从喜温蓝藻SynechococuselongatusT3分离出一种限制性内切核酸酶,命名为SelI.此酶是限制性内切核酸酶Sau96I的同切酶,它们的识别序列GGNCC.但是SelI的反应条件与Sau96I不同.SelI是一种热稳定的限制性内切核酸酶,最适反应温度的范围是50~57℃  相似文献

通过不同微肥对 Laccaria laccata S238 与 Gyroporus cyanescens等菌根真菌菌丝体生长的研究证实:1) 浓度为 0.01% 的 Na2 B4 O7 对 L.laccata S238 的菌丝体生长促进作用明显,其它浓度的 Na2 B4 O7 以及 M n S O4、( N H4 )6 Mo7 O24、 Zn S O4 对 L.laccata S238 均有抑制作用.在培养 L.laccata S238 时应单独添加低浓度的 Na2 B4 O7, 以促进其正常生长;2) A 处理的 A1 对 G.cyanescens 菌丝体生长具有一定的促进作用,但不是很显著,其它微肥对其生长具有明显的抑制作用  相似文献

本文研究了安徽产百合属(LiliumL.)2种和2变种的染色体数目和核型,结果如下:野百合(L.browniiF.E.Brown)观察了2个居群。歙县居群观察到两种细胞型,细胞型Ⅰ:2n=26=1m+2Sm+10St+10t+3T;细胞型Ⅱ:2n=24+2Bs=2m+2Sm+8St(2sc)+12t+2Bs;石台居群核型公式为2n=24+1Bs=3m+1Bm+6St+12t+2T+1Bs。百合(L.browniiF.E.Brownvar.viridu-lumBaker)核型公式为2n=24+1Bs=4m(2sc)+14St(2sc)+6t+1Bs;药百合(L.speciosunThunb.var,gloriosoidesBaker)核型公式为2n=24=4m+9St(2sc)+10t+1T;条叶百合(L.callosumSieb.etZuce.)核型公式为2n=24=2m+2Sm+4St(2sc)+16t。以上的核型类别全部属于“3B”型,其中野百合2n=26的染色体数目和核型为首次记录。  相似文献

报道了草珊瑚(Sarcandraglabra)细胞悬浮培养的某些特性。细胞悬浮培养的摇床转速对草珊瑚细胞生长至关重要、培养物在100r/min速度下振荡培养几乎难以存活,最佳的摇床转速为40r/min(23.5h/d+80r/min(0.5h/d),其生长可增加5倍左右。细胞悬浮培养的接种量以2.0gFW/瓶合适,其培养物的月产率13.22gFW/瓶(或18.73gDW/1)。细胞生长的时间进程是,接种0-5d,细胞增殖十分缓慢,10-20d迅速生长,20d后基本停止生长。培养基的pH迅速下降时是细胞增殖缓慢的时期,当培养基pH值开始升高时,细胞增殖也显著加快。黄酮苷类物质的合成在0-20d迅速增加,20d左右达到高峰。  相似文献

利用内生固氮菌在植和体内固氮有可能开辟一条非豆科作用直接利用生物固氮的捷烃.KlebsiellaaxytocaSA2是一株从生长在流沙地的先锋植物-沙竹(Pxam-maochloamongolica)内分离到的固氮菌。水稻幼苗接种SA2限菌培养,免疫金银染色根和芭匠石切片确证了SA2的内生特性。SA2可以从水稻侧根发生时产生的裂隙和根表皮胞间入侵支细胞间隙内向和纵向迁移。SA2可以进入根中柱,在导  相似文献

筛选两株生长快、产木聚糖酶活力高的菌种;黑曲霉(Aspergillusniger,AN01)、链霉菌(Streptomycessp.Str7B),酶活力分别达128mg/L·min和176mg/L·min;酶反应最适温度分别为60℃与50℃;最适pH值为5.0和6.0,并分别在pH2.2~5.0,5.8~6.4酶活性稳定;在60℃条件下保温1.5h,酶活力分别剩余20.5%,88.5%,其中AN01株原酶液在90℃保温10min,活力仍剩余14.5%.Cu2+对酶活表现出极强抑制,Fe2+,Mg2+,Ca2+等离子则有促进作用;用纸层析法探讨了不同培养时间各种产物产生的情况.  相似文献

利用真空下氩气保护液相重结晶的方法,对热蒸镀和磁迭射制得的InSb薄膜进行热处理,X射线衍和组织分析的结果表明:热处理前薄膜为InSb,In,Sb各相的混合物;重结晶后,基本为InSb单相,InSb晶粒呈规则状外形长大,热处理后室温下的电子迁移率大大提高,由原来的1.31×10^4cm^2/(V.s)提高到4.47×1064cm^2/(V.s)(热蒸镀)和2.15×10^3cm^2/(V.s)提高  相似文献

本文以哺乳动物细胞表达载体pRc/CMV为运载体,构建了受控于CMV启动子的hSOD基因的表达载体pRc/CMV_SOD,用脂质体转染法转染大白鼠肺上皮细胞L2细胞后,经抗生素G418选择性培养,获得与细胞染色体DNA稳定整合的hSODcDNA导入细胞,经SouthernBlots和WesternBlots分析,结果表明,hSOD在细胞中得到稳定表达,同时测定细胞SOD酶活性,hSODcDNA导入细胞的SOD酶活性是空载体导入细胞和父本细胞的约2倍,并且在选择细胞克隆之后的60天内是持续的.  相似文献

当大蒜细胞培养在MS液体培养基中,细胞生长迅速,脱分化的细胞仍具有合成和积累SOD的能力.培养15天,大蒜细胞生长和SOD酶积累都达到高峰期,细胞合成量鲜重为120.8g/L,细胞单位酶活力为每克细胞5.22×10-7mol/s,细胞生长最适温度为28℃,最适pH值为5.8。植物生长调节物质(2,4—D、KT)对大蒜培养细胞生长和SOD酶积累都有显著的影响,其浓度有一个最佳值。适当地提高培养基中物质的浓度,有利于促进大蒜细胞的生长和胞内SOD酶的积累。培养基中添加一些刺激剂,可大大地提高大蒜细胞内SOD酶的产量。  相似文献

对绿色巴夫藻pavlavaviridis进行了生态条件试验,认为该藻能适应光强500-2500Lux;适温范围5-33℃,最适20-26℃;密度1.005-1.040g/cm3,最适1.005g/cm3;pH5-9都可以满足其正常生长,通过单因子和正交试验表明:培养液的较佳配方是在天然海水中加入(NH2)2CO-N20mg/dm3,KH2PO4-p1mg/dm3,FeSO4-Fe0.2mg/dm3,维生素B11mg/dm3和维生素B120.005mg/dm3。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
