hypothalamic area in prepuberal female rats, 15 and 20 days old, but not in younger animals. The ovarian hormone induced no changes in the protein synthetic activity of middle and posterior hypothalamus and cerebral cortex in prepuberal female rats of different ages. Estradiol did not modify the protein synthesis of the hypothalamus and cerebral cortex in prepuberal male rats.Supported by a grant from the Consejo Nacional de Investigaciones Cientifícas y Técnicas de la República Argentina.     

Effects of isaxonine phosphate and analogs on fibroblast metabolism in culture

Summary Human skin fibroblasts in confluent cultures were incubated for 24 h in the presence of isaxonine phosphate (Nerfactor) and several related factors. The incorporation of¹⁴C-proline into secreted proteins and the release of collagen into the medium were inhibited. When the cells incubated for an additional period of 24 h after thorough washing, protein and collagen syntheses were found to be identical to those of controls, demonstrating that the inhibition of protein synthesis was independent of any toxic effect. When cells were incubated in the presence of both isaxonine and colchicine, the secretion of collagen was more inhibited than by colchicine alone, and proteins accumulated in the cells.

Summary After intravenous injection of a³⁵S-labelled heparinoid (N-formyl-chitosan-polysulfuric acid ester) into rabbits, a highly purified clearing factor preparation was obtained from the blood plasma. The heparinoid content of this preparation is approximately 40 times higher, weight for weight, than that of the other plasma proteins.      

Synthesis of a ubiquitously present new HSP60 family protein is enhanced by heat shock only in the Malpighian tubules ofDrosophila

A homologue of the chaperonin protein of the HSP60 family has not been shown so far inDrosophila. Using an antibody specific to HSP60 family protein in Western blotting and immunocytochemistry, we showed that a 64-kDa polypeptide, homologous to the HSP60, is constitutively present in all tissues ofDrosophila melanogaster throughout the life cycle from the freshly laid egg to all embryonic, larval and adult stages. A 64-kDa polypeptide reacting with the same antibody in Western blots is present in all species ofDrosophila examined. Using Western blotting in conjunction with³⁵S-methionine labeling of newly synthesized proteins and immuno-precipitation of the labeled proteins with HSP60-specific antibody, it was shown that synthesis of the 64-kDa homologue of HSP60 is appreciably increased by heat shock only in the Malpighian tubules, which are already known to lack the common HSPs.     

Metallothionein induced in the frogXenopus laevis

Summary A low molecular weight cadmium- and copper-binding protein, induced in the liver of the frog,Xenopus laevis, by the administration of cadmium, was shown to consist of a single isoprotein and was characterized as an amphibian metallothionein based on its high cysteine and metal content, its low molecular weight, and the lack of aromatic amino acids.The authors thank Dr K. Effect of low density lipoprotein on proteoglycan synthesis by aorta cells in culture

Summary Increasing the content of human serum low density lipoprotein in the growth medium led to greater incorporation of³⁵S-sulfate into proteoglycan (mostly into dermatan sulfate) by primary aorta cells but did not affect similar incorporation by fibroblast cells. These results suggest a mechanism which can explain the increased deposition of lipid in aorta due to hyperlipidemia.Acknowledgements. High affinity recognition of a <Emphasis Type="Italic">Phytophthora</Emphasis> protein by <Emphasis Type="Italic">Arabidopsis</Emphasis> via an RGD motif

The RGD tripeptide sequence, a cell adhesion motif present in several extracellular matrix proteins of mammalians, is involved in numerous plant processes. In plant-pathogen interactions, the RGD motif is believed to reduce plant defence responses by disrupting adhesions between the cell wall and plasma membrane. Photoaffinity cross-linking of [¹²⁵I]-azido-RGD heptapeptide in the presence of purified plasma membrane vesicles of Arabidopsis thaliana led to label incorporation into a single protein with an apparent molecular mass of 80 kDa. Incorporation could be prevented by excess RGD peptides, but also by the IPI-O protein, an RGD-containing protein secreted by the oomycete plant pathogen Phytophthora infestans. Hydrophobic cluster analysis revealed that the RGD motif of IPI-O (positions 53–56) is readily accessible for interactions. Single amino acid mutations in the RGD motif in IPI-O (of Asp⁵⁶ into Glu or Ala) resulted in the loss of protection of the 80-kDa protein from labelling. Thus, the interaction between the two proteins is mediated through RGD recognition and the 80-kDa RGD-binding protein has the characteristics of a receptor for IPI-O. The IPI-O protein also disrupted cell wall-plasma membrane adhesions in plasmolysed A. thaliana cells, whereas IPI-O proteins mutated in the RGD motif (D56A and D56E) did not.Received 23 October 2003; received after revision 5 December 2003; accepted 12 December 2003     

Aberrant,canavanyl protein formation and the ability to tolerate or utilize L-canavanine

Summary L-Canavanine, 2-amino-4-(guanidinooxy)butyric acid, and L-arginine incorporation into de novo synthesized proteins was compared in six organisms. Utilizing L-[guanidinooxy¹⁴C]canavanine and L-[guanidino¹⁴C]arginine at substrate saturation, the canavanine to arginine incorporation ratio was determined in de, novo synthesized proteins.Caryedes brasiliensis andSternechus tuberculatus, canavanine utilizing insects;Canavalia ensiformis, a canavanine storing plant; and to a lesser extentHeliothis virescens, a canavanine resistant insect, failed to accumulate significant canavanyl proteins. By contrast,Manduca sexta, a canavanine-sensitive insect, andGlycine max, a canavanine free plant, readily incorporated canavanine into newly synthesized proteins. This study supports the contention that the incorporation of canavanine into proteins in place of arginine contributes significantly to canavanine's antimetabolic properties.

Summary Mature chloroplasts ofClivia miniata etBilbergia sp. show the intensive incorporation of thymidine³H into DNA and uridine³H into RNA. On the contrary, phenylalanine³H is mostly incorporated in the proplastids.
