Treatment with the co-transport inhibitor, furosemide decreased36Cl flux across perfused Malpighian tubules ofLocusta. However, exclusion of36Cl from the bathing medium had not effect on22Na+ flux whereas substitution of bathing medium Na+ by K+ increased36Cl flux. Summary A comparative account of numerical chromosome variation in 2× and 4× callus cultures ofHyoscyamus muticus examined at monthly intervals reveals that both types of calli tend to attain a genomic flux around 4×. Parallels apparently exist between the course of ploidy change and chromosomal flow in culture and evolutionary processes in nature, suggesting the utility of the in vitro system as a rapid approach for recognizing trends in genome evolution.  相似文献

Summary An SH reactive agent, N-ethyl maleimide (10–3 M for 2 min in the luminal fluid) selectively inhibits the urea transpithelial flux across the toad gall bladder. Acetyl-coenzyme A synthetase (AMP forming; Acs) is an enzyme whose activity is central to the metabolism of prokaryotic and eukaryotic cells. The physiological role of this enzyme is to activate acetate to acetyl-coenzyme A (Ac-CoA). The importance of Acs has been recognized for decades, since it provides the cell the two-carbon metabolite used in many anabolic and energy generation processes. In the last decade researchers have learned how carefully the cell monitors the synthesis and activity of this enzyme. In eukaryotes and prokaryotes, complex regulatory systems control acs gene expression as a function carbon flux, with a second layer of regulation exerted posttranslationally by the NAD+/sirtuin-dependent protein acetylation/deacetylation system. Recent structural work provides snapshots of the dramatic conformational changes Acs undergoes during catalysis. Future work on the regulation of acs gene expression will expand our understanding of metabolic integration, while structure/function studies will reveal more details of the function of this splendid molecular machine.Received 4 December 2003; received after revision 2 March 2004; accepted 16 March 2004  相似文献

The duck-billed platypus, or Ornithorhynchus, was the subject of an intense debate among natural historians in the late eighteenth and early nineteenth centuries. Its paradoxical mixture of mammalian, avian and reptilian characteristics made it something of a taxonomic conundrum. In the early 1820s Robert Jameson (1774–1854), the professor of natural history at the University of Edinburgh and the curator of the University's natural history museum, was able to acquire three valuable specimens of this species. He passed one of these on to the anatomist Robert Knox (1791–1862), who dissected the animal and presented his results in a series of papers to the Wernerian Natural History Society, which later published them in its Memoirs. This paper takes Jameson's platypus as a case study on how natural history specimens were used to create and contest knowledge of the natural world in the early nineteenth century, at a time when interpretations of the relationships between animal taxa were in a state of flux. It shows how Jameson used his possession of this interesting specimen to provide a valuable opportunity for his protégé Knox while also helping to consolidate his own position as a key figure in early nineteenth-century natural history.  相似文献

Summary Microorganisms are useful model systems for the study of intracellular transport of lipids. Eukaryotic microorganisms, such as the yeastSaccharomyces cerevisiae, are similar to higher eukaryotes with respect to organelle structure and membrane assembly. Experiments in vivo showed that transport of phosphatidylcholine between yeast microsomes and mitochondria is energy independent; transfer of phosphatidylinositol to the plasma membrane and the flux of secretory vesicles take place by different mechanisms. Linkage of transfer and biosynthesis of phospholipids was demonstrated in the case of intramitochondrial phospholipid transfer. A yeast phosphatidylinositol/phosphatidylcholine transfer protein, which is essential for cell viability, was isolated and characterized. Another phospholipid transfer protein present in yeast cytosol, which has a different specificity, is currently under investigation. Transfer of phospholipids between cellular membranes was also demonstrated with prokaryotes. The cytoplasm and the periplasma of the gram-negative facultative photosynthetic bacteriumRhodopseudomonas sphaeroides contain phospholipid transfer proteins; these seem to be involved in the biosynthesis of prokaryotic membranes.  相似文献

The PAS domain kinase PASKIN, also termed PAS kinase or PASK, is an evolutionarily conserved potential sensor kinase related to the heme-based oxygen sensors of nitrogen-fixing bacteria. In yeast, the two PASKIN homologs link energy flux and protein synthesis following specific stress conditions. In mammals, PASKIN may regulate glycogen synthesis and protein translation. Paskin knock-out mice do not show any phenotype under standard animal husbandry conditions. Interestingly, these mice seem to be protected from the symptoms of the metabolic syndrome when fed a high-fat diet. Energy turnover might be increased in specific PASKIN-deficient cell types under distinct environmental conditions. According to the current model, binding of a putative ligand to the PAS domain disinhibits the kinase domain and activates PASKIN auto- and target phosphorylation. Future research needs to be conducted to elucidate the nature of the putative ligand and the molecular mechanisms of downstream signalling by PASKIN. Insect flight is the most energy-demanding exercise known. It requires very effective coupling of adenosine triphosphate (ATP) hydrolysis and regeneration in the working flight muscles.31P nuclear magnetic resonance (NMR) spectroscopy of locust flight muscle in vivo has shown that flight causes only a small decrease in the content of ATP, whereas the free concentrations of inorganic phosphate (P i ), adenosine diphosphate (ADP) and adenosine monophosphate (AMP) were estimated to increase by about 3-, 5- and 27-fold, respectively. These metabolites are potent activators of glycogen phosphorylase and phosphofructokinase (PFK). Activation of glycolysis by AMP and P i is reinforced synergistically by fructose 2,6-bisphosphate (F2,6P2), a very potent activator of PFK. During prolonged flight locusts gradually change from using carbohydrate to lipids as their main fuel. This requires a decrease in glycolytic flux which is brought about, at least in part, by a marked decrease in the content of F2,6P2 in flight muscle (by 80% within 15 min of flight). The synthesis of F2,6P2 in flight muscle can be stimulated by the nervous system via the biogenic amine octopamine. Octopamine and F2,6P2 seem to be part of a mechanism to control the rate of carbohydrate oxidation in flight muscle and thus function in the metabolic integration of insect flight.Dedicated to Dr. Ernst Zebe, Emeritus Professor of Zoology (University of Münster) on the occasion of his 70th birthday.  相似文献

A key statement of the 1965 Monod-Wyman-Changeux (MWC) model for allosteric proteins concerns the distinction between the ligand-binding function ( ) and the relevant state function ( ). Sequential models predict overlapping behavior of the two functions. In contrast, a straightforward experimental consequence of the MWC model is that for an oligomeric protein the parameters which characterize the two functions should differ significantly. Two situations, where \bar Y$$ " align="middle" border="0"> and the system ishyper-responsive or where and the system ishypo-responsive, have been encountered. Indeed, the hyper-responsive pattern was first observed for the enzyme aspartate transcarbamoylase, by comparing with monitored by a change in sedimentation. Extensions of the theory to ligand-gated channels led to the suggestion that, on the one hand, hyper-responsive properties also occur with high-affinity mutants. On the other hand, native channels of the acetylcholine neuronal7 receptor and low-affinity mutants of the glycine receptor can be interpreted in terms of the hypo-responsive pattern. For the ligand-gated channels, whereas is detected directly by ion flux, ligand binding has rarely been measured and the formation of desensitized states may complicate the analysis. However, stochastic models incorporating both binding and channel opening for single molecules predict differences that should be measurable with new experimental approaches, particularly fluorescence correlation spectroscopy.  相似文献

X-ray astronomy began in 1948 with the first detection of X-rays from the Sun. Astronomical X-ray observations need to be performed from high-altitude rockets and satellites because the Earth's atmosphere absorbs X-rays. Currently about 100,000 X-ray sources are known all over the sky. The Sun is by far the strongest source. The outermost solar atmosphere, the corona, emits X-rays due to its high temperature of a few million K. Solar X-ray emission is highly variable. Eruptions lead to variations of the X-ray flux on time scales of minutes. The average X-ray flux varies with the 11-year sunspot cycle by a factor of about 1000. Solar X-rays have a profound influence on the Earth's upper atmosphere.  相似文献

Résumé Un modèle cinétique simple a été développé pour l'épiderme; les lois cinétiques de premier ordre peuvent lui être appliquées fructueusement. Il est envisagé que la régulation homéostatique de l'épiderme peut se faire par l'ajustement des quantités de nourriture qui lui parviennent. Le flux nutritif atteignant l'épiderme est composé du flux de diffusion, qui prédomine normalement, et du flux actionné par l'évaporation, à la surface de la peau, de l'eau provenant de l'ultra-filtrat du sérum. Ce dernier pourrait prédominer quand l'épiderme aurait été lésé, et serait augmenté quand la peau ne constituerait pas une barrière suffisante au passage de l'eau.Par analogie avec les cultures de microbes en «steady state», il est envisagé que le nombre total de cellules épidermiques par unité de surface cutanée et leur taux de croissance sont des fonctions du taux du transfert de nourriture à l'épiderme. L'équation hyperbolique deMonod est donc modifiée et appliquée à l'épiderme. Une évaluation approximative de cette équation suggère que le flux nutritif vers l'épiderme est augmenté en cas de psoriasis aussi bien que dans la peau normale lorsqu'elle est dépouillée du stratum corneum. On considère que dans le psoriasis le dérangement cinétique de l'épiderme et le dérangement fonctionnel de la barrière de la peau au passage de l'eau sont intimement liés.

Recent discoveries revealing that carbohydrate modifications play critical roles in a wide variety of biological processes have brought wide recognition to the field of glycobiology. Growing attention has focused on the function of unusual O-linked carbohydrate modifications such as O-fucose. O-fucose modifications have been described in several different protein contexts, including epidermal growth factor-like repeats and thrombospondin type 1 repeats. The O-fucose modifications on thrombospondin type 1 repeats have only recently been described, but the site of modification occurs in a region proposed to play a role in cell adhesion. O-fucose modifications on epidermal growth factor-like repeats have been described as important players in several signal transduction systems. For instance, Notch, a cell-surface signaling receptor required for many developmental events, bears multiple O-fucose saccharides on the epidermal growth factor-like repeat of its extracellular domain. The O-fucose moieties serve as a substrate for the β1,3 N-acetylglucosaminyltransferase activity of Fringe, a known modifier of Notch function. The alteration of O-fucose structures by Fringe influences the ability of Notch ligands to activate the receptor and provides a means to regulate Notch signaling. Thus, O-fucose and Fringe provide a clear example of how carbohydrate modifications can have direct functional consequences on the proteins they modify. RID="*" ID="*"Corresponding author.  相似文献
