Zusammenfassung An Schilddrüsenschnitten wurde der subzelluläre Einbau von Phosphor in Phospholipide und die Wirkung von Thyreotropin und von LATS untersucht, wobei die mikrosomale Fraktion die höchste Radioaktivität aufwies.     

A simple method of purifying human sera for the assay of long-acting thyroid stimulator

Résumé Pour purifier les sérums pour l'étalonnage du LATS, on les agite deux fois avec l'échangeur DEAE-Sephadex. Le surnageant contient la quantité entière du LATS (96 ± 8%) et un taux fort réduit des autres protéines. Le traitement élimine la toxicité des sérums et la plupart de l'iodure et de la thyroxine sérique.

Identification of human serum proteins binding iron,copper, and thyroid hormones by starch gel electrophoresis

Résumé On a analysé au moyen d'électrophorèse sur gel d'amidon les protéines qui, dans le sérum humain, lient le fer, le cuivre et l'hormone thyroïdienne, en utilisant comme témoins des protéines purifiées et la radioactivité. Le fer est lié par la -globuline qui, dans certaines familles, paraît se diviser en deux composants. Le cuivre est lié par la céruloplasmine, qui émigre dans la position ₂ rapide. La protéine principale liant la thyroxine est la préalbumine.     

Effect of long acting somatostatin-analogue,SMS 201995, on gut hormone secretion in normal subjects

Summary SMS 201 995 is a new long acting analogue of somatostatin. We have investigated its effect on basal and meal stimulated secretion of gut hormones and have shown that after a single s. c. injection of 50 g it lowers significantly the basal plasma levels of pancreatic polypeptide, secretin, motilin, pancreatic glucagon and insulin, it also effectively suppresses the postprandial release of pancreatic polypeptide, gastrin, secretin, gastric inhibitory peptide, pancreatic glucagon and insulin. Except for the usual brief discomfort of an injection, no symptoms or untoward effects were observed.     

Immunofluorescent localization of beta-lipotropin (beta-LPH) and beta-endorphins in normal and anencephalic human fetal pituitary gland]

The use of antibodies to alpha-(17-39) ACTH, beta-LPH, alpha- and beta-endorphins has enabled the detection of immunoreactive cells in human fetal pituitary glands as early as the eight week of gestation. The same cells reacted with the different antibodies. Immunoreactive cells were also observed in the pituitary of 6 human anencephalic fetuses.

Collagenolytic enzymes in human serum

Zusammenfassung Mit Hilfe des synthetischen Kollagenasesubstrates PZ-Pro-Leu-Gly-Pro-D-Arg wurden im humanen Serum kollagenolytische Enzyme nachgewiesen, deren Spezifität der Clostridiopeptidase A entspricht. Die Spaltungsgeschwindikeit zeigt in Abhängigkeit von pH ein Optimum bei pH 7,2 und bei pH 8,0. Das spricht für die Existenz zweier Enzyme. Zur quantitativen Bestimmung der Enzymaktivitäten wurde eine Methode ausgearbeitet, mit der eine Reihe von normalen humanen Seren analysiert wurde.     

Immunofluorescent antibody studies of a murine leukemia virus: Comparison of human,bovine and murine systems

Zusammenfassung Antikörper bei genesenden Mäusen nach Infektion mit Leukämie-Viren (S-63) erzeugt, zeigen eine Kreuzreaktion mit den Leukämie-Antigenen aus Rinder- und Menschenserum und Gewebe. Diese Befunde sprechen dafür, dass die Viren-Antigene in den drei Systemen sehr ähnlich sind.

Determination of rubidium in human serum

Field desorption mass spectrometry (FD-MS) combined with stable isotope dilution has been used to determine rubidium concentrations from human serum. Samples obtained from 110 healthy volunteers (50 males, 60 females) were examined. The rubidium concentrations found varied from 0.96 to 3.56 mumoles/l, the average value being 1.96 mumoles/l. The precision of the measurements within a batch was 0.8%. The time for 1 analysis, including sample preparation is about 30 min, the total sample consumption is 100-200 mul. The corresponding potassium concentrations were also determined from all these serum samples; a weak trend towards higher potassium levels with increasing rubidium concentration is found.     

Formation of anaphylatoxin in human serum

Zusammenfassung Es ist möglich, auch in menschlichem Serum eine Anaphylatoxinbildung (AT) durch Kontaktaktivierung oder Kobragift zu induzieren. Wegen der geringen Mengen, die entstehen, muss das wirksame Prinzip vor dem biologischen Nachweis angereichert werden. Menschliches AT verhält sich in allen untersuchten Eigenschaften wie AT aus anderen Plasmaarten. Es unterscheidet sich von dem darmkontrahierenden Spaltprodukt aus der menschlichen Komplementkomponente C3, das mithin nicht als AT angesprochen werden kann.     

Detection of theH-RAS oncogene in human thyroid anaplastic carcinomas

Summary We have transfected high-molecular-weight DNA from human thyroid carcinomas into murine 3T3 cells. As a result we identified several foci of morphologically distinct transformed cells in each of the tumour DNA transfected cultures. After a total of three rounds of transfection, the transformed cells were shown to form tumours in nude mice. Southern blot analysis of DNA prepared from third-round transfectants demonstrated the presence of human Alu repetitive sequences and, after hybridization with probes for known oncogenes, indicated the presence of the humanH-RAS oncogene in 3T3 cells transfected with three out of four anaplastic carcinoma DNA samples. It appears therefore that activation ofRAS genes may be an important event in the development of the anaplastic thyroid tumours.
