Bioactivity-directed fractionation of a root extract ofDuroia hirsuta (Rubiaceae), a toxic and potentially allelopathic understory tree from the western Amazon, has led to the isolation of the tetracyclic iridoid lactone, plumericin (1). Bioassays showing plumericin strongly inhibited lettuce radicle elongation at a concentration (IC50) of 35.8 m/ml (123 M). The isolation of a highly potent inhibitor of plant growth fromDuroia hirsuta supports the hypothesis that the lack of vegetation surrounding this tree is the result of allelopathy.  相似文献

How a conformationally disordered polypeptide chain rapidly and efficiently achieves its well-defined native structure is still a major question in modern structural biology. Although much progress has been made towards rationalizing the principles of protein structure and dynamics, the mechanism of the folding process and the determinants of the final fold are not yet known in any detail. One protein for which folding has been studied in great detail by a combination of diverse techniques is hen lysozyme. In this article we review the present state of our knowledge of the folding process of this enzyme and focus in particular on recent experiments to probe some of its specific features. These results are then discussed in the context of the ‘new view’ of protein folding based on energy surfaces and land scapes. It is shown that a schematic energy surface for lysozyme folding, which is broadly consistent with our experimental data, begins to provide a unified model for protein folding through which experimental and theoretical ideas can be brought together.  相似文献

In The Structure of Scientific Revolutions, Kuhn famously advanced the claim that scientists work in a different world after a scientific revolution. Kuhn's view has been at the center of a philosophical literature that has tried to make sense of his bold claim, by listing Kuhn's view in good company with other seemingly constructivist proposals. The purpose of this paper is to take some steps towards clarifying what sort of constructivism (if any) is in fact at stake in Kuhn's view. To this end, I distinguish between two main (albeit not exclusive) notions of mind-dependence: a semantic notion and an ontological one. I point out that Kuhn's view should be understood as subscribing to a form of semantic mind-dependence, and conclude that semantic mind-dependence does not land us into any worrisome ontological mind-dependence, pace any constructivist reading of Kuhn.  相似文献

Zusammenfassung Eine genetisch bedingte Bewegungsstörung in der Hausmaus «reeler» wurde untersucht. Der Erbgang der «reeler»-Mutation ist einfach rezessiv. In homozygotem Zustand führt das Gen zu Gleichgewichtsstörungen und leichtem Zittern.Das Studium der Morphologie des Gehirns der «reeler»-Mäuse ergab, dass die Cytoarchitektonik des Kleinhirns in dieser Mutante verändert und teilweise zerstört ist.

It is well documented that activation of calpain, a calcium-sensitive cysteine protease, marks the pathology of naturally and experimentally occuring neurodegenerative conditions. Calpain-mediated proteolysis of major membrane-skeletal protein, αII-spectrin, results in the appearance of two unique and highly stable breakdown products, which is an early event in neural cell pathology. This review focuses on spectrin degradation by calpain within neurons induced by diverse conditions, emphasizing a current picture of multi-pattern neuronal death and a recent success in the development of spectrin-based biomarkers. In this article, we analyze the third of three papers, in which Einstein presented his quantum theory of the ideal gas of 1924–1925. Although it failed to attract the attention of Einstein’s contemporaries and although also today very few commentators refer to it, we argue for its significance in the context of Einstein’s quantum researches. It contains an attempt to extend and exhaust the characterization of the monatomic ideal gas without appealing to combinatorics. Its ambiguities illustrate Einstein’s confusion with his initial success in extending Bose’s results and in realizing the consequences of what later came to be called Bose–Einstein statistics. We discuss Einstein’s motivation for writing a non-combinatorial paper, partly in response to criticism by his friend Ehrenfest, and we paraphrase its content. Its arguments are based on Einstein’s belief in the complete analogy between the thermodynamics of light quanta and of material particles and invoke considerations of adiabatic transformations as well as of dimensional analysis. These techniques were well known to Einstein from earlier work on Wien’s displacement law, Planck’s radiation theory and the specific heat of solids. We also investigate the possible role of Ehrenfest in the gestation of the theory.  相似文献

Summary A method for detecting signal in the presence of noise in a highly specific was is described. Using action potential interval data from 12 neurons in rat cerebellum, we have demonstrated that the sequential ordering of spike intervals contains both noise and signal. We have identified and quantified the magnitude of relative entropy (uncertainty) for specified sets of interval patterns, ranging in length from 3–5 successive intervals. Some of these sets have exceptionally low entropy and thus seem to be especially meaningful as a set (word) to the brain.  相似文献

Summary Electron microscopy demonstrated the presence of glycocalyx on the cell surface ofChattonella antiqua, which was previously believed to be naked. Histochemical analysis of this structure indicates that the glycocalyx consists of at least two different types of acidic complex carbohydrates, sulfated and nonsulfated, together with a neural carbohydrate-protein complex with 1,2-glycol groups and -D-mannosyl and -D-glucosyl residues.  相似文献

ATP-dependent potassium (KATP) channels occupy a key position in the control of insulin release from the pancreatic β cell since they couple cell polarity to metabolism. These channels close when more ATP is produced via glucose metabolism. They are also controlled by sulfonylureas, a class of drugs used in type 2 diabetic patients for triggering insulin secretion from β cells that have lost part of their sensitivity to glucose. We have demonstrated the existence of endogenous counterparts to sulfonylureas which we have called ‘endosulfines.’ In this review, we describe the discovery, isolation, cloning, and biological features of the high-molecular-mass form, α-endosulfine, and discuss its possible role in the physiology of the β cell as well as in pathology. In this paper I direct attention to one feature of Hacking’s recent work on styles of reasoning and argue that this feature is of far greater philosophical significance than Hacking’s limited discussion of this suggests. The feature in question is his use of ‘legendary beginnings’ in setting out a given style, viz. the method of introducing a style of reasoning by recounting a popular and quasi-mythical narrative that ties the crystallisation of that style to a particular person in a particular place and at a particular time. Whilst Hacking both deploys and discusses this method, his comments suggest that this is primarily a stylistic device employed for reasons of expedience. In contrast, it is argued here that recounting the legendary origins of a style of reasoning affords a distinctive way of vindicating that style, a vindication from within the style itself.  相似文献

This study is the foundation of a new interpretation of the introduction and the three first books of Diophantus’s Arithmetica, one that opens the way to a historically correct contextualization of the work. Its purpose, as indicated in the title, is to renew the traditional discussion on the methods of problem-solving used by Diophantus, through the detailed exposition of a new analytical framework that aims to give an account of the coherence and progressive nature of the material included in the three first books of the Arithmetica. One outcome of this new ‘toolbox’ is a new conspectus of the problems and solutions contained in the latter, which is presented in appendix. The first part of the article clarifies, as a necessary preliminary, the key notions and terminology underlying our analysis. Among these new concepts is the notion of “method of invention,” which accounts in general for any way, by which “positions” (hypostaseis) are used in the Arithmetica. The next part proposes a complete inventory of the various methods of invention found in the three first books. Finally the last part presents the above mentioned conspectus and proposes a series of preliminary conclusions that can be drawn from it.  相似文献

This paper provides a detailed account of the period of the complex history of British algebra and geometry between the publication of George Peacock's Treatise on Algebra in 1830 and William Rowan Hamilton's paper on quaternions of 1843. During these years, Duncan Farquharson Gregory and William Walton published several contributions on ‘algebraical geometry’ and ‘geometrical algebra’ in the Cambridge Mathematical Journal. These contributions enabled them not only to generalize Peacock's symbolical algebra on the basis of geometrical considerations, but also to initiate the attempts to question the status of Euclidean space as the arbiter of valid geometrical interpretations. At the same time, Gregory and Walton were bound by the limits of symbolical algebra that they themselves made explicit; their work was not and could not be the ‘abstract algebra’ and ‘abstract geometry’ of figures such as Hamilton and Cayley. The central argument of the paper is that an understanding of the contributions to ‘algebraical geometry’ and ‘geometrical algebra’ of the second generation of ‘scientific’ symbolical algebraists is essential for a satisfactory explanation of the radical transition from symbolical to abstract algebra that took place in British mathematics in the 1830s–1840s.  相似文献

This paper argues that essential features of Feyerabend's philosophy, namely his radicalization of critical rationalism and his turn to relativism, could be understood better in the light of his engagement with early Greek thought. In contrast to his earlier, Popperian views he came to see the Homeric worldview as a genuine alternative, which was not falsified by the Presocratics. Unlike socio–psychological and externalist accounts my reading of his published and unpublished material suggests that his alternative reconstruction of the ancient beginnings of the Western scientific tradition motivate and justify his moderate Protagorean relativism.  相似文献
