Degradation of type I collagen fibrils synthesized by human dental pulp cells in explant culture exposed toActinomyces viscosus. Electron microscope immunotyping

Summary Actinomyces viscosus Be 66, added to pulpal cells in culture, does not cause apparent cellular damage. The extracellular matrix consists of altered collagen fibrils and thin filaments, immunochemically identified as type I collagen. Dense core vesicles in cerebral cortex of the human fetus.

The present study clearly demonstrates that by the 15th week of gestation dense core vesicles appear within the human cerebral cortex. These vesicles can be identified within axon cylinders, axon growth cones, and axon synaptic terminals. The role of these vesicles is speculative, yet, their very presence at this early fetal stage seems to reflect an advanced state of synaptic vesicle development.

Zusammenfassung Einige proteolytische Fermente (wie Pronase, Pepsin und Trypsin) beweisen eine beschränkte Wirkung auf das Kollagenmolekül. Die Wirkung dieser Fermente wurde durch die Aminosäuresequenz, die für den Hauptteil des Kollagenmoleküls typisch ist (das heisst Gly.Pro.X), beschränkt. Dieser Typ der Sequenz wurde als C-Endsequenz im Restkollagen nach der Pronasewirkung bestimmt.

Studies on the structure of collagen V. The site of binding of trivalent iron on collagen

Zusammenfassung Die Reaktion zwischen Kollagen und Fe^III wurde studiert. Es zeigte sich, dass das Eisen an die Sequenzen des Typs Ala-Asp-Gly gebunden wird. Weiter wurde gefunden, dass 1 mol Ratten-Tropokollagen 1 mol Cystein als Sequenz Cys-Ala-Asp-Gly enthält.

A cell culture substrate obtained from heat-fused collagen.

Coverslips coated with rat-tail collagen dried at 37 degrees C were placed in a hot-air sterilizing oven at 160 degrees C for 2 h. The resulting transparent sterile film was found to be a useful multipurpose substrate for cell culture and for subsequent histological sectioning.     

Electronmicroscopic identification of type C particles in cultured murine neuroblastoma.

Monolayer of murine neuroblastoma were treated with dexamethasone and examined by electronmicroscopy. Most of the treated cells were morphologically differentiated and exhibited type C virus particles which were budding from the cell surface. This in vitro system may be of great value for exploring the oncogenic potential of the virus, and its possible role in cell differentiation.     

Distribution of hydroxyproline and hydroxylysine deficient collagen in individual collagen fractions in the granuloma tissue

Zusammenfassung Vom atypischen C¹⁴-Kollagen, das unter der Wirkung von 0.5 mM 2,2-Dipyridyl im karrageenen Granulationsgewebe entstanden ist, wurden neutralsalzlösliche, säurelösliche und unlösliche Fraktionen isoliert. Die Erscheinung der Radioaktivität in der unlöslichen Fraktion beweist, dass atypisches Kollagen von Zellen transportiert wird und im extrazellulären Raum aggregieren kann. Die niedrigere spezifische Aktivität des säurelöslichen Kollagens zeigt, dass es sich um ein Abbauprodukt des unlöslichen Kollagens handelt.     

Inhibition of collagen synthesis by interleukin-1 in three-dimensional collagen lattice cultures of fibroblasts

Interleukin-1 (Il-1) was added to collagen lattice cultures of human skin fibroblasts. No cell division was induced, the ability of fibroblasts to contract the lattices was decreased and a dose-related inhibition of collagen synthesis without effect on non-collagen proteins was found. Indomethacin had no influence on these effects.

The role of proteolytic processing in the morphogenesis of virus particles.

Proteinases are encoded by many RNA viruses, all retroviruses and several DNA viruses. They play essential roles at various stages in viral replication, including the coordinated assembly and maturation of virions. Most of these enzymes belong to one of three (Ser, Cys or Asp) of the four major classes of proteinases, and have highly substrate-selective and cleavage specific activities. They can be thought of as playing one of two general roles in viral morphogenesis. Structural proteins are encoded by retroviruses and many RNA viruses as part of large polyproteins. Their proteolytic release is a prerequisite to particle assembly; consequent structural rearrangement of the capsid domains serves to regulate and direct association and assembly of capsid subunits. The second general role of proteolysis is in assembly-dependent maturation of virus particles, which is accompanied by the acquisition of infectivity.
