Potassium permeability was monitored in human sweat glands and rat submandibular glands. Acetylcholine increased permeability in both tissues and the responses consisted of transient, calcium-independent and sustained, calcium-dependent components. Amiloride, a drug which inhibits Na+?H+ countertransport, impaired the regulation of potassium permeability in sweat glands but not in the submandibular gland. It is suggested that the stimulus-permeability coupling process in the sweat gland may be sensitive to the lowering of internal pH.  相似文献

Acetylcholine releases calcium from cytoplasmic stores and permits an influx of calcium in salivary acinar cells. The resultant rise in [Ca2+]i causes an increase in potassium permeability which is an important part of the secretory response. We have investigated the effects of 12-0-tetradecanoyl phorbol-13-acetate, a potent activator of protein kinase C, upon this regulation of potassium permeability in superfused pieces of rat submandibular salivary gland. This compound inhibited the initial [Ca2+]o-independent component of the response of acetylcholine but had no effect upon the subsequent [Ca2+]o-dependent phase. This compound does not, therefore, appear to inhibit receptor-regulated calcium influx.  相似文献

However, no renin-like activity could e detected in glands from male and female Wistar rats using either pH 5.8 or 7.4 for assay and a radioimmunoassay specific for renin's reaction product, angiotensin I. Résumé Des glandes sousmaxillaires de chiens ont été coupées en morceaux et mises dans un fixatif composé de 2% KSB(OH)6 et soit de 6.25% d'aldéhyde glutarique, soit de 2% d'osmium et préparées pour l'examen au microscope électronique. Quand les tissus sont fixés à l'aldéhyde glutarique, le précipité est principalement extracellulaire. Par contre, avec le fixatif à l'osmium, le précipité est plus diffus et se trouve partout dans la cellule. Both acetylcholine and a Ca-ionophore, A23187, are comparatively strong stimulants of eccrine sweat secretion in vitro. Nevertheless, the contraction of the secretory coil was seen only after stimulation with acetylcholine but not with alpha or beta adrenergic drugs or with A23187. Summary Acini were enzymatically dissociated from rat submandibular gland and their mucous glycoproteins radiolabelled with14C-glucosamine. Sera from cystic fibrosis patients stimulated the release of labelled TCA/PTA — insoluble material from the cultured acinar cells to a significantly higher degree than did control sera.This work was supported by the Canadian Cystic Fibrosis Foundation and the Sellers Foundation.  相似文献

Summary Proteins secreted by the rat submandibular gland after administration of autonomic agonists have been fractionated by sodium dodecyl sulphate (SDS) — polyacrylamide gel electrophoresis. α-adrenoceptor, β-adrenoceptor and muscarinic agonists were all found to cause the secretion of different protein populations. Disulfide-groups of elaunin fibres of sweat gland basement membrane are demonstrated a) by thiosulfation/aldehyde-fuchsin staining or thiosulfation/Alcian Blue + 0.8 M MgCl2 staining, and b) by identifying SH-groups after reduction with sodiumthioglycollate.
