IGF-ⅡP3脱氧核酶对人肝癌细胞凋亡作用的影响   总被引:2,自引:1,他引:1  
目的 研究IGF-ⅡP3DRz对肝癌细胞生长的抑制作用.方法 用噻唑蓝比色法(MTT)检测DRz对肝癌细胞SMMC-7721的增殖抑制作用;流式细胞技术检测分析细胞凋亡;倒置显微镜、光镜Giemsa染色观察细胞形态.结果 DRz作用SMMC-7721细胞24 h后,细胞存活率明显下降,而且脱氧核酶的浓度增加,抑制作用增强(P<0.01);DRz可诱导SMMC-7721细胞凋亡,细胞形态结构呈现典型的凋亡特征.结论 DRz可通过阻滞细胞周期发展发挥抑制SMMC-7721细胞增殖的作用,DRz还可诱导细胞发生凋亡.  相似文献

本研究旨在探究NADPH氧化酶活化所致氧化胁迫在七叶皂苷诱导SH-SY5Y神经母细胞瘤凋亡中的重要作用.以体外培养SH-SY5Y细胞作为研究对象,采用MTT法测定细胞增殖活力,流式细胞术测定细胞周期、细胞凋亡和活性氧指标,Western Blot测定凋亡蛋白Caspase 3变化.结果表明,七叶皂苷处理致显著细胞增殖抑制(P 0. 05)、细胞内活性氧水平明显增加(P 0. 05)、细胞周期阻滞于S期(P 0. 05)、细胞凋亡数目增加、Caspase 3蛋白切割水平显著增加(P 0. 05); NADPH氧化酶抑制剂DPI预处理明显逆转七叶皂苷所致的细胞增殖抑制、活性氧增加、周期阻滞和凋亡(P 0. 05).提示NADPH氧化酶活化所致氧化胁迫参与七叶皂苷所引起SH-SY5Y细胞的凋亡.  相似文献

本文通过MTT法和流式细胞术研究橙皮苷对人食管癌Eca9706细胞增殖的抑制作用及对细胞周期和凋亡的影响,结果显示:不同浓度橙皮苷能有效抑制Eca9706细胞增殖,且呈剂量依赖性(P<0.01),IC50为84μg/mL;22、43、84μg/mL的橙皮苷作用于Eca9706细胞48h,细胞周期在S期的比例分别为25.16%、30.13%、31.05%,比对照组明显升高(P<0.002);细胞凋亡率分别为6.23%、8.19%、10.62%,与对照组相比细胞凋亡率明显增加,差异有统计学意义(P<0.05).以上结果说明,橙皮苷通过干预细胞周期和凋亡抑制食管癌Eca9706细胞增殖.  相似文献

应用MTT实验、细胞计数法检测吲哚乙酸结合辣根过氧化物酶,流式细胞仪(FCM)和DNA末端标记法(TUNEL)检测IAA/HRP对细胞增殖周期和细胞凋亡的作用,研究了吲哚乙酸(indole-3-acetic acid IAA)结合辣根过氧化物酶(horseradish peroxidase HRP)对K562细胞增殖的影响.结果表明,与对照组相比,IAA和HRP单独处理组对K562细胞的生长具有部分抑制作用,但无显著性差异;而IAA与HRP结合后对K562细胞的抑制作用明显增强.流式细胞仪检测结果表明,K562细胞阻滞于G2/M期.IAA/HRP具有明显的诱导K562细胞凋亡的作用,且诱导凋亡的作用与IAA浓度呈正相关(r=0.971,P<0.01).IAA/HRP能明显抑制K562细胞生长,将细胞周期阻滞于G2/M期,诱导细胞凋亡.  相似文献

目的观察硒蛋氨酸(selenome-thionine)对DU145前列腺癌细胞增殖及凋亡的影响,并初步探讨其作用机制.方法采用MTT比色法观察1.25,2.50,5.00μmol.L-1硒蛋氨酸对DU145细胞增殖活力的抑制作用;吖啶橙染色观察硒蛋氨酸诱导细胞凋亡情况;流式细胞术分析细胞周期及凋亡,并计算细胞平均凋亡指数;免疫细胞化学技术观察细胞内激活的caspase3的表达情况.结果经1.25,2.50,5.00μmol.L-1硒蛋氨酸处理48 h后,DU145细胞生长抑制率分别为16.35%,38.10%和73.54%,3组间比较差异有显著性(P<0.01);随硒蛋氨酸浓度升高细胞生长抑制率增加,呈明显的剂量依赖性.吖啶橙荧光染色可见经1.25,2.50,5.00μmol.L-1硒蛋氨酸处理后的细胞呈明显凋亡形态,并随药物剂量增加凋亡细胞数增多;流式细胞术结果显示,经1.25,2.50,5.00μmol.L-1硒蛋氨酸处理48 h后,其细胞的凋亡率分别为5.34%,8.71%和13.6%,3组间比较差异有显著性(P<0.05),随硒蛋氨酸浓度升高细胞凋亡率升高,呈剂量依赖关系;免疫细胞化学染色结果显示,随着硒蛋氨酸剂量增加,细胞内激活的caspase3表达上调.结论硒蛋氨酸对DU145细胞具有明显的抑制作用,其机制可能与激活的caspase途径诱导细胞凋亡有关.  相似文献

目的 观察澳洲茄胺盐酸盐对HeLa细胞c-myc,H-rass和hTRT基因表达的影响.方法 用鼠抗人c-myc McAb,H-ras McAb和hTRT PcAb作为一抗,生物素化羊抗鼠IgG作为二抗,用免疫组化方法测定澳洲茄胺盐酸盐对HeLa细胞c-myc,H-ras和hTRT基因的表达.结果 澳洲茄胺盐酸盐处理的HeLa细胞与对照组相比,c-myc,H-ras和hTRT基因的表达均明显降低(P<0.05).结论 澳洲茄胺盐酸盐下调HeLa细胞c-myc和H-ras基因的表达,抑制细胞增殖,诱导分化;澳洲茄胺盐酸盐下调HeLa细胞hTRT基因的表达,抑制端粒酶活性,抑制细胞生长.  相似文献

目的:研究黄芩苷(BA)抑制Jurkat T细胞(人T淋巴细胞白血病细胞株)的生长和诱导其凋亡的作用及机制。方法:以噻唑兰(MTT)比色法测定BA对细胞生长的抑制率;胞内[Ca2 ]i和线粒体跨膜电势(ΔΨm)分别用F luo-4/AM和D iOC6(3)荧光探针标记,流式细胞仪检测;PI染色流式细胞术分析细胞周期分布及凋亡率;凋亡细胞形态以Hoechst33342/PI染色后荧光显微镜观察。结果:BA明显抑制Jurkat细胞增殖(P<0.01),并呈时间、浓度依赖性;BA可浓度依赖的诱导Jurkat细胞[Ca2 ]i浓度上升、线粒体膜电势ΔΨm降低,将细胞周期阻滞在S期,凋亡率增加,荧光显微镜下可见经BA(100 mg/L)作用的细胞呈现典型的凋亡核固缩表现,胞核呈致密浓染的颗粒状荧光。结论:BA体外显著抑制Jurkat T细胞增殖及诱导其凋亡,其机制可能与胞内[Ca2 ]i及线粒体依赖的凋亡通路有关。  相似文献

目的:观察选择性COX-2抑制剂尼美舒利对人类骨髓白血病K562细胞增殖和凋亡的影响。方法:分别以尼美舒利25、50、100μg.mL-1体外处理骨髓白血病K562细胞,采用MTT比色法、流式细胞仪分析技术及DNA片段凝胶电泳等方法,检测尼美舒利对K562细胞增殖和凋亡的影响。结果:尼美舒利剂量依赖性地抑制K562细胞增殖,其中等剂量组(50μg.mL-1)的抑制作用强于阿霉素组(25μg.mL-1);尼美舒利可诱导细胞凋亡,使细胞阻滞于G0/G1期,S期细胞明显减少,其诱导细胞凋亡的作用也呈剂量依赖性(剂量由低至高,凋亡率分别为(3.4±2.8)%,(16.8±1.8)%和(42.7±2.5)%;DNA条带分析也进一步证实了尼美舒利的促凋亡作用。结论:尼美舒利在体外可显著抑制K562细胞增殖,该作用可能与其干扰细胞周期、诱导细胞凋亡有关。  相似文献

研究西达本胺(Chidamide)对结肠癌细胞系HCT-15的增殖抑制作用及机制. 以不同浓度西达本胺处理HCT-15细胞,CCK-8法检测细胞增殖情况及细胞的药物敏感性;平板克隆形成实验检测细胞体外克隆形成能力;流式细胞术检测细胞凋亡;EdU实验检测细胞增殖周期;Western blot检测乙酰化组蛋白、线粒体凋亡途径相关蛋白、细胞周期相关蛋白表达水平的变化. 结果表明:西达本胺抑制HCT-15细胞增殖呈浓度和时间依赖性,细胞体外克隆形成能力减弱,凋亡增多,细胞分裂相明显减少、总周期变慢,乙酰化组蛋白H3和H4、线粒体介导的相关凋亡蛋白表达上调、p21、p27表达上调、CDK2、CyclinA2蛋白表达下调. 西达本胺可抑制HCT-15细胞增殖,通过线粒体途径诱导细胞凋亡并阻滞细胞周期.  相似文献

为探讨1,25-二羟维生素D3[1,25-dihydroxy vitamin D3,1,25(OH)2D3]对人卵巢癌细胞HO8910生长和增殖的作用,选用人卵巢癌细胞HO8910进行体外培养。培养时添加不同浓度的1,25-二羟维生素D3,采用MTT比色法测定细胞生长抑制率,采用流式细胞术(flow cytometry,FCM)进行细胞周期及细胞凋亡分析。结果:不同浓度的1,25-二羟维生素D3对HO8910细胞生长均起到不同程度的抑制作用(P<0.05),并呈浓度和时间依赖关系;能使细胞周期时相发生改变,G1期细胞增多(P相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
