In a first experiment, cells were cultured in media supplemented by nickel sulphate, irradiated in same media and cultured in same media after irradiation. In a second experiment, cells were cultured during 18 hrs. in media supplemented by nickel sulphate, and then cells were washed and cultured in normal media where they were irradiated. The nickel sulphate toxicity appears as a creasing function of the nickel sulphate concentration and the nickel sulphate action endurance. The nickel sulphate toxic effect is amplified by X-Rays. This amplification is a time function that depends on the X-Ray dose, nickel sulphate concentration and period of time from the outset of culture to the irradiation. The nickel sulphate toxic effect appears faster when nickel works after X-Rays.  相似文献

Nickel is titrated by atomic absorption in nuclear and non-nuclear fractions of cells cultured in media supplemented by nickel sulphate. The living cell percentages are estimated in the same culture conditions. We try to connect the nickel presence in cellular fractions and the living cell percentages. These used cells seem to have mortality probability when nuclear fraction has 55 x 10(-6) of nickel and non-nuclear fraction has 33 x 10(-6) of nickel. We discuss the nickel stimulating effect for the start of DNA synthesis and the nickel penetration into cells.  相似文献

Nickel enzymes are a relatively new class of metalloenzymes. The seven known nickel enzymes are urease, hydrogenase, CO-dehydrogenase, methyl-coenzyme M reductase, Ni-superoxide dismutase, glyoxalase I and cis-trans isomerase. The requirement for nickel implies the presence of a nickel-processing system, since free transition metals are harmful to the cell. A nickel-processing system involves the recognition and transport of nickel into the cell and the handling of the nickel once it enters the cell until it is inserted into the nickel enzyme. Several mechanisms for nickel transport have been identified and will be reviewed here. Accessory proteins required for the biosynthesis of the nickel active site have been identified. Accessory proteins bind the nickel when it enters the cell and are proposed to assist with the insertion of nickel into the enzyme. The function of the characterized nickel-processing proteins is described, and models for nickel insertion into the nickel enzymes are presented.  相似文献

Summary Sulphated proteoglycans were isolated using a dissociative method of extraction. Cerebrum accounted for the major portion of proteochondroitin sulphate in brain, whereas the brain stem fraction contained over half the proteoheparan sulphate. Summary Snails (Helix aspersa) contain sulphate-reducing bacteria in their crops. Feeding such animals on food containing sulphate and molybdate ions does not induce a copper deficiency and in fact the bacteria appear to facilitate metal absorption. Summary Proteins secreted by the rat submandibular gland after administration of autonomic agonists have been fractionated by sodium dodecyl sulphate (SDS) — polyacrylamide gel electrophoresis. α-adrenoceptor, β-adrenoceptor and muscarinic agonists were all found to cause the secretion of different protein populations. Summary L-Aspartic acid was injected into a lateral cerebral ventricle of the sheep at ambient temperatures between 0° and 40°C. Doses of 100 or 500 nmoles·kg–1 caused a rise in heat production and/or a decrease in heat loss; rectal temperature rose. Atropine sulphate attenuated or prevented these effects.  相似文献

In addition to several anomalous structures, other general forms of definitely rod-shaped microorganisms have been found by scanning and transmission electron microscopy in the lung tissue taken at autopsy from a patient who succumbed to confirmed Legionnaires' disease with extensive necrotizing lobar pneumonia. The microorganisms were greatly varied in size and shape. They were micrographed in the act of fission. These forms have been found to some extent throughout the tissue. No nickel was demonstrated, either in the lung tissue or in the microorganisms.  相似文献

Syndecans are a family of integral membrane proteoglycans with conserved membrane-spanning and intracellular domains but with structurally distinct extracellular domains (ectodomains). They are known to function as heparan sulphate co-receptors in fibroblast growth factor signalling as well as to link cells directly to the extracellular matrix. These and other biological activities of syndecans involve specific interactions of the heparan sulphate side chains of syndecans with cytokines and extracellular matrix proteins. Four different vertebrate syndecans, designated as syndecans 1–4 (or syndecan, fibroglycan, N-syndecan and amphiglycan, respectively), are known. During embryonic development, syndecans have specific and highly regulated expression patterns that are distinct from the expression in adult tissue, suggesting an active role in morphogenetic processes. The developmental expression of syndecans is particularly intense in mesenchymal condensates and at epithelium mesenchyme interfaces, where a number of heparan sulphate-binding cytokines and matrix components are also expressed in a regulated manner, ofter spatially and temporally co-ordinated with the syndecan expression. Recent evidence indicates that the regulation of heparan sulphate fine structure (mainly the number and arrangement of sulphate groups along the polymer) provides a mechanism for the cellular control of syndecan-protein interactions. Furthermore, morphogenetically active cytokines such as fibroblast growth factor-2 and transforming growth factor-β participate in the regulation of syndecan expression and glycosaminoglycan structure. This review discusses the developmental expression and binding functions of syndecans as well as the molecular regulation of specific heparan sulphate-protein interactions.  相似文献

The distribution of eight components of the extracellular matrix in immature human placenta was studied by an indirect immunofluorescence method with monospecific antibodies. In the stroma of the term chorionic villi, collagen types I, III, IV, V, and fibronectin formed a mesh of fibers and conglomerates. Heparan sulphate proteoglycan formed multiple conglomerates, whereas laminin comprised small, scanty, discrete granules. Collagen type IV, laminin, entactin, and heparan sulphate proteoglycan were confined to the basement membrane of the trophoblast. Sometimes, only collagen type IV was identified in fetal vascular basement membrane.  相似文献

The range of (presumed) polyketide constituents in comatalid crinoids has been extended to include bianthronyls, meso-naphthodianthrones and sulphate esters of various of the polyketides. Several species of fish are deterred from eating food by the inclusion in it of these sulphates at the concentration found in crinoids. Sodium 2-hydroxy-anthraquinone sulphate and anthraquinone-2-suphonate were also active as antifeedants for the species tested.  相似文献

Following the suppression of renewal of rat taste cells by vinblastine sulphate, the preference for sucrose decreased markedly while the aversion to quinine did not change. Molecular genetic analyses of osteochondrodysplasias in mice and humans have recently led to the identification of mutations in genes encoding structural proteins growth factor receptors and sulphate transporters. Further analyses of such inherited disorders, using rapid techniques for gene mapping, positional cloning and mutation detection, will undoubtedly uncover other genes that are important for skeletal development. Together with studies of transgenic mice, in which specific genes that are expressed in the skeleton are mutated, these analyses will provide insight into genes that are essential for skeletal morphogenesis.  相似文献

Bright light (2000-3000 lux) of sufficient intensity to suppress human melatonin secretion, acts as a strong zeitgeber in the entrainment of circadian rhythms in man. In polar conditions, light of this intensity is not experienced for several weeks during the winter. The entrainment of human circadian rhythms, in particular that of melatonin, is clearly of interest in these circumstances. Urinary 6-hydroxy melatonin sulphate (aMT6s) is a good index of melatonin secretion in man. In a limited study of seven male volunteers living on an Antarctic base the overall 24-h rhythm of aMT6s excretion was maintained at four different times of year (spring, summer, autumn and winter) and no significant seasonal effects were noted. Cortisol excretion, appeared to be markedly affected by the season although other factors such as social and environmental stress cannot be discounted. These observations suggest that in the absence of a strong light-dark cycle melatonin production may be entrained by other factors.  相似文献

Keratan sulphate (KS) is the predominant glycosaminoglycan (GAG) in the cornea of the eye, where it exists in proteoglycan (PG) form. KS-PGs have long been thought to play a pivotal role in the establishment and maintenance of the array of regularly-spaced and uniformly-thin collagen fibrils which make up the corneal stroma. This characteristic arrangement of fibrils allows light to pass through the cornea. Indeed, perturbations to the synthesis of KS-PG core proteins in genetically altered mice lead to structural matrix alterations and corneal opacification. Similarly, mutations in enzymes responsible for the sulphation of KS-GAG chains are causative for the inherited human disease, macular corneal dystrophy, which is manifested clinically by progressive corneal cloudiness starting in young adulthood.  相似文献
