Résumé La stabilité de l'iodure d'argent a été étudiée in vivo, par rapport à l'élimination de l'argent et du iode radioactif. Le foie semble responsable de la désiodation. Dans le foie, au bout de 6 h, 83% de l'iode est eliminé, mais seulement 16% de l'argent. Les expériences in vitro indiquent que le facteur responsable de la désiodation est thermostable et plus concentré dans le foie et dans le sang.  相似文献

Development and application of therapeutic oligonucleotides rely on proper analysis of binding and uptake. We have used several model oligodeoxynucleotides (ODNs) to analyze binding/uptake by rat and human leukocytes. Here we describe: (1) differences between in vivo and in vitro uptake of ODNs to rat leukocytes, (2) differences after injection of lipopolysaccharide (LPS), (3) large in vitro differences between primary mononuclear cells in PBS, plasma and blood, and (4) differences of ODN uptake between rat and human leukocytes. Our data show that ODN uptake by primary blood cells was different in PBS, plasma and blood. In addition, LPS treatment increased ODN uptake by leukocytes in blood, indicating that pathological conditions may influence ODN uptake. Furthermore, ODN uptake in rat and human blood is also different, suggesting that preclinical ODN uptake data from rat blood cannot easily be extrapolated to the human condition. Zusammenfassung Es wird mit indirekter Immunofluoreszenz gezeigt, dass Leberzellkerne mit Antikörpern bedeckt werden können, wenn sie systemischemLupus erythromatosus-Serum (SLE) für weniger als 2 sec ausgesetzt werden, und dass die Färbungsintensität vom Titer des Antikörpers abhängt.Diese Ergebnisse deuten darauf hin, dass das in SLE-Geweben durch direkte Immunofluoreszenz nachgewiesene nukleare-Globulin hauptsächlich nukleare post-Biopsiereaktionen wiederspiegelt und weniger in vivo-Reaktionen, und dass in beiden Fällen nur abgestorbene oder nicht intakte Zellen vom Antikörper durchdrungen werden.

The effects of cholesterol and cholesterol-derived oxysterols (cholestanone, cholestenone, coprostanone and epicoprostanol) on adipocyte ghost membrane fluidity were studied using a fluorescence depolarization method. The fluorescence anisotropy of the treated membranes was determined using 1,6-diphenyl-1,3,5-hexatriene (DPH) and 1-(4-trimethylammoniumphenyl)-6-phenyl-1,3,5-hexatriene (TMA-DPH). Cholestanone and cholesterol decreased membranes fluidity at both the concentrations tested (10 & 50 M) while the rest of the sterols did not exert any significant effect on membrane fluidity. In the presence of epinephrine, cholestanone partitioned more towards the lipid core but cholesterol partitioning was not affected. The fusion activation energies (E) obtained for membranes preincubated with cholestanone (8.6 kcal/mol) and cholesterol (8.2 kcal/mol) were not significantly different from that of untreated membranes (8.3 kcal/mol). Membranes preincubated with cholestanone and cholesterol did not exhibit any change in lipid phase throughout the temperature range (10–45°C) tested. The sterols were found to inhibit fisetin-induced phospholipid methylation in isolated rat adipocytes in the rank order of cholesterol > epicoprostanol > cholestanone=cholestenone=coprostanone, while basal methylations was unaffected. When adipocytes were preincubated with the sterols before the addition of fisetin, cholestanone and cholestenone showed 74% and 66% inhibition of maximal methylation respectively. These results indicated that cholesterol oxysterols interact differently with rat adipocyte membranes, with cholestanone interacting more with phospholipids located at the inner lipid bilayer (e.g. phosphatidylethanolamine) while cholesterol interacts more with phosphatidylcholine located at the outer lipid bilayer. This differential interaction may cause selective changes in membrane fluidity at different depths of the bilayer and thus may modulate the activities of membrane-bound proteins such as enzymes and receptors.  相似文献

Summary Glucagon infusion caused a marked increase of brown fat venous FFA concentration, but the extent of increase was less in cold-acclimatized rats than in warm-acclimatized ones. Systemic venous FFA was not appreciably influenced by glucagon. Zusammenfassung Junge, ausgewachsene weibliche Ratten nahmen nach 8 Wochen fettreicher Diät mehr Fett auf als solche mit einer fettarmen Diät, aber gleichviel Kalorien. Bei Weibchen mit körperlicher Bewegung war der Cholesteringehalt des Blutes und des Gewebes höher als bei eingesperrten, sitzenden Tieren.
