Summary A blue carotenoid-protein complex (max 635 nm) was extracted and purified from the carapace of the crayfishProcambarus clarkii. The complex was further liberated from astaxanthin, its prostetic group, causing dissociation into apoprotein subunits. Reconstitution of the complex from the various sub-units (isolated by chromatofocusing) plus astaxanthin was attempted. Apoprotein-size pigments of rose-purple color (max 545 nm) were obtained. It was found that both monomers are required in order to a blue complex fairly similar in structure ot the native one. However, the native conformation was not completely recovered, as indicated by some differences in the UV spectrum.  相似文献

Despite knowledge of the existence of the pigment called scytonemin for over 100 years, its structure has remained unsolved until now. This pigment, the first shown to be an effective, photo-stable ultraviolet shield in prokaryotes, is a novel dimeric molecule (molec. wt. 544) of indolic and phenolic subunits and is known only from the sheaths enclosing the cells of cyanobacteria. It is probable that scytonemin is formed from a condensation of tryptophan-and phenylpropanoid-derived subunits. The linkage between these units is unique among natural products and this novel ring structure is here termed the scytoneman skeleton. Scytonemin absorbs strongly and broadly in the spectral region 325–425 nm (UV-A-violet-blue, with an in vivo maximum at 370 nm). However, there is also major absorption in the UV-C ( max=250nm) and UV-B (280–320 nm). The pigment has been recently shown to provide significant protection to cyanobacteria against damage by ultraviolet radiation. The pigment occurs in all phylogenetic lines of sheathed cyanobacteria and possibly represents a UV screening strategy far more ancient than that of plant flavonoids and animal melanins. How diverse organisms deal with UV radiation is considered of vital importance to global ecology.  相似文献

Summary The anthocyanidin investigated here exists below pH 4 as a cationAH 2 + (wave length of absorption maximum max=459 mµ), between pH 5 and 7 in the neutral formAH ( max=492 mµ) and above pH 8 as an anionA ( max=537 mµ). At pH 5 the freshly dissolved substance is partially converted into a colourless formBH ( max=372 mµ) and a chemical equilibrium betweenAH andBH is reached within 1 h. A kinetic study of the process of formation ofBH shows that an intermediate productZ is formed. This process can be reversed by light exposure. It can be concluded from a kinetic investigation by using flash light thatBH is transformed by the absorbing light into a new substanceL ( max 275 mµ and 225 mµ), andL changes partially intoBH, partially intoZ, which itself is transformed partially intoAH 2 + ,AH,A , partially intoBH. The reactionZ AH,AH 2 + ,A is proportional to the concentration of protons [H +], the reactionZ BH independent of [H +]. Thus a photochemical production ofAH 2 + ,AH,A fromBH is readily obtained in the presence ofH + and not obtained in the absence ofH +.  相似文献

Summary Ethanol may modulate endogenous opioid systems by disrupting opioid receptor signalling. Low concentrations of ethanol slightly potentiate -opioid receptor binding by increasing receptor Bmax, and, in some cases, chronic ethanol exposure decreases the density or affinity of the -opioid receptors. By contrast, high concentrations of ethanol acutely decrease -opioid receptor binding by decreasing receptor affinity, whereas chronic exposure of animals and neuronal cell lines to lower concentrations of ethanol leads to possibly adaptive increases in the density or affinity of the -opioid receptors. In the neuronal cell line NG108-15, ethanol does not up-regulate the -opioid receptor by blocking receptor degradation or endocytosis, but protein synthesis is required for this response. Up-regulation of the -opioid receptor renders ethanol-treated NG108-15 cells 3.5-fold more sensitive to opioid inhibition of adenylyl cyclase. Long-term treatment with ethanol also increases maximal opioid inhibition in NG108-15 cells, possibly by decreasing levels of Gs and its mRNA. Ethanol differentially modulates signal transduction proteins in three additional neuronal cell lines, N18TG2, N4TG1, and N1E-115. Ethanol-treated N18TG2 cells show the least up-regulation of the -opioid receptor, little heterologous desensitization of adenylyl cyclase, and no changes in Gs or Gi. By contrast, ethanol-treated N1E-115 cells show the largest up-regulation of the -opioid receptor, the most heterologous desensitization of adenylyl cyclase, and concentration-dependent decreases in Gs and increases in Gi. Further analysis of these related neuronal cell lines may help to identify the molecular elements that endow some, but not all, neuronal cells with the capacity to adapt to ethanol.  相似文献

Zusammenfassung Von den Blutzellen derPhallusia mamillata Cuvier zeigen nur die stark vakuolisierten (compartment cells) im UV-Licht eine stabile blaue Fluoreszenz ( max =5200 und 4600 Å). Sie ist in den Zellgranula lokalisiert. Bei den Blutzellen vonCiona intestinalis L. tritt dagegen eine gelbe Fluoreszenz ( max = 5900, 5200 und 4600 Å) auf, die nur von den Vanadocyten ausgeht. Diese Fluoreszenz, welche auch von den Testazellen der Oocyten gegeben wird, geht innerhalb einiger Minuten in die blaue Fluoreszenz über.

Summary The intensity of sound-induced convulsions in the genetically epilepsy-prone rat (GEPR) was reduced in a dose related fashion by intracerebroventricular administration of dobutamine, (1 agonist), terbutaline (2 agonist) or phenylephrine (1 agonist). BHT-920 (2 agonist) did not cause a dose-related decrease in sound-induced convulsion intensity. Binding studies showed that whole brain and receptor densities (Bmax) were normal while the Kd was increased for the ligand in GEPR brain.Acknowledgment. Summary The conversion of cholesterol into 19-nor-5-cholestan-3-ol by the spongeAxinella polypoides involves a partial loss (40%) of the 3-hydrogen atom; moreover administration to the sponge of [4-14C]cholesterol tritiated at C-4 and C-7 showed that the 4- and 7-hydrogen atoms are retained in this conversion. Summary Critical concentration of unbound cadmium in the rabbit renal cortex was around 13 g Cd/g, which was far lower than the critical concentration of total cadmium in the renal cortex, 200–300 g Cd/g, and a little lower than the critical concentration of non-MT-Cd in the renal cortex, 35–60 g Cd/g, respectively. Summary Scarites buparius has 2n=37 and n=17+X1X2Y. This multiple sex-chromosomes system seems to be the result of a reciprocal translocation between the primitive X-chromosome and an autosome. Summary The author treats 2 topics connected with his studies on the cytogenetics of the African pigmy-mice. (1)Mus (Leggada) bufo Th. shows the primitive complement already detected in other species or subspecies (minutoides ssp 1,indutus, tenellus, setulosus): 2N=36,N.F.=36, sex chromosomesPR. (2) The deletion of the short arm of oneX-chromosome (X dc ) was previously observed as heterozygous mutation by 4 in a sample of 7 and 5 . A more important sample (18 , 21 ) gives the following results: 11 X-X, 10 X-X dc , 18 X-Y. The occurrence of X dc -X dc and of X dc -Y is also very improbable. Summary Studies of the interaction between vinblastine-like alkaloids and their receptor, i.e. tubulin, are reported. They shed some light on the structure-activity relationships in this medicinally important series: the configurations at C14 and C16 as well as the presence of the methoxycarbonyl group on C16 seem to play an essential role in the determination of biological activity.This work was supported by CNRS (A.T.P. Zusammenfassung Die Existenz von zwei verschiedenen isomeren Homoallylkationen in der Umlagerung von Steroid- 5-19-Methansulfonsäure-Estern zu 5, 19-cyclo-6-hydroxylierten Steroiden und deren Umlagerung zu 5(10)-B-homo-7-hydroxylierten Steroiden wurde bewiesen, und eine neue Art der Homoallylumlagerung eines 5(10)-B-homo-7-hydroxylierten Steroids zum 5, 6-Methano-9-en und zum 5, 6-Methano-10-ol wurde beobachtet.
