研究AP-1及其相关基因uPA、uPAR在人肺癌中的表达及其与肺癌临床病理特征的关系.通过免疫组织化学方法检测101例肺癌组织及7例正常肺组织中AP-1、uPA、uPAR的表达情况,利用CMIAS2000型多功能真彩病理图像分析系统测量计算各病例中c-jun、c-fos、uPA、uPAR蛋白阳性细胞的平均光密度(DAO)和积分光密度(DIO).c-jun、c-fos在肺癌组织中表达的阳性率及其共表达率均显著高于正常肺组织;二者在NSCLC的表达显著强于SCLC;不同组织学类型肺癌中二者表达具有显著性差异,腺癌中表达最强,小细胞肺癌中表达最弱;c-jun、c-fos与肺癌分级、合并分期、淋巴结转移均呈显著正相关.uPA、uPAR与肺癌淋巴结转移密切相关,与合并分期呈显著正相关;uPAR与肺癌分级呈显著正相关;各指标之间均存在显著正相关关系.研究结果表明uPA、uPAR参与了肺癌侵袭转移,AP-1信号传导通路在该过程中起了重要调节作用.  相似文献

目的:研究正常状态下黑线仓鼠脑内前脑皮层、海马和下丘脑区c-fos的基础表达。方法:采用免疫组织化学方法(SABC)检测c-fos蛋白在黑线仓前脑皮层、海马和下丘脑区的表达,以昆明品系小鼠为对照,建立足底电击急性应激小鼠模型。结果:正常生活状态下黑线仓鼠脑内前脑皮层、海马和下丘脑区c-fos蛋白广泛表达;其中海马CA1和CA3区c-fos阳性神经元相对数量较少,海马DG区、前脑皮层和下丘脑区c-fos阳性神经元相对数量较多;与正常小鼠相比,前脑皮层c-fos阳性神经元显著增多(P〈0.01)。应激小鼠各脑区c-fos阳性神经元数量明显增多,与正常小鼠比较有统计学显著差异(P〈0.05,P〈0.01)。各脑区c-fos阳性神经元平均目标灰度值结果类似。结论:正常状态下黑线仓鼠脑内c-fos蛋白的表达可能与动物的行为活动和内分泌、空间认知功能以及复杂运动的调控密切相关。  相似文献

Previous studies have suggested a role for c-fos in cellular differentiation in fetal membranes, haematopoietic cells and teratocarcinoma stem cells. In other cell types, such as fibroblasts, c-fos expression is normally very low, but is rapidly induced by peptide growth factors, implicating c-fos in growth control mechanisms. Here, we show that the TPA (12-O-tetradecanoylphorbol-13-acetate)-induced macrophage-like differentiation of HL60 human promyelocytic precursor cells is accompanied by the induction of both c-fos mRNA and protein within 15 min after treatment, suggesting a functional role for c-fos in this differentiation system. In quiescent terminally differentiated macrophages, expression of c-fos is inducible by the macrophage-specific growth factor colony-stimulating factor-1 (CSF-1). The kinetics of c-fos induction, however, are entirely different from those in growth factor-stimulated fibroblasts, supporting the view that the c-fos gene product may serve different functions in different cell types.  相似文献

探讨复方天麻制剂外周镇痛机理,即对神经递质和c-fos基因表达的影响.以福氏完全佐剂制作慢性疼痛模型,高效液相色谱法测定血清中5-HTP、5-HI AA含量,免疫组织化学法检查外周组织c-fos的表达.实验结果表明,复方天麻制剂降低血清5-HTP、5-HI AA含量,抑制外周组织c-fos表达;通过降低血清吲哚类神经递质含量,抑制疼痛基因表达达到外周镇痛作用.  相似文献

Transfer of mouse or human c-fos proto-oncogenes into F9 teratocarcinoma stem cells results in expression of c-fos mRNA and protein. 目的:观察中药雪芙蓉胶囊对雄性大鼠力竭跑台时间、血清激素水平及下丘脑室旁核c—fos表达的影响。方法:将30只雄性成年大鼠随机分为3组,即安静对照组、训练组、中药训练组。中药训练组每天灌服雪芙蓉胶囊(5mg/kg),8周后测试大鼠力竭跑台时间,检测血清睾酮及皮质醇含量,用免疫组织化学结合图像半定量方法对c—fos神经元的数量、面积及灰度进行测量和分析。结果:与训练组比较,中药训练组大鼠力竭跑台时间延长、血清睾酮水平增高(P〈0.01,P〈0.05)、血清皮质醇水平下降(P〈0.05);下丘脑室旁核c—fos免疫阳性细胞数量和阳性产物面积表达均降低(P〈0.05,P〈0.01),灰度没有显著差异(P〉0.05)。结论:中药雪芙蓉胶囊可提高大鼠的运动能力,延缓中枢疲劳,机理可能与此中药提高力竭运动大鼠血清睾酮水平,同时降低大鼠血清皮质醇水平及下丘脑室旁核c—fos的过度表达有关。  相似文献

Alterations in neuronal gene expression have been proposed to account for permanent changes in brain function such as learning and memory. In particular, it has been suggested that protooncogenes such as c-fos may be rapidly induced in conditions that lead to neuronal plasticity and evoke permanent changes in the expression of effector genes. Concentrations of the c-fos proto-oncogene increase rapidly following depolarization-induced calcium influx in non-dividing neuronally differentiated PC 12 cells. Recently, the presence and induction of c-fos in the adult brain and spinal cord has been observed. Here we report that electrically-induced seizure activity, which leads to a permanent increase in the response of the brain to future seizures (kindling), rapidly and transiently increases c-fos protein-like immunoreactivity in the nuclei of granule cells in the rat dentate gyrus. These results suggest that c-fos protein is present within the nuclei of adult mammalian neurons, and could be involved in plastic changes in the nervous system associated with seizure activity.  相似文献

摘要: 目的探索低频电刺激对中国树鼩死亡后脏器中激素水平的影响。方法给予中国树鼩低频电刺激,分别于死亡后0 h、3 h、6 h、12 h、18 h、24 h、36 h 和72 h 取甲状腺、肝脏、脾脏,用放射免疫法测定内皮素( endothelin,ET) 、心钠素( atrial natriuretic factor,ANF) 、血栓素( thromboxane,TX) 水平; 剥取死后0 h 中国树鼩中脑腹侧背盖区( ventral tegmental area,VTA) 检测c-fos 表达。结果电刺激后,中国树鼩尸体脏器中内皮素、心钠素、血栓素水平比对照组显著升高,其含量随死亡后时间的延长而降低,且呈现一定的趋势; VTA c-fos 表达明显增强。结论低频电刺激能引起中国树鼩脏器中激素的合成、释放和脑组织c-fos 表达。  相似文献

目的:观察电针(EA)对炎症痛模型佐剂性关节炎(AA)大鼠炎症局部c-fos表达的影响,以探讨针刺对炎症痛的外周镇痛机制。方法:以福氏完全佐剂造成炎症痛模型AA、选取悬钟、昆仑穴EA治疗,采用免疫组织化学方法检测c-fos表达。结果:EA可以抑制AA大鼠炎症局部c-fos的表达。结论:c-fos可能参与了EA对炎症痛的外周镇痛作用。  相似文献

There has been much interest in the biochemical and biophysical processes that couple extracellular signals to alterations in gene expression. While many early events associated with the treatment of cells with growth factors have been described (for example, ion flux and protein phosphorylation), it has proved difficult to establish biochemical links to gene expression. Recently, the study of such genomic control signals has been facilitated by the demonstration that the c-fos proto-oncogene is rapidly and transiently induced by treatment of several cell types with polypeptide growth factors and other growth modulating substances. In one particular system it has been shown that nerve growth factor (NGF) causes a transient induction of c-fos in the phaeochromocytoma cell line PC12, within 15 min. Furthermore, the magnitude of this induction can be modulated with pharmacological agents such as peripheral-type benzodiazepines (BZDs). Thus, the study of c-fos expression in PC12 cells could yield valuable clues to the coupling mechanisms linking cell surface activation to genomic events. Here we demonstrate that c-fos is induced in PC12 cells either by receptor-ligand interaction or by agents or conditions that effect voltage-dependent calcium channels.  相似文献

It has been suggested that the proto-oncogenes c-fos and c-myc participate in the control of genetic events which lead to the establishment of prolonged functional changes in neurons. Expression of c-fos and c-myc are among the earliest genetic events induced in cultured fibroblast and phaeochromocytoma cell lines by various stimuli including growth factors, peptides and the intracellular second messengers diacylglycerol, cAMP and Ca2+. We report here that physiological stimulation of rat primary sensory neurons causes the expression of c-fos-protein-like immunoreactivity in nuclei of postsynaptic neurons of the dorsal horn of the spinal cord. Activation of small-diameter cutaneous sensory afferents by noxious heat or chemical stimuli results in the rapid appearance of c-fos-protein-like immunoreactivity in the superficial layers of the dorsal horn. However, activation of low-threshold cutaneous afferents results in fewer labelled cells with a different laminar distribution. No c-fos induction was seen in the dorsal root ganglia, gracile nucleus or ventral horn. Thus, synaptic transmission may induce rapid changes in gene expression in certain postsynaptic neurons.  相似文献

目的:探讨川芎嗪干预脑缺血/再灌注损伤后对Bcl-2蛋白、c-fos蛋白、Caspase-3蛋白表达的影响及其保护大脑损伤的作用机制.方法:将SD大鼠30只,随机分为3组,为假手术组、模型组和治疗组,每组10只.建立脑缺血/再灌注模型.治疗组用川芎嗪干预,采用免疫组化法测定川芎嗪干预大鼠脑缺血24h后对Bcl-2蛋白、c-fos蛋白、Caspase-3蛋白表达的影响.结果:在脑缺血/再灌注损伤24h后,治疗组脑组织Bcl-2蛋白阳性神经元数较模型组明显增多,差异有统计学意义(P<0.01);治疗组c-fos蛋白和Caspase-3蛋白表达阳性神经元数较模型组明显减少,差异有统计学意义(P<0.01).结论:川芎嗪能上调Bcl-2蛋白表达,下调c-fos蛋白和Caspase-3蛋白表达,川芎嗪可能通过抑制脑缺血/再灌注损伤后细胞凋亡的机制,从而对脑脑缺血/再灌注损伤有保护作用.  相似文献
