空间环境对恶性黑色素瘤血管生成拟态的影响

观察空间诱变后恶性黑色素瘤B16细胞株的在小鼠体内肿瘤组织中血管生成拟态数目的改变,并探讨可能的机理。应用细胞低温长期生存系统,将B16细胞株搭载于中国第20号返回式卫星,返地后单克隆化,从得到的110株单克隆空间诱变B16细胞株中随机选取7株,编号为38#～44#,常规培养6代后和对照细胞株在荧光倒置显微镜下观察肿瘤细胞的形态;同时采用MTT(3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide,又称噻唑蓝)法,检测细胞增殖情况,将增殖差异较大的39#和44#细胞株接皮下种于C57BL/6J小鼠,两周后颈椎脱臼法处死小鼠,取出瘤体,经福尔马林固定后,采用CD31和PAS套染的方法观测肿瘤组织血管生成拟态的情况,并推测其与肿瘤生长速度和转移可能关系。     

空间环境对黑色素瘤B16细胞致瘤基因和蛋白质表达的影响

初步探讨了空间环境对黑色素瘤B16细胞致瘤基因和蛋白质表达的影响。选择经第20颗返回式卫星搭载的B16细胞,进行体外和体内实验后,筛选出性状变异明显的空间诱变B16细胞株,检测空间诱变B16细胞基因和蛋白质表达的变化。将筛选出的空间诱变B16细胞接种于C57BL/6小鼠,2周后,处死荷瘤小鼠,取出移植瘤,利用免疫组化方法,检测接种空间诱变B16细胞的荷瘤小鼠移植瘤细胞Melan-A和VEGF蛋白的表达情况;利用RT-PCR方法,检测蛋白质水平上表达变化明显的空间诱变B16细胞melan-a,gp100,bag-3和l-pgds基因的表达情况。与对照细胞相比,1株空间诱变B16细胞荷瘤小鼠移植瘤细胞Melan-A和VEGF蛋白表达明显增加,其他细胞2种蛋白质的表达无明显变化。RT-PCR结果显示:此株细胞的melan-a,gp100和l-pgds基因表达增加,bag-3基因表达降低。空间环境的复合因素可诱导B16细胞基因和蛋白质表达产生变化。     

空间诱变致成瘤性减弱的B16细胞株形态学观察

观察动物接种实验致瘤性显著减弱的空间诱变小鼠黑色素瘤B16细胞株的细胞形态、细胞骨架和表面结构与对照细胞株的差异。分别使用激光共聚焦显微镜观察筛选所得阳性细胞株与对照细胞株,以及用荧光素标记的鬼笔环肽染色,观察细胞骨架的异同。应用原子力显微镜(AFM)对筛选所得阳性细胞株和对照细胞株分别成像,观察空间诱变细胞株与对照组细胞株所得图像的差异。激光共聚焦显微镜观察结果显示,从空间诱变细胞株中筛选出的8#细胞株的细胞骨架荧光较对照组增强,细胞骨架纤维比对照组粗大。AFM图像显示,8#细胞株分泌的纤维粘连蛋白的纤维形态和分布与对照组细胞有较大差异,其纤维较为扁平、走行弯曲或呈轻度迂曲状;对照组细胞纤维呈山脊状、放射状,源自细胞伪足部细胞膜。在8#细胞株图像中有少量“孔洞”样结构,而在对照组细胞中未观察到类似结构。     

太空诱变宫颈癌细胞的差异表达基因初探

将搭载于“神舟四号”飞船飞行返地后的宫颈癌Caski细胞进行单克隆化,筛选出生长速度快于对照组、编号为44F10的细胞克隆,G1期细胞减少,S期细胞增多,成瘤能力增强;编号为48A9的细胞克隆细胞学行为与之相反,与对照组差异均有显著性(P<0.05)。为了解太空诱变肿瘤细胞生物学行为改变的机制,从分子水平入手研究经太空诱变的宫颈癌细胞和地面对照细胞的差异表达基因。应用含2747个人类肿瘤相关基因的Oligo双通道芯片研究差异表达基因。分别抽提44F10、48A9组和地面对照细胞的总RNA,逆转录cDNA并标记探针。将实验组和对照组cDNA探针混合,分别与同一张芯片杂交后,用不同的波长扫描荧光强度,从而筛选出差异基因。44F10组有16个基因呈现差异表达,48A9组有36个基因呈现差异表达。差异性表达主要涉及细胞凋亡、细胞增殖、细胞周期调控和信号转导的基因。促进细胞增殖的基因在44F10组中表达上调,而在48A9组中限制细胞增殖的基因表达上调。研究表明,太空诱变宫颈癌细胞的差异表达基因导致了细胞生物学行为的改变。     

空间环境对黑色素瘤B16细胞体内增殖及免疫原性的影响

初步探讨空间环境对黑色素瘤B16细胞增殖及免疫原性的影响。选择4株经体外实验筛选出生物学性状变异明显的第20颗返回式卫星搭载的空间培养B16细胞,检测其体内增殖能力及免疫原性的变化。将筛选出的4株空间培养B16细胞接种于C57BL/6小鼠,其中一组接种于腹腔,观察荷瘤小鼠生存期;另一组接种于腋下皮肤,检测小鼠出瘤时间。接种至2周时,处死,分离荷瘤小鼠移植瘤和血清。称重荷瘤小鼠移植瘤,观察空间培养B16细胞增殖能力;利用HE染色法观察荷瘤小鼠移植瘤切片细胞形态;采用放射免疫分析法(Radioimmunoassay,RIA)测定荷瘤小鼠血清白介素-2(InterLeukin-2,IL-2)浓度,观察空间培养B16细胞免疫原性变化情况。与对照细胞相比,1株空间培养B16细胞的荷瘤小鼠生存期明显缩短,出瘤时间明显延长。HE结果显示:此株空间培养B16肿瘤组织内有淋巴细胞浸润;RIA结果显示:3株空间培养B16细胞荷瘤小鼠血清IL-2浓度明显增加。空间环境的复合因素可诱导B16细胞增殖能力及免疫原性产生变化。     

过表达IL-8受体对脐带源间充质干细胞生物活性的影响

目的探究腺病毒介导的IL-8受体过表达对脐带源间充质干细胞生物活性的影响。方法本研究利用含有IL-8RA和IL-8RB的c DNA和绿色荧光蛋白(GFP)基因的重组腺病毒载体(p Ad-IL-8RA-GFP、p Ad-IL-8RB-GFP和p Ad-Null-GFP)分别转染分离培养的P3-5代脐带源间充质干细胞,倒置荧光显微镜观察GFP的表达,流式细胞仪检测干细胞标签蛋白CD29、CD44、CD34、HLA-DR的表达,CCK-8实验检测转染后细胞的增殖能力,粘附实验及Transwell趋化实验观察过表达IL-8RA/B的脐带源间充质干细胞的迁移能力。结果脐带源间充质干细胞形态均一、生长状态良好;干细胞特异性相关蛋白CD29、CD44阳性表达,CD34、HLA-DR阴性表达;与对照组比较,IL-8RA/B过表达的脐带源间充质干细胞的增殖能力无明显改变,但是过表达IL-8RA/B的间充质干细胞粘附在损伤内皮上以及迁移到小室下方的细胞数目更多。结论以上结果表明IL-8RA/B的过表达不影响脐带源间充质干细胞的增殖能力,但是能够促进脐带源间充质干细胞向IL-8趋化和向损伤内皮细胞粘附。     

A549-KIF4A过表达细胞株的构建与鉴定

非小细胞肺癌A549细胞中驱动蛋白KIF4A的表达量极低.为研究KIF4A在肺癌发生和发展过程中的作用,利用脂质体转染的方法,将p EGFP-KIF4A质粒和p EGFP-C1质粒分别转入A549细胞中,用G418筛选得到单克隆,并扩增得到稳定表达GFP-KIF4A和GFP的细胞株.利用Western Blot以及免疫荧光染色法对得到的细胞株进行鉴定.结果表明:A549-GFP-KIF4A细胞中GFP-KIF4A蛋白的表达水平较高,A549-GFP-C1中仅表达GFP蛋白.免疫荧光检测表明:A549-GFP-KIF4A过表达细胞中GFP-KIF4A荧光融合蛋白的定位与内源KIF4A的定位相同.     

熊果酸对H22荷瘤小鼠抗肿瘤及免疫调节作用

目的 探讨熊果酸（UA）对H22荷瘤小鼠抗肿瘤作用及免疫功能的影响.方法 皮下移植建立H22荷瘤小鼠模型,腹腔注射不同剂量UA,检测抑瘤率和免疫器官指数,MTT法检测脾脏T、B淋巴细胞增殖能力,流式细胞术检测CD4＋、CD8＋T细胞亚群含量及比例,ELISA法检测血清细胞因子IL-2、IL-4和TNF-α的表达量.结果 UA对小鼠皮下移植性肿瘤H22有显著的抑制作用,可降低免疫器官中异常增大的脾指数,增强脾脏中T、B淋巴细胞增殖能力,提高淋巴细胞亚群CD4＋T细胞表达及CD4＋/CD8＋T细胞亚群比例,促进血清IL-2、TNF-α表达,降低IL-4表达.结论 UA可抑制小鼠肝癌H22肿瘤生长,体内可以提高荷瘤小鼠的免疫能力,其抗肿瘤作用可能与机体的免疫调节作用相关.     

胃印戒细胞癌中CD44v6，Survivin与PTEN的表达及临床病理学意义

探讨黏附相关因子变异体CD44v6、凋亡抑制基因Survivin和抑癌基因FIEN在胃印戒细胞癌组织中的表达与其临床病理特征之间的关系,及三者的关系。表明应用免疫组织化学SP法,检测CD44v6、Survivin和FIEN蛋白在73例胃印戒细胞癌组织中的表达。结果胃印戒细胞癌中CD44v6、Survivin和PIEN蛋白的阳性表达率分别为78．1％（57／73）、63．0％（46／73）、34．2％（25／73）。CD44v6随胃印戒细胞癌浸润深度加深、淋巴结的转移逐渐上升（P〈0．05）;随浸润深度加深、淋巴结的转移、器官转移、临床分期增加Survivin蛋白的阳性表达率逐渐上升（P〈0．01）,而FTEN基因的阳性表达率逐渐降低（P〈0．05）。在胃印戒细胞癌组织中CD44v6与PFEN蛋白的表达呈负相关（相关系数r=-0．214,P〈0．05）,Survivin与PTEN的表达呈负相关（r=-0．404,P〈0．01）,CD44v6与Survivin的表达呈正相关（r=0．28,P〈0．01）。因此可知,CD44v6、Survivin和PIEN基因的异常表达与胃印戒细胞癌临床病理特征有关,PIEN和Survivin基因表达可以作为判断胃印戒细胞癌生物学行为的指标;CD44v6与Survivin的协同作用,以及两者与PTEN拮抗作用共同参与胃印细胞癌的发生发展过程。     

AG490下调STAT3活化增强葫芦素B对B16F10黑素瘤细胞的抑制作用

目的:研究Jak2/STAT3特异性抑制剂AG490对葫芦素B(CuB)抑制B16F10黑素瘤细胞效应的影响,并探讨其作用机制.方法:用MTS法检测细胞增殖状态;用碘化丙锭(PI)染色分析细胞凋亡和细胞周期分布;用免疫印迹检测p-STAT3以及STAT3蛋白的表达情况.结果:CuB以时间与浓度依赖方式抑制BI6F10黑素瘤细胞的生长,AG490预处理能增强CuB对B16F10细胞的抑制作用;PI染色分析显示,AG490能增强CuB诱导B16F10细胞凋亡的比率;免疫印迹检测表明,CuB引起Bi6F10细胞中STAT3的活化(p-STAT3),AG490能明显下调pSTAT3蛋白的表达水平.结论:阻断Jak/STAT3的活化可增强CuB对B16F10黑素瘤细胞的抑制作用.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
