Purification of α-ketoaldehyde dehydrogenase from the human liver and its possible significance in the control of glycation

Summary Alfa-ketoaldehyde dehydrogenase, which was extracted and purified from human livers, may act on carbonyl compounds, such as 3-deoxyglucosone, and be involved in the control of glycation (Maillard reaction) in the body.     

Histochemistry of 3β-hydroxysteroid dehydrogenase in monkey adrenal cortex

Zusammenfassung Es wird über mit Hydrocortisonacetat, ACTH und Aldosteronacetat induzierte morphologische und histochemische Veränderungen in der Nebennierenrinde desM. rhesus berichtet. Die bedeutende Verminderung der Steroid-3-ol-dehydrogenasischen Tätigkeit nach Cortisol und deren Zunahme nach ACTH wird hervorgehoben.     

On the production of 12α-hydroxysteroid dehydrogenase fromClostridium group P,strain C48-50 ATCC 29733

Summary The production of 12-hydroxysteroid dehydrogenase ofClostridium group P strain C48-50 was optimized when the organism was grown in the presence of 2% fructose and 0.1% dithiothreitol. It appears that an initial redox potential of less than –160 mV (achieved by autoclaving in the presence of dithiothreitol, dithioerythritol or cysteine) is important in the production of this enzyme.     

Histochemistry of steroid-3β-ol dehydrogenase in the human adrenal cortex

Zusammenfassung Bericht über histochemische Untersuchungen an menschlichen Nebennieren, die chirurgisch bei Fällen von inoperablem Mammacarcinom (mit oder ohne vorhergehende Stimulation mit exogenen ACTH) sowie bei Fällen von Cushingscher Krankheit entfernt worden waren.Anhand der Verteilung der verschiedenen Enzyme und insbesondere des Steroid-3-ol Dehydrogenase wird auf Fragen der Beziehungen der Morphologie und Funktion der normalen Drüse eingegangen und die Cushingsche Krankheit interpretiert.

Die Streuung von -Teilchen an schweren Kernen ist einem optischen Beugungsphänomen ähnlich. Resonanzen wie sie in der Streuung an leichten Kernen beobachtet werden, sind akustischen Resonanzen vergleichbar.     

Synthesis of a pregnancy-associatedα-macroglobulin by human leucocytes

Un test in vitro utilisant l'incorporation de¹⁴C-glutamine ou³H-leucine dans la glycoprotéine a montré que celle-ci peut être synthétisée par des leucocytes.

In situ demonstration of the activity of 3β-hydroxysteroid dehydrogenase on steroid hormone secreting cells within the paraaortic lymph nodes of golden hamster

Summary The in situ activity of 3-hydroxysteroid dehydrogenase was detected in clustered cells which showed steroid-producing morphology, within the capsule of the paraaortic lymph node. In light and electron microscopic studies, the positive reaction products were detected on intracapsular cell clusters. This result indicates that these unique cells may have a steroid secreting function within the lymph nodes.     

Effect of tea consumption on the levels ofα-ketoglutarate and pyruvate dehydrogenase in rat brain

Summary Administration of tea to rats fed on a normal diet results in a marked drop in brain levels of total thiamine as well as of -ketoglutarate and pyruvate dehydrogenase activities. The patterns of decrease in both enzyme activities are similar to that of total thiamine content; they drop to about 65% of the control at 14–20 weeks after continuous consumption of tea.Supported by a grant from the International Foundation for Science (IFS), Sweden.     

Adenosine uptake in pyrimidine 5′-nucleotidase deficient human erythrocytes via a high affinity transport system

Summary Using a pulse-labeling technique,¹⁴C-adenosine uptake into pyrimidine 5-nucleotidase (P5N) deficient erythrocytes (RBC) was found to be impaired. The Lineweaver-Burk plot showed K_m values of 2.0×10^–3 mM and 0.2 ×10^–3 mM for normal RBC and P5N deficient RBC, respectively. These results indicate that P5N is one of regulators of the adenosine transport system and/or is associated with adenosine carrier protein.     

Folding and assembly defects of pyruvate dehydrogenase deficiency-related variants in the E1α subunit of the pyruvate dehydrogenase complex

The pyruvate dehydrogenase complex (PDC) bridges glycolysis and the citric acid cycle. In human, PDC deficiency leads to severe neurodevelopmental delay and progressive neurodegeneration. The majority of cases are caused by variants in the gene encoding the PDC subunit E1α. The molecular effects of the variants, however, remain poorly understood. Using yeast as a eukaryotic model system, we have studied the substitutions A189V, M230V, and R322C in yeast E1α (corresponding to the pathogenic variants A169V, M210V, and R302C in human E1α) and evaluated how substitutions of single amino acid residues within different functional E1α regions affect PDC structure and activity. The E1α A189V substitution located in the heterodimer interface showed a more compact conformation with significant underrepresentation of E1 in PDC and impaired overall PDC activity. The E1α M230V substitution located in the tetramer and heterodimer interface showed a relatively more open conformation and was particularly affected by low thiamin pyrophosphate concentrations. The E1α R322C substitution located in the phosphorylation loop of E1α resulted in PDC lacking E3 subunits and abolished overall functional activity. Furthermore, we show for the E1α variant A189V that variant E1α accumulates in the Hsp60 chaperonin, but can be released upon ATP supplementation. Our studies suggest that pathogenic E1α variants may be associated with structural changes of PDC and impaired folding of E1α.     

Orally administered melatonin stimulates the 3α/β-hydroxysteroid oxidoreductase but not the 5α-reductase in the ventral prostate and seminal vesicles of pinealectomized rats

Summary Pinealectomized rats were treated orally with melatonin (MEL) for 14 days. Prostates and seminal vesicles were investigated for the activity of the 4-3-ketosteroid-5-oxidoreductase (5-R) and of the 3/-hydroxysteroid-oxidoreductase (3-HSO). The activity of the 3a-HSO was significantly stimulated (p<0.01, Friedman test) when compared to controls. The activity of the 5-R did not change significantly.This work was supported by the Deutsche Forschungsgemeinschaft, Sonderforschungsbereich 34 (Endokrinologie).

Résumé Le 3, 17, 21-trihydroxy 5-pregnane (allo-tétrahydro-17-hydroxy-cortexone) et le 3,21-dihydroxy 5-prégane (allo-tétrahydro-cortexone) ont été identifiés dans l'urine humaine. Ces composés avaient les mêmes migrations chromatographiques avant ou après acétylation ou oxydation que les respectifs stéroïdes de synthèse.     

Inhibition of human pancreatic elastase II activity on human aortic elastin by human α2-macroglobulin

Summary Human 2-macroglobulin-human pancreatic elastase II binding were investigated using a homologous substrate, human aortic elastin, in order to test the enzymatic activity. We demonstrated that two moles of 2-M are required to inhibit one mole of HPE_II when the enzyme is added to a mixture of elastin and 2-M. In addition, when the elastase-2-M complex is prepared under some circumstances, it exhibits an elastinolytic activity.

Leydig cell andΔ 5-3β-hydroxysteroid dehydrogenase activity in the testis of toad (Bufo melanostictus) following cold exposuredehydrogenase activity in the testis of toad (Bufo melanostictus) following cold exposure

Summary Leydig cell nuclear area and ⁵-3-hydroxysteroid dehydrogenase activity were increased in the testis ofBufo melanostictus 2 days after a single short-term cold exposure. Both parameters returned to normal values 4 days later.     

Purification and characterization of aβ-glucosidase (linamarase) from the haemolymph ofZygaena trifolii Esper, 1783 (Insecta,Lepidoptera)

Summary A -glucosidase (linamarase) was purified 52-fold with a recovery of 27% from the haemolymph of the larvae ofZygaena trifolii, ESPER, 1783 (Lepidoptera, Zygaenidae). The final enzyme preparation was found to be nearly homogeneous on both disc polyacrylamide gel electrophoresis and SDS-polyacrylamide gel electrophoresis. The molecular weight of the enzyme was determined to be about 130 kDa; it consisted of two subunits of about 66 kDa. The enzyme showed an optimum between pH 4.5 and 5 with linamarin and a broad optimum between pH 3.5 and 6.5 for p-nitrophenyl--D-glucoside; the temperature optimum was 40°C. The -glucosidase showed a high specificity for its endogenous substrates linamarin and lotaustralin. Among the other natural and artificial substrates tested, only prunasin and p-nitrophenyl--D-glucoside were hydrolyzed by the enzyme, whereas linustatin, salicin, cellobiose and trehalose were not. The enzyme is strongly inhibited by -glucosylpiperidine.
