Cell pairs isolated from adult rat and guinea pig ventricles were used to study the electrical properties of the nexal membrane. Each cell of a pair was connected to a voltage-clamp system so as to enable whole-cell, tight-seal recording. The current-voltage relationship of the nexal membrane was found to be linear, revealing a resistance rn of 2-4 M omega. rn was insensitive to the sarcolemmal membrane potential (range: -90 to +30 mV), and exerted no time-dependent gating behavior (range: 0.1 to 10 s). Lowering pHi yielded a small increase in rn. Vigorous elevations in [Ca2+]i gave rise to an increase in rn which was associated with a cell shortening. Uncoupling caused by aliphatic alcohols or halothane did not produce cell shortening. Cell pairs were also used to study action potential transfer.  相似文献

Summary Internal longitudinal resistance (ri), a determinant of cardiac conduction, is affected by changes in intracellular calcium and protons. However, the role and mechanism by which H+ and Ca2+ may modulate ri is uncertain. Cable analysis was performed in cardiac Purkinje fibers to measure ri during various interventions. In some experiments, intracellular pH (pHi) was recorded simultaneously to study the pHi-ri relation. Both intracellular Ca2+ and H+ independently modified ri. However, internal resistance of cardiac fibers was insensitive to pHi changes compared to other tissues. A latent period preceded the pHi-related changes in ri and the amount of change depended upon methodology. The results suggest that direct action of protons on ri may be subordinate to other regulatory processes. Ionic regulation of internal longitudinal resistance may occur by more than one mechanism: i) direct cationic binding to sites on junctional membrane proteins; and ii) H+- or Ca2+-dependent phosphorylation of junctional proteins.  相似文献

Histamine release from rat peritoneal mast cells induced by anti-IgE was essentially complete within 4–5 min. Xestobergsterol A and B, which are constituents of the Okinawan marine spongeXestospongia bergquistia Fromont, dose-dependently inhibited anti-IgE-induced histamine release from rat mast cells. The IC50 values of xestobergsterol A and B for histamine release in mast cells activated by anti-IgE were 0.07 and 0.11 M, respectively. Anti-IgE stimulated PI-PLC activity in a mast cell membrane preparation. Xestobergsterol A dose-dependently inhibited the generation of IP3 and membrane-bound PI-PLC activity. Moreover, xestobergsterol A inhibited Ca2+-mobilization from intracellular Ca2+-stores as well as histamine release in mast cells activated by anti-IgE. On the other hand, xestobergsterol B did not inhibit the membrane-bound and cytosolic PI-PLC activity, IP3 generation or the initial rise in [Ca2+]i in mast cells activated by anti-IgE. These results suggest that the mechanism of inhibition by xestobergsterol A of the initial rise in [Ca2+]i, of the generation of IP3, and of histamine release induced by anti-IgE, was through the inhibition of PI-PLC activity.  相似文献

Summary Sucrose acts in vitro as a mixed inhibitor of (V+K+n) type towards honeybee hemolymph PNP--D-glucosidase activity. Between the stages of emergence and foraging, there is little change in the effect of the inhibitor on VM (fi=from 1.31 to 1.35 respectively) or n (fi=from 1.09 to 1.07) but K is more markedly affected (fi –1=from 1.17 to 1.87)M. These observations reflect the decrease of Ki from 277 to 98 mM and of I50 from 154 to 111 mM, but Ki scarcely alters during development (from 477 to 425 mM). These inhibitory effects of sucrose are intermediate between those previously reported for trehalose and glucose.Acknowledgments. Particular appreciation is due to Mrs C. Pyrophosphate (PPi) is a non-hydrolyzable ATP analogue known to affect the binding between myosin heads and actin. By using a dynamic laser light scattering method, we have shown that 1–2 mM PPi enhances the increase in values induced by Ca2+ in isolated thick myofilaments fromLimulus striated muscle. However, similar treatment has no effect on the values of filaments suspended in either relaxing solution or ATP-free solution. is the average linewidth of the photoelectron count autocorrelation function of the light scattered. PPi had no effect on the increase of values by Sr2+ but it substantially increased the values of the thick myofilaments suspended in Ba2+-substituted Ca2+ activating solution. The results show that PPi also affects the energy-requiring cyclic cross-bridge motions.  相似文献

Waste water purification is characterized by intensive mineralization and nitrification processes. Because of the high O2 demand, temporarily anaerobic conditions may be produced, and denitrification by nitrifying organisms as well as heterotropic denitrification may contribute to N2O release. In situ measurements (1993–1994) suggest that N2O is released from activated sludge in a domestic waste water treatment plant at an average rate of 1040 g m–2h–1 with a range between zero and 6198 g m–2h–1. The production of N2O seems to be related to the concentration of NO 2 and NO 3 as well as to the pH. In the waste water about 75–200 g N2O l–1 is dissolved. This N2O is released after discharge into the receiving waters. The N2O is produced essentially by nitrification rather than by heterotropic denitrification. On a long-term scale the increasing use of mechanical-biological waste water purification plants world-wide may add increasingly to the anthropogenic production of N2O, although the present amount of N2O produced is negligible compared to its global terrestrial production.  相似文献

Summary The reaction of excess nitrosyl chloride with vanadium tetrachloride under specific conditions furnishes the previously unreported compound VCl4, 2NOCl. The formation of a 12 adduct shows that vanadium (IV) can behave as a typical tetravalent metal in the MIVCl4-NOCl system. Spectroscopic measurements reveal both nitrosonium and hexachlorometallate ions in the addition compound and indicate structural analogy with (NO)2-TiCl6. The complex may thus be designated as dinitrosonium hexachlorovanadate, (NO)2VCl6. However, this compound is probably not purely ionic, since the relatively low frequency observed for the IR-absorption of the NO+ ion (2146 cm–1 compared with the normal range 2150 to 2400 cm–1) and the appreciable volatility of the compound suggest a significant degree of covalent bonding.

Enterocytes isolated from rat jejunum were tested for the existence of a Cl/HCO 3 exchange, previously evidenced in basolateral membrane vesicles but not in brush border. Cells were found to retain functional integrity and transport capabilities long enough to allow Cl fluxes to be measured. Both efflux and uptake experiments indicate that a Cl/HCO 3 antiport, inhibited by 4,4-diisothiocyanostilbene-2-2-disulfonic acid (DIDS), is functional under resting conditions.  相似文献

Summary Zn2+ (10–100 M) elevated the frequency of miniature end-plate potentials (MEPPs) in the mouse diaphragm. The effect did not depend on external Ca2+. Botulinum type A toxin (BTXA, 50 ng/ml) abolished MEPPs almost completely within 30 min. Zn2+ (100 M) restored MEPPs and increased their frequency after they had been abolished by BTXA in Ca2+-free solutions. The antagonistic effect of Zn2+ in the Ca2+-free solution was reduced by exposing the diaphragm to the toxin in the Ca2+-free solutions containing high K+. Thus, the action of BTXA is probably enhanced by depolarization of the motor nerve terminals.  相似文献

Summary Phospholipase digestion of rat intestinal epithelial cell membranes was performed in order to study the influence of membrane phospholipids on the binding activity of VIP receptors. Phospholipases A2 and C strougly (ED504×10–2 and 4×10–1 g/ml, respectively) and rapidly reduced125I-VIP binding to membranes whereas phospholipase D was ineffective. This suggests an important role of both hydrophobic and hydrophilic groups of phospholipids on VIP receptor binding activity.This work was supported by INSERM (CRL 827017) and the Fondation pour la Recherche Médicale Française.  相似文献

Changes in cytosolic Ca2+ play an important role in a wide array of cell types and the control of its concentration depends upon the interplay of many cellular constituents. Resting cells maintain cytosolic calcium ([Ca2+]i) at a low level in the face of steep gradients of extracellular and sequestered Ca2+. Many different signals can provoke the opening of calcium channels in the plasma membrane or in intracellular compartments and cause rapid influx of Ca2+ into the cytosol and elevation of [Ca2+]i. After such stimulation Ca2+ ATPases located in the plasma membrane and in the membranes of intracellular stores rapidly return [Ca2+]i to its basal level. Such responses to elevation of [Ca2+]i are a part of an important signal transduction mechanism that uses calcium (often via the binding protein calmodulin) to mediate a variety of cellular actions responsive to outside influences.  相似文献

Summary A method of estimating the photosynthetic rate of soybean leaves using an oxygen electrode is presented. The procedure is rapid, requires small samples and compares favourably with estimates by other techniques. Light saturation occurs at 1200 E·m–2·sec–1. In intact tissue, [d-Ala2,MePhe4, Gly-ol5] enkephalin (10–5 M;-ligand), diminsihed short-circuit current (Isc) and increased water, Na+ and Cl net fluxes in vitro under open circuit conditions; it also inhibitedL-valine absorption andL-valine-dependent variations of short-circuit current (Isc, val). Naloxone (10–6 M) antagonized these effects. In the absence of the muscularis and myenteric plexus this enkephalin or morphine (-ligand) reduced Isc and Isc, val. These enkephalin effects occurred at different times. Different concentrations of enkephalin were tested for their effects on Isc, val. [d-Ala2,d-Leu5] enkephalin (mainly a -ligand) significantly decreased Isc but not Isc, val. The reduction ofL-valine absorption does not depend on the effects on basal ion transport. Interaction of opioids with-receptors located in the submucosal plexus and/or in the epithelial cell accounts for this reduction. This enkephalin effect seems to be at least partially under the control of the myenteric plexus.  相似文献

Summary The possibility that intracellular Ca2+, which mediates neurotransmitter release, regulation of membrane permeability, microtubule polymerization and axonal transport, is influenced by gonadal steroids via a Na–Ca exchange mechanism was examined. The resting Ca2+ uptake into synaptosomes was measured using crude synaptosomal pellets (P2 fraction), isolated from the brain stem, mesencephalic reticular formation (MRF), nucleus caudatus (NC) and the hippocampus of intact, long-term ovariectomized (OVX) and OVX plus progesterone (P) or estradiol-17 benzoate (EB) treated adult female rats. Irrespective of the brain structure investigated, the uptake was 1) markedly increased in synaptosomes from OVX animals in comparison to intact controls, and 2) reduced to near control values in synaptosomes from OVX rats treated s.c. with a single dose of 2 mg P or 5 g EB. Since Ca2+ influx into synaptosomes was shown earlier to depend on external sodium concentration, which was the same in all experiments described in this work, the results obtained indicate that ovarian steroids modulate basal synaptic activity in the rat brain by suppressing Na-dependent Ca2+ efflux from the nerve cell.  相似文献

Summary The contraction induced by a Ca2+-independent myosin light chain kinase (MLCK-) was characterized in terms of isometric force (Fo), immediate elastic recoil (SE), unloaded shortening velocity (Vus), shortening under a constant load and ATPase activity of chemically skinned smooth muscle preparations. These parameters were compared to those measured in a Ca2+-induced contraction to assess the nature of cross bridge interaction in the MLCK-induced contraction. Fo developed in chicken gizzard fibers as well as SE were similar in contractions elicited by either agent. Vus in the contraction induced by MLCK-(0.36 mg/ml) was similar though averaged 39.3±8.9% less than Vus induced by Ca2+ (1.6x10–6M) in the control fibers. Addition of Ca2+ (1.6x10–6M) to a contraction induced by MLCK-resulted in small increases in both Fo and Vus. Shortening under a constant load was similar for both types of contractions. The contraction induced by MLCK-was accompanied by an increased rate of ATP hydrolysis. The MLCK-induced contraction is thus kinetically similar though not identical to a contraction induced by Ca2+. We conclude that with respect to actin-myosin interaction, MLCK- and Ca2+-induced contractions are similar.  相似文献

Summary Using metal-ion buffers it was possible to remove Zn2+, Mg2+ and Mn2+ ions of pig kidney alkaline phosphatase reversibly. The dissociation constants obtained are KEMg:4·10–7 M, KEMn:4·10–8 M and KEZn:8·10–13 M (22°C, pH:9.6, :0.07).Acknowledgement: The authors thank Dr.H. U. Summary The O2– and Ca2+-paradoxes have a number of features in common and it is suggested that release of cytosolic proteins in both paradoxes is initiated by the activation of a sarcolemma NAD(P)H dehydrogenase which can generate a transmembrane flow of H+ and e and also oxygen radicals or recox cycling which damage ion channels and membrane proteins (phase I). Entry of Ca2+ through the damaged ion channels then exacerbates the damage by further activating this system, either directly or indirectly, and the redox cycling and/or oxygen radicals cause further damage to integral and cytoskeletal proteins of the sarcolemma resulting in microdamage to the integrity of the membrane (phase II) and the consequent release or exocytosis of cytoplasmic proteins and, under specialised condition, the blebbing of the sarcolemma. The system may be primed either by removal of extracellular Ca2+ or by raising [Ca2+]i by a variety of measures, these two actions being synergistic. The system is initially activated in the Ca2+-paradox by the membrane perturbation associated with removal of extracellular Ca2+; prolonged anoxia in the metabolically active cardiac muscle causes a depletion of the ATP supply, particularly in the absence of glucose, and hence a rise in [Ca2+]i in phase I of the oxygen paradox with the consequent activation of the NAD(P)H oxidase at the sarcolemma. Oxygen radicals are probably generated in both paradoxes and may have a partial role in the genesis of damage, but are not essential in the Ca2+-paradox which continues under anoxia. Massive entry of Ca2+ also activates an intracellularly localised dehydrogenase (probably at the SR) which produces myofilament damage by redox cycling.  相似文献

Summary Streptomyces species 3M grew in peptone yeast extract medium with 1000 g/ml K2Cr2O7. Incubation of the chromate with different cell fractions in the presence of NADH and NADPH resulted in a decrease of Cr6+ in the reaction mixture. The level of Cr6+ was reduced by 82.7% by a particulate cell fraction obtained by centrifugation at 105,000×g for 1 h, in the presence of NADH. The reducing enzyme was associated with this cell fraction. The enzyme was constitutive and reduced Cr6+ to Cr3+.  相似文献

Summary Hexoprenaline potentiated the14C-aminopyrine uptake (a reliable index of H+ generation) of isolated rat gastric cells stimulated by 10–6–10–4 mol/l carbachol, and inhibited that in response to 10–4 mol/l histamine without and in the presence of propranolol. It is concluded that hexoprenaline acts as a partial agonist on parietal cell H2-receptors and that -adrenoceptor activation may functionally modualte gastric acid secretion.Acknowledgments. S. Bioenergetic and metabolic status have been correlated with tissue oxygenation in murine fibrosarcomas (FSaII) of varying sizes (44–600 mm3). Ratios of -nucleoside triphosphates to inorganic phosphate (NTP/Pi) and phosphocreatine to inorganic phosphate (PCr/Pi) ratios derived from31P nuclear magnetic resonance spectroscopy (NMR) were positively correlated to median tissue O2 tension (pO2) values using O2-sensitive needle electrodes. pH declined during growth with intracellular acidosis being evident in tumors >350 mm3. Whereas lactic acid formation greatly contributed to this decline in small and medium-sized tumors, adenosine triphosphate (ATP) hydrolysis and slowing down of the activities of pumps involved in cellular pH regulation seem to be major factors responsible for intracellular acidification in bulky tumors. PCr levels decreased at an early growth stage, whilst ATP concentrations dropped in bulky malignancies only, coinciding with a decrease in adenylate energy charge and a substantial rise in the levels of total Pi On average, median pO2 values of ca. 10 mmHg represent a critical threshold for energy metabolism. At higher median O2 tensions, levels of ATP, phosphomonoester (PME) and total Pi were relatively constant. This coincided with intracellular alkalosis or neutrality and stable adenylate ratios. On average, median pO2 values <10 mmHg coincided with intracellular acidosis, ATP depletion, a drop in energy charge and rising Pi levels.  相似文献
