噬菌体展示蛋白质芯片的无标记检测

为获得丰富的特异识别分子和实现无标记、高灵敏度检测,利用噬菌体展示技术,选择在pⅢ蛋白上展示12肽的噬菌体M13作探针,采用羧基-氨基共价耦联法制备噬菌体展示蛋白质芯片.在此基础上,与表面等离子体共振传感技术结合,使用Biacore3000仪器,检测噬菌体展示蛋白质芯片与特异性抗体的反应,测定反应动力学常数.实验结果表明:当抗体浓度为0.4 μmol·L-1时,响应信号可达365±8个单位;不同浓度的抗体与芯片反应的数据较好地与S型曲线吻合.该方法可望用于噬菌体展示蛋白质芯片的检测.     

人源噬菌体抗体库的构建及抗D-二聚体抗体的获得

目前国际和国内筛选蛋白所用的技术主要是噬菌体展示技术和酵母双杂交技术。噬菌体展示技术在抗体的筛选方面已显示出良好的应用前景。本实验应用噬菌体展示技术展示出血栓病人的抗体，形成对血栓病人的抗体库。并从中淘选出能与血栓降解产物之一的D-二聚体有特异结合作用的抗体Fabp13。抗D-二聚体的Fab在XL1-Blue中得到可溶表达。再利用抗D-二聚体抗体进行竞争ELISA检验，证实p13具有抗D-二聚体的活性。从而我们筛选得到了抗D-二聚体抗体Fab p13。     

噬菌体筛选技术筛选与联萘酚(BINOL)相结合的抗体

利用噬菌体展示技术筛选只结合1种对应体的BINOL抗体,建立人类单链抗体(scFv)噬菌体文库--Griffin.1文库,以固相化抗原淘筛抗体库,ELISA鉴定噬菌体抗体.从经过4轮富集的次级抗体库中挑选到噬菌体克隆,用该克隆制备的单链抗体阳性克隆,经ELISA证实具有良好的抗原特异性.从人类scFv噬菌体文库中获得抗BINOL的特异性抗体.     

轮状病毒间接免疫荧光的检测方法

以兔抗SA11多克隆抗体为一抗、 Alexa 488标记的山羊抗兔抗体为二抗, 通过对反应条件的优化建立轮状病毒（RV）检测的间接免疫荧光法（IFA）. 实验结果表明, IFA最佳工作条件为: MA104细胞密度为每孔2×10⁴个, 胰酶活化轮状病毒质量浓度为1 μg/mL, 培养时间为18 h, 一抗最适稀释度为1∶800, 二抗最适稀释度为1∶500.     

抗克伦特罗噬菌体单链抗体库的构建、筛选及鉴定

利用噬菌体展示技术构建克伦特罗(CBL)单链抗体(scFv)库,从中筛选CBL特异性噬菌体scFv,从而成功扩增出抗CBL的VL,VH基因片段并采用重叠延伸PCR拼接为全长的scFv基因片段,抗体库库容约为1.6×104,经4轮吸附—洗脱—扩增的富集,采用酶联免疫吸附测定(ELISA)法筛选到6个具有CBL结合活性的噬菌体scFv,为进一步大量表达CBL单链抗体奠定了基础.     

多头绒泡菌线粒体中肌动蛋白的免疫细胞化学鉴定

自多头绒泡菌(Physarum polycephalum Schw.)原质团中分离线粒体,以兔抗肌动蛋白抗体为一抗,荧光素标记的羊抗兔IgG和辣根过氧化物酶标记的羊抗兔IgG作二抗进行间接免疫荧光和蛋白质免疫印迹实验.结果显示,线粒体中含有与肌动蛋白抗体呈阳性反应的抗原.以兔抗肌动蛋白抗体作一抗,金颗粒标记的蛋白A作二抗进行的间接免疫电子显微镜实验结果进一步证明了肌动蛋白是多头绒泡菌线粒体的组成成分,并在线粒体中呈散在分布.     

鼠源抗AFB1噬菌体单链抗体库的构建与鉴定

研究以AFB1-BSA免疫的小鼠脾脏细胞为试验材料,利用RT-PCR技术,克隆了抗AFB1抗体重链和轻链可变区基因VH和VL,利用连接肽(Gly4Ser)3将VH和VL链接成单链抗体基因scFv,通过噬菌体展示载体pCANTAB-5E携带将其电转化E.coli TG1,经氨苄青霉素平板筛选,构建了库容为2.13×109 cfu/μg DNA的噬菌体单链抗体库,抗体库的克隆阳性率达到100%,且多样性良好,为高活性抗AFB1单链抗体的筛选提供了材料基础。     

人源性抗HBc单链噬菌体抗体库的构建

利用RT－PCR和噬菌体表面展示技术，直接从乙肝病毒核心抗体（抗HBc）阳性患淋巴细胞中提取总RNA，反转录成cDNA；合成全套人抗体可变区引物扩增抗体可变区基因，并将重、轻链可变区基因进行拼接装配成单链抗体（ScFv）基因，重组于噬菌粒载体pHEN1，转化抑制型大肠杆菌E.coliTG1，以辅助噬菌体援救后，构建成人源性单链噬菌体库。库容量达106。     

利用十二肽库筛选心肌型脂肪酸结合蛋白特异性的亲和配体

采用噬菌体表面展示十二肽库对心肌型脂肪酸结合蛋白（H FABP）特异性亲和配体进行筛选, 经3轮筛选及序列比对后得到一个酶联免疫测定分析 (ELISA)检测阳性特征序列： W P N H H M L H K R W P. 对此序列进行肽合成, 并标记上荧光标记物芘, 经高效液相纯化、 冻干及质谱确定其分子量后制成荧光肽探针检测急性心肌梗塞病人血样, 结果均呈阳性. 结果表明, 所获得的肽探针具有较好的临床应用效果.心肌型脂肪酸结合蛋白; 噬菌体表面展示十二肽库; 亲和配体; 酶联免疫测定分析     

一种适用于教学的酶联免疫吸附试验的抗原抗体系统制备

目的:探讨设计一种教学成本低、便于教学使用、有助于培养医学生综合实验设计能力和提高实验基本操作技能的酶联免疫吸附实验抗原抗体系统.方法:以提纯的人IgG免疫家兔获取兔抗人IgG抗体,再用其包被酶标反应板,以人血清(人IgG)作抗原,用酶标记羊抗人IgG抗体结合抗原及催化底物显色反应.结果:该双抗体夹心法,底物显色反应清楚肉眼即可作出判定,非特异干扰极小.结论:我们设计的酶联免疫吸附实验-双抗体夹心法原理清晰、便于理解和掌握、教学成本较低、结果易于观察、便于教学大量使用.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
