Biosynthesis of ascorbic acid was found in the kidneys (mesonephros and metanephros) of the chick embryo as well as in the yolk sac membrane. The activity of L-gulonolactone oxidase in the yolk sac membrane suggested that it was the major source of ascorbic acid in the chick embryo.  相似文献

In situ precipitation of calcium (Ca2+) with fluoride and antimonate shows that Ca2+-specific precipitate is localized almost exclusively within lipid droplets of neuroepithelial cells during neural tube formation in chick and mouse embryos. The density of Ca2+ precipitate within lipid droplets is generally greater in the apical ends of cells situated in regions of the neuroepithelium that are actively engaged in bending. These findings suggest that lipid droplets, in addition to providing a source of metabolic fuel for developing neuroepithelial cells, also serve as Ca2+-storage and-releasing sites during neurulation.This study was supported by grants from the NIH (NS23200), the BRSG fund of UMDNJ, and the Busch Fund of Rutgers University. Summary The presence of transforming growth factor activity in early chick embryos was directly demonstrated by the ability of limb and tail buds to induce anchorage independent division in NRK 49f cells. Colony number increased with limb bud number and developmental stage. Medium conditioned by tail buds contained some stimulating effect, and strongly promoted the action of other transforming growth factors.  相似文献

The effect of carbachol on twitch tension of atrial preparations from chick embryos of different incubation ages (3–14 days) was studied. At every age carbachol evoked negative (at low concentrations) and positive (at higher concentrations) inotropic responses. Maximal response values for both effects increased with age; in 3- and 5-day atria the positive inotropic response prevailed. The muscarinic antagonist pirenzepine inhibited the positive (on 5-day atria) and negative (on strips of 14-day atria) inotropic effects of carbachol with pA2 values of 6.8 and 8.0, respectively, suggesting that muscarinic receptors mediating these effects belong to different receptor subtypes.  相似文献

A population of ventral neural tube cells has recently been shown to migrate out of the hind brain neural tube via the vagus nerve and contribute to the developing gastrointestinal tract. Since liver is also innervated by the vagus nerve, we sought to determine if these cells also migrate into the liver. Ventral neural tube cells in the caudal hindbrain of chick embryos were tagged with a replication-deficient retroviral vector containing the LacZ gene on embryonic day 2. Embryos were processed for detection of labeled cells on embryonic day 5 and 11. Labeled cells were seen in the liver on both days and identified as hepatocytes. Previously, it was believed that all hepatocytes develop from the gut endoderm. Results of the present study show an additional source for the formation of liver cells. Summary Ascorbic acid is utilized during the post-embryonic differentiation of skeletal muscle fibres in chick. While the fibres lose their heterogeneity with regard to ascorbic acid, they continue to exhibit differences in their metabolic rates in terms of the succinate dehydrogenase activity throughout life.The investigations were carried out at the Department of Zoology, Panjab University, Chandigarh. 烯二炔类抗生素以其独特的化学结构和强烈的抗肿瘤活性成为化学、生物和医药领域重点关注的对象。本文简要介绍了近年来关于烯二炔化合物的生物合成研究进展,以及运用组合生物合成的原理创造新型烯二炔结构类似物的前景。烯二炔类抗生素为我们研究复杂天然产物的生物合成提供了一个很好的模型,其基因和生化水平机制的阐明将进一步丰富组合生物合成的内容和手段,最终有利于发现和发展新型的抗肿瘤药物。  相似文献

Amphibians and reptiles evolved with the capacity to synthesize ascorbic acid. Some higher vertebrates, like bats, guinea pigs, primates, and humans have lost the microsomal enzyme gulonolactone oxidase, and in cases of ascorbic acid deficiency suffer from symptoms of scurvy. The question of whether the capacity to synthesize ascorbate is also present in lower vertebrates could throw light on the evolution of this pathway. In order to find out whether ascorbic acid synthesis took place in two primitive Actinopterigian fish, the paddlefish (Polydon spathula) and the white sturgeon (Acipenser transmontanus) were fed with a scorbutogenic diet or diet(s) supplemented with a graded level of ascorbic acid. We found no growth depression nor external symptoms of scurvy, which would be pronounced in modern bony fishes (Teleostei) under similar conditions. The tissue level of ascorbate in both these primitive species indicated that vitamin C in intestine and liver is not depleted when fed a scorbutogenic diet. Gulonolactone oxidase activity was found in the kineys of the Actinopterigian fishes. Thus, I question the accepted evolutionary pathway for ascorbic acid biosynthesis in lower vertebrates and suggest that the modern bony fishes,Teleostei, lost their ability to express the gulonolactone oxidase genes after they had separated during the Silurian from their common ancestor with the coelacanths (Latimeria) and Dipnoi.  相似文献

Summary Increase in nuclear size in liver has been used as an index of polyploidy. It has long been considered that the occurrence of polyploidy and multinuclearity are characteristics of mammalian liver. The present study shows the occurrence of these phenomena in the liver of birds, so these features are not confined to mammals. 3 classes of nuclear size groups have been identified. The simultaneous occurrence of polyploidy and binuclearity indicates some sort of interrelationship between them.  相似文献

Accumulation of cadmium in organs of guinea pigs after subchronic oral cadmium treatment (1 mg Cd/animal/24 h) was in the following order: kidneys > liver > heart > testes > brain. The preventive effects of high doses of ascorbic acid (AA) against cadmium deposition were more pronounced in the testes, heart and brain, and in the kidney only after short-term cadmium treatment. Ascorbic acid had no protective effect on cadmium accumulation in the liver.  相似文献

Summary A mutant mouse strain ddY/DAO lacks D-amino acid oxidase activity and accumulates free neutral D-amino acids in its tissues. In this study, D-aspartate oxidase activity and D-aspartate content in the tissues of these mutant mice were compared with those of normal mice. No significant difference was observed, indicating that the metabolism of acidic D-amino acids was unaffected in the mutant.  相似文献

The physiological role of D-amino acid oxidase (EC 1. 4. 3. 3) in mouse brain is described. The presence of D-enantiomers of neutral common amino acids was surveyed in the brain. D-serine was shown to be present at high concentration only in regions where the enzyme activity was low. In normal mice whose D-amino acid oxidase activity was much higher in the cerebellum than in the cerebrum, free D-serine content was apparently lower in the cerebellum than in the cerebrum. In mice of a mutant strain lacking D-amino acid-oxidase activity, the free D-serine level was remarkably high both in the cerebrum and cerebellum. The results suggest that the enzyme is involved in the elimination of free D-serine in the cerebellum.  相似文献

Summary Polyclonal antibodies were prepared from rabbit sera after immunization with holo- and apo-D-amino acid oxidase purified fromR. gracilis. Both anti-holo- and anti-apoenzyme IgG fractions (as well as affinity-purified IgG) were highly specific: in blot-transfer analyses after SDS-PAGE only a 39 kDa band, corresponding to enzyme monomer, was recognized even in the partially purified yeast extract. No cross-reaction was detected with pig kidney D-amino acid oxidase. As a difference from the mammalian enzyme, yeast D-amino acid oxidase anti-holo- and anti-apoenzyme IgGs had different properties in inactivation and precipitation experiments, indicating the existence of different antigenicity sites related to the FAD-binding domain in the enzyme.  相似文献

The patterns of Glut1 and Glut3 glucose transporter protein and mRNA expression were assessed during embryogenesis of chicken brain and skeletal muscle, Glut4 protein levels were also evaluated in skeletal muscle and heart, and Glut1 was examined in the developing heart and liver. Glut1 protein expression was detectable throughout brain ontogeny but was highest during early development. Glut1 mRNA levels in the brain remained very high throughout development. Glut3 protein was highest very early and very late and mRNA was highest during the last half of development. In embryonic skeletal muscle, the levels of Glut1and Glut3 proteins and mRNA were highest very early, and declined severely by mid-development. Glut1 protein and mRNA in the heart also peaked early and then decreased steadily. Although Glut1 mRNA levels were consistently high in the embryonic liver, Glut1 protein expression was not detected. These results suggest that (1) Glut1 is developmentally regulated in chick brain, skeletal muscle, and heart, (2) Glut1 mRNA is present in liver but does not appear to be translated, (3) Glut3 in brain increases developmentally but is virtually absent in muscle, and (4) Glut4 protein and mRNA appear to be absent from chick heart and skeletal muscle.
