Lack of pressor effect of dopamine in the pentobarbital-anesthetized rat

Summary The blood pressure and heart rate responses to intravenous dopamine infusion at 2.5, 5.0 and 10.0 g·min^–1·100 g^–1 were studied in conscious and pentobarbital-anesthetized Sprague — Dawley rats. In the conscious rats, dopamine caused a significant dose-related increase in the mean arterial blood pressure which was abolished in the anesthetized rats. The heart rate increased significantly only at the highest dose infused. The responses to equipressor doses of noradrenaline (40 ng·min^–1·100 g^–1) and phenylephrine (1.0 g·min^–1·100 g^–1) were also suppressed in the anesthetized rats. The results suggest that pentobarbital anesthesia depresses the blood pressure response to dopamine infusion in the rat through a depression of activation of alpha-adrenoceptors.16 June 1986     

Adenosine uptake in pyrimidine 5′-nucleotidase deficient human erythrocytes via a high affinity transport system

Summary Using a pulse-labeling technique,¹⁴C-adenosine uptake into pyrimidine 5-nucleotidase (P5N) deficient erythrocytes (RBC) was found to be impaired. The Lineweaver-Burk plot showed K_m values of 2.0×10^–3 mM and 0.2 ×10^–3 mM for normal RBC and P5N deficient RBC, respectively. These results indicate that P5N is one of regulators of the adenosine transport system and/or is associated with adenosine carrier protein.     

Physicochemical studies of indocyanine green (ICG): absorbance/concentration relationship,pH tolerance and assay precision in various solvents

Summary Indocyanine green (ICG) obeyed the Beer-Lambert law within the concentration range 1.25 g/ml–10.0g/ml in distilled water, methanol, dimethylformamide (DMF), 1.2-propanediol and aqueous buffers (pH 9.0), but only up to 7.5 g/ml in human bile and 0.5% human albumin, and only to 5.0 g/ml in human duodenal fluid. ICG was rapidly (<1 h) decomposed to a colorless derivative at pH<5 and >11, but remained relatively stable for 48 h at pH 8–10. ICG is an indicator and a weak acid with a pKa of 3.27. In bile stabilized with 25% methanol, the precision of the method (CV) is 5% and the accuracy is 106–127%.     

Soybean photosynthesis: simultaneous14CO2 and O2 estimates

Summary A method of estimating the photosynthetic rate of soybean leaves using an oxygen electrode is presented. The procedure is rapid, requires small samples and compares favourably with estimates by other techniques. Light saturation occurs at 1200 E·m^–2·sec^–1. The apparent K_m for HCO ₃ ^– is 3.2 mM at pH 7.6.     

Calcium release from frog sarcoplasmic reticulum by an imidazolyl reagent

Summary Calcium is released from the isolated heavy sarcoplasmic reticulum (SR) of frog skeletal muscle upon application of 0.1–1 mM diethylpyrocarbonate (DEP, an imidazolyl reagent). The Ca-ATPase activity of SR was suppressed by 20% in the presence of 1 mM DEP. More than 1 mM of free magnesium ion or 5 M ruthenium red eliminated the effect of DEP on calcium release but not on Ca-ATPase activity. A plausible site of DEP action is on the calcium channel.     

Hepatic glucose signals vagally modulate the cyclicity of gastric motility in rats

The cyclicity and intensity of gastric motlity were examined following glucose injection into the portal vein with an intragastric balloon in anesthetized rats. Enhanced gastric motility caused by insulin administration was influenced by 4 mM glucose (25 l) injected into the portal vein; glucose provoked a shift in the cyclicity power spectrum without any change in intensity. The peak power spectrum shifted from 4.0–5.0 cpm to 2.0–3.0 cpm. Hepatic branch vagotomy abolished the response.The results suggest that glucose signals in the hepatic vagal branch modulate the cyclicity of gastric motiligy.     

Green tea epigallocatechin-3-gallate is an inhibitor of mammalian histidine decarboxylase

(–)-Epigallocatechin-3-gallate, an antiproliferative and antiangiogenic component of green tea, has been reported to inhibit dopa decarboxylase. In this report, we show that this compound also inhibits histidine decarboxylase, the enzymic activity responsible for histamine biosynthesis. This inhibition was proved by a double approach, activity measurements and UV-Vis spectra of enzyme-bound pyridoxal-5-phosphate. At 0.1mM (–)-epigallocatechin-3-gallate, histidine decarboxylase activity was inhibited by more than 60% and the typical spectrum of the internal aldimine form shifted to a stable major maximum at 345nm, suggesting that the compound causes a stable change in the structure of the holoenzyme. Since histamine release is one of the primary events in many inflammatory responses, a new potential application of (–)-epigallocatechin-3-gallate in prevention or treatment of inflammatory processes is suggested by these data.Received 8 April 2003; received after revision 20 May 2003; accepted 3 June 2003     

Über «therapeutische» und «toxische» Digitoxigeninwirkungen auf elektrophysiologische Messgrössen des Meerschweinchenvorhofs

Summary Therapeutic concentrations of digitoxigenin (10^–7–3× 10^–7 g/ml), producing positive inotropic effects, did not alter resting and action potentials of electrically driven guinea-pig auricles. However, toxic concentrations of digitoxigenin (5 × 10^–7–10^–8 g/ml), producing arrhythmias and contracture of the myocardium, had a marked influence upon resting and action potentials and conduction velocity.     

Choleretic and cholestatic effects of infused bile salts in the rat

Summary In rats, at low infusion rates taurocholate (TC), taurochenodeoxycholate (TCDC) and taurodeoxycholate (TCD) each produced an increase in bile flow of 20–50%. However, at high infusion rates (5–20 moles min^–1 kg^–1) the cholestatic effects of the bile salts were revealed and the relative toxicity of the bile salts was seen to be TDC>TCDC>TC.     

On the use of phosphamidon for eradication of fresh water fish predators

Zusammenfassung Laboratoriumsexperimente mit Phosphamidon ergeben eine günstige kritische Dosisempfindlichkeit für Labeo-Karpfen (150–250 ppm). Es gelingt so leicht, Wasserinsekten (2–10 ppm) und Raubfischstadien (20–125 ppm) aus den Frischwasserreservoiren zu eliminieren.

Kinetics and inotropic action of probenecid in guinea-pig heart in vitro

Summary Probenecid (100–750 g·ml^–1) was found to inhibit cardiac contraction force in untreated and digoxin-treated (100 g·ml^–1) isolated right guinea-pig atrium in vitro by a reversible process, without influencing beating frequency. At low concentrations (1.5–60 g·ml^–1),¹⁴C-probenecid was accumulated into right atrium by an oxygen-dependent process. Correlation between uptake and negative inotropic action of probenecid could not be found.     

Studies on hydrogen,halides and thiocyanate ions in dioxane-water mixtures at various temperatures

Summary The free enrgies of transfer (G _t ⁰ ) of H⁺, Cl^–, Br^–, I^– and SCN^– ions from water to 10, 20, 30 and 40% dioxanewater mixtures have been studied from potentiometric measurements. For anions the G _t ⁰ values are in the order Cl^–>Br^–>SCN^–>I^–.     

The effect of insulin on the electrophoretic mobility of rat hepatocytes

Summary The addition of insulin (10 U) to a suspension of isolated hepatocytes in Krebs-Ringer bicarbonate solution. causes an increase in the negative electrophoretic mobility of the cells from –1.68 m sec^–1 V^–1 cm to 2.26 m sec^–1 V^–1 cm. This observation supports the findings by other workers that the binding of insulin to its receptor leads to a marked change in the membrane.Acknowledgment. Variations de l'activité des auxines-oxydases dans les racines duLens

Summary 2, 4-dichlorophenol (DCP), 2, 4-dinitrophenol (DNP) and 2, 4-dinitro-o-cresol (DNC) applied at a concentration of 1.10^–7 to 1.10^–3 M, produce a dramatic increase in IAA-oxidase activity in young cells ofLens root. IAA destruction by breis from root tissues was inhibited by Mn⁺⁺ ion at low concentrations of DCP (1·10^– to 1.10^–7 M), but was enhanced by Mn⁺⁺ ion at higher (1.10^–5 to 1.10^–3 M) DCP levels.

Dopamine and cerebral cortical blood flow in the rat

Summary Dopamine topically applied to the cerebral cortex (1–20 g/ml) or administered i.v. (0.5–64 g/kg/min) has no effects on cerebral cortical blood flow in the rat.     

‘Binding’ of glycine andγ-aminobutyric acid to synaptosomal fractions of 6 regions of the feline brain; effects of strychnine

Summary GABA (6×10^–6 M) binding to synaptosome-enriched fractions of cat CNS exhibited a clear rostro-caudal gradient, whereas glycine (6×10^–6 M) binding was greatest to particles of cerebellar cortex, and this was followed by medulla caudate nucleus cerebral cortex pons > corona radiata. Strychnine-SO₄ (10^–3 or 10^–4 M) inhibited the binding of GABA and glycine in all brain regions studied; at 10^–5 M this drug inhibited the binding of both GABA and glycine only to particles of the cerebral cortex.This study was supported by Centro Nacional Ramón y Cajal and Fundación Juan March. The levels of metallothionein-like proteins in animal tissues

Summary The level of metallothionein-like proteins was determined in different tissues of 6 animal species. The highest concentrations were found in pig and rat tissues. The organs richest in metallothionein-like proteins included: kidneys (101–305 g/g), intestine (127–257 g/g) and liver (54–496 g/g).Studies performed under the Polish-American agreement 05-009-2 with National Institute for Occupational Safety and Health, PHS, USA.     

The uptake of iodinated protein by spleen cells of guinea-pigs in vitro

Zusammenfassung 6×10^–10 g bis 10^–4 g J 125 Rinderserum-Albumin (BSA) bzw. 1,0×10^–7 bis 1,0×10^–6 g J 125 Rinderserum--Globulin (BGG) werden 3 × 10⁷ Milzzellen normaler Meerschweinchen in vitro angeboten. Unabhängig von der Höhe des Angebotes beträgt die Menge des aufgenommenen BSA 1%, die des aufgenommenen BGG 10% des Angebotes. Durch 4C Inkubations-temperatur wird die Aufnahme z.T. behindert. Die Aufnahme findet nach wenigen Sekunden Inkubationszeit statt und ist nach etwa 2–3 min abgeschlossen.

Nitric oxide-related pancreatic endocrine responses to hyperglycaemia in the conscious calf

Mean plasma insulin concentration was reduced and mean plasma glucose concentration increased following the administration of N-nitro-L-arginine methyl ester (L-NAME; 100 mol kg^–1 i.a.) in conscious calves given continuous infusions of exogenous glucose (30–60 mol min^–1 kg^–1 i.v.). It is concluded that the rise in plasma insulin concentration which occurs in these animals in response to glucose is mediated, at least in part, by a nitric oxide-related factor (NOx).
