It has been shown previously that deoxyguanosine residues in DNA are hydroxylated at the C-8 position both in vitro and in vivo to produce 8-hydroxydeoxyguanosine (8-OH-dG) by various agents that produce oxygen radicals such as reducing reagents-O2, metal ions-O2, polyphenol-H2O2-Fe3+, asbestos-H2O2 or ionizing radiation. These agents are mostly either mutagenic or carcinogenic; therefore, the formation of 8-OH-dG can also be considered a likely cause of mutation or carcinogenesis by oxygen radicals. It is of interest to know whether the 8-OH-dG residue in DNA is misread during DNA replication. To answer this question, we have examined the effect of the 8-OH-dG residue in DNA on the fidelity of DNA replication using a DNA synthesis system in vitro with Escherichia coli DNA polymerase I (Klenow fragment). The synthetic oligodeoxynucleotides, with or without an 8-OH-dG residue in a specified position, were chemically synthesized and used as templates for DNA synthesis under the conditions of the dideoxy chain termination sequencing method. Surprisingly, in addition to misreading of the 8-OH-dG residue itself, pyrimidines next to the 8-OH-dG residue (G has not yet been tested) were also misread.  相似文献

The boundary between the Earth's metallic core and its silicate mantle is characterized by strong lateral heterogeneity and sharp changes in density, seismic wave velocities, electrical conductivity and chemical composition. To investigate the composition and properties of the lowermost mantle, an understanding of the chemical reactions that take place between liquid iron and the complex Mg-Fe-Si-Al-oxides of the Earth's lower mantle is first required. Here we present a study of the interaction between iron and silica (SiO2) in electrically and laser-heated diamond anvil cells. In a multianvil apparatus at pressures up to 140 GPa and temperatures over 3,800 K we simulate conditions down to the core-mantle boundary. At high temperature and pressures below 40 GPa, iron and silica react to form iron oxide and an iron-silicon alloy, with up to 5 wt% silicon. At pressures of 85-140 GPa, however, iron and SiO2 do not react and iron-silicon alloys dissociate into almost pure iron and a CsCl-structured (B2) FeSi compound. Our experiments suggest that a metallic silicon-rich B2 phase, produced at the core-mantle boundary (owing to reactions between iron and silicate), could accumulate at the boundary between the mantle and core and explain the anomalously high electrical conductivity of this region.  相似文献

Seismological observations have revealed the existence of low-velocity and high-attenuation zones above the discontinuity at 410 km depth, at the base of the Earth's upper mantle. It has been suggested that a small amount of melt could be responsible for such anomalies. The density of silicate melt under dry conditions has been measured at high pressure and found to be denser than the surrounding solid, thereby allowing the melt to remain at depth. But no experimental investigation of the density of hydrous melt has yet been carried out. Here we present data constraining the density of hydrous basaltic melt under pressure to examine the stability of melt above the 410-km discontinuity. We infer that hydrous magma formed by partial melting above the 410-km discontinuity may indeed be gravitationally stable, thereby supporting the idea that low-velocity or high-attentuation regions just above the mantle transition zone may result from the presence of melt.  相似文献

The distribution of geochemical species in the Earth's interior is largely controlled by fractional melting and crystallization processes that are intimately linked to the thermal state and evolution of the mantle. The existence of patches of dense partial melt at the base of the Earth's mantle, together with estimates of melting temperatures for deep mantle phases and the amount of cooling of the underlying core required to maintain a geodynamo throughout much of the Earth's history, suggest that more extensive deep melting occurred in the past. Here we show that a stable layer of dense melt formed at the base of the mantle early in the Earth's history would have undergone slow fractional crystallization, and would be an ideal candidate for an unsampled geochemical reservoir hosting a variety of incompatible species (most notably the missing budget of heat-producing elements) for an initial basal magma ocean thickness of about 1,000 km. Differences in 142Nd/144Nd ratios between chondrites and terrestrial rocks can be explained by fractional crystallization with a decay timescale of the order of 1 Gyr. These combined constraints yield thermal evolution models in which radiogenic heat production and latent heat exchange prevent early cooling of the core and possibly delay the onset of the geodynamo to 3.4-4 Gyr ago.  相似文献

Ribbon-type synapses in inner hair cells of the mammalian cochlea encode the complexity of auditory signals by fast and tonic release through fusion of neurotransmitter-containing vesicles. At any instant, only about 100 vesicles are tethered to the synaptic ribbon, and about 14 of these are docked to the plasma membrane, constituting the readily releasable pool. Although this pool contains about the same number of vesicles as that of conventional synapses, ribbon release sites operate at rates of about two orders of magnitude higher and with submillisecond precision. How these sites replenish their vesicles so efficiently remains unclear. We show here, using two-photon imaging of single release sites in the intact cochlea, that preformed vesicles derived from cytoplasmic vesicle-generating compartments participate in fast release and replenishment. Vesicles were released at a maximal initial rate of 3 per millisecond during a depolarizing pulse, and were replenished at a rate of 1.9 per millisecond. We propose that such rapid resupply of vesicles enables temporally precise and sustained release rates. This may explain how the first auditory synapse can encode with indefatigable precision without having to rely on the slow, local endocytic vesicle cycle.  相似文献
