Summary In chromosome preparations from swine lymphocyte cultures, quadriradial figures and homolog associations were seen only of chromosome 10 and at a low frequency. The frequency of quadriradial figures was substantially increased by mitomycin C treatment, whereby 26.2% of the quadriradials were formed from the same chromosome pair with the number 10.  相似文献

The influence of inhibition or stimulation of cellular DNA synthesis on tick-borne virus antigen production in persistently infected cell culture was studied. Either mitomycin C or cytosine-arabinoside caused cessation of antigen-containing cell number increase. Stimulation of cellular DNA synthesis by growth medium change increased the level of antigen-containing cells. When HEp-2-Sof culture was synchronized, a correlation was observed between the entrance of cells into DNA synthesis phase and the increase of proportion of antigen-containing cells.  相似文献

Summary Cells fromXenopus embryos blocked at the blastula stage by treatment with hydroxyurea have been isolated and cultured in vitro. The morphology of these cells has been compared with that of cells from normal embryos using scanning electron microscopy. Cells from such hydroxyurea-blocked embryos do not show the features, or changes in features, in culture shown by cells from normal embryos.Acknowledgments. We wish to thank Mr C. Summary Attention has previously been drawn to a specific effect of NHCP on embryonicPleurodeles cell differentiation. With a modified NHCP labelling technique, autoradiography has revealed a cytoplasmic concentration of labelled NHCP and has not revealed any difference between homospecific and heterospecific NHCP penetration.This work has been supported financially by research grants from the C.N.R.S. Heated albumin microspheres with an average diameter of 45 +/- 8 microns and containing mitomycin C, released, in vitro, about 20% of this antibiotic over a 3-day period. VX-2 tumors were implanted into the hind leg of rabbits and the drug-containing microspheres were injected into the femoral artery of these animals. High levels of the drug were maintained for several hours in the tumor and growth of the tumor was inhibited considerably, compared to findings in control rabbits given the conventional mitomycin C. Half the number of the rabbits treated with our new method are alive with no evidence of tumor.  相似文献

Summary Pea embryos grown in the presence of 2.5 mM hydroxyurea from the beginning of imbibition showed a mitotic peak the height of which was considerably less than that of the control, material. Soon after the mitotic peak, there was a complete cessation of mitosis in the treated material. The extent of suppression of mitosis during the first post-dormancy cell cycle caused by hydroxyurea has been used to obtain an estimate of the relative proportion of 2C and 4C cells in the embryo.  相似文献

CBA Mice were immunized by two intraperitoneal injections of 30 X 10(6) DBA/2 or C57BL/6 spleen cells at days--12 and--2. Peritoneal cell population was obtained at day zero by washing the peritoneal cavity of Mice. Adherent cells were then separated using a 2 hrs. incubation in "Falcon" plates followed by washing. This macrophage-rich peritoneal cell population was found nonspecifically cytotoxic against 51Cr labeled tumoral target cells: P815 X DBA/2 mastocytoma cells, EL4 X C57BL/L lymphoma cells and spontaneous lymphoma AKR cells (same H--2k as CBA). This adherent peritoneal cell cytoxicity was demonstrated after 24 hrs. incubation with the target cells. It was found in nonspecific combination as well as when using target cells syngeneic to the donor. These findings suggest that adherent peritoneal cell cytotoxicity could be at least partly due to macrophages and result from factor (s) released by sensitized lymphocytes in vivo in the same way as has been previously demonstrated in vitro.  相似文献

Summary The activities of catalase (E.C.1.11.1.6) and glutathione peroxidase (E.C.1.11.1.9) were compared in red blood cells from humans, ducks and normal and acatalasemic mice. In the cells from both strains of mice, an equally high activity of GSH-Px was found which could be inhibited completely by iodoacetate but was not sensitive to N-ethylmaleimide.Acknowledgment. Summary An extensive cytoplasmic fibrillar system has been observed in fibroblast-like cells of regenerating tendon. It consists of bundles of actin filaments, which often show a cross-striated appearance due to electron dense bodies occurring throughout their length. The functional role of this contractile apparatus seems to be related to the process of movement and orientation of the newly formed cells and to the retraction of the regenerating tendon.Work was supported by grant C. Summary This paper deals with the fine structure of the fibrillar centres of the nucleolus inAllium cepa cells in ultrathin, sections of in vivo fixed roots. The ultrastructural observations have allowed us to consider each nucleolar fibrillar centre as an active zone in the nucleolar chromatin loop, and to propose a possible model for the organization of the different components of the nucleolus within it.Acknowledgments. Summary Male mice were injected i.p. with 2.5 mg/kg mitomycin C, 100 mg/kg ethyl nitrosourea or saline and mated with untreated virgin females five weeks later. Sperm from 64 of the F1 male progeny were analyzed histochemically for acrosin, succinic dehydrogenase and alpha-glycerophosphate dehydrogenase activity. The frequency of F1 males with sub-normal sperm enzyme activity was significantly higher among progeny from treated males than in controls. Indole-3-carbinol (I3C) has been found to act against several types of cancer, while ultraviolet B (UVB) is known to induce the apoptosis of human melanoma cells. Here, we investigated whether I3C can sensitize G361 human melanoma cells to UVB-induced apoptosis. We examined the effects of combined I3C and UVB (I3C/UVB) at various dosages. I3C (200 μM)/UVB (50 mJ/cm2) synergistically reduced melanoma cell viability, whereas I3C (200 μM) or UVB (50 mJ/cm2), separately, had little effect on cell viability. DNA fragmentation assays indicated that I3C/UVB induced apoptosis. Further results show that I3C/UVB activates caspase-8, −3, and Bid and causes the cleavage of poly(ADP-ribose) polymerase. Moreover, I3C decreased the expression of the anti-apoptotic protein, Bcl-2, whereas UVB increased the translocation of Bax to mitochondria. Thus, an increased Bax/Bcl-2 ratio by I3C/UVB may result in melanoma apoptosis. In conclusion, our study demonstrated that I3C sensitizes human melanoma cells by down-regulating Bcl-2. Summary Neurosecretory substance has been identified in the rostral process of bipolar neurosecretory cells of the caudal neurosecretory system in 4 Indian catfishes, using acid violet stain.
