Influence of the thymus on the incidence of secondary and parabiotic disease

The secondary disease in P leads to F1 strain combination depends upon the immune status of both partners. It is most clearly expressed if both donors and recipients are immunologically crippled by thymectomy and irradiation. Conspicuous reduction in the immune potential of parental parabionts induced by thymectomy and irradiation displayed their marked incidence of parabiotic death.

A partially purified thymic extract has determined the development of leucosis in 10 thymectomized irradiated CC57Bl mice out of 25. The pure hormone isolated from the same extract prevented the development of leucosis in intact irradiated Mice. This seems to indicate the presence of a second active substance in the extract (enhancing the development of the leucosis).     

Influence of a mixture of chemical protectors on the lymphoid regeneration of bone marrow and thymus in irradiated mice

Summary In mouse, the administration of chemical protectors before an irradiation induces a more rapid bone marrow regeneration and an increased lymphoid rebound. In the thymus the late atrophy is reduced. Separately administrated, the protectors decrease greatly the thymocytes number but have no effect on the marrow population.

Zusammenfassung Die Radiogoldclearance thymektomierter erwachsener Mäuse ist gegenüber scheinthymektomierten bzw. den Kontrolltieren vermindert. Eine zusätzliche Immundepression durch Ganzkörperbestrahlung mit 350 R führt gegenüber den einzelnen Vergleichsgruppen zu keiner Änderung der Radiogoldclearance. Daraus kann mit aller Vorsicht geschlossen werden, dass der Thymus sowohl die Immunreaktionen als auch die Phagozytosefunktion als übergeordnetes Zentrum steuert. Jedes System scheint aber für sich eine funktionelle Einheit zu bilden.

Zusammenfassung Es wird gezeigt, dass an erwachsenen und neugeborenen Ratten die Jodaufnahme des Thymus bei den letzteren viel höher ist als die Jodaufnahme der Thyroidea. Dies unterstützt die These, wonach die Jodspeicherkapazität der Thyroidea das Resultat einer Differenzierung darstellt, indem auf früherer ontogenetischer Stufe für den ganzen Vorderdarm eine Jodspeicherkapazität besteht.     

Melatonin biosynthesis in the thymus of humans and rats

Melatonin is an indoleamine widely distributed in the evolution that shows a great functional versatility, playing an important role as a transmitter of photoperiodic information and exhibiting antioxidant, oncostatic, anti-aging and immunomodulatory properties. In vertebrates, this molecule is produced by the pineal gland and other extrapineal sites. The present study was carried out to investigate the presence of melatonin in thymus and the possibility of an endogenous melatonin synthesis in this organ, in which T cells are matured. In this work, we demonstrate in humans and rats that thymus contains melatonin, expresses the mRNAs encoding N-acetyltransferase and hydroxyindol-O-methyltransferase, the two key enzymes of the melatonin synthesis, and has this biosynthetic machinery activated. In addition, rat thymocytes cultured for 24 h exhibited high levels of melatonin. The results presented here suggest that human and rat thymuses are able to synthesize melatonin, which could have intracrine, autocrine and paracrine functions. The effects of parabiotic union with a normal partner on the blood tissue of the b/b rat suffering from an inheritable anemia

Summary The parabiotic union of a b/b rat, suffering from a red blood cell anemia, with a normal partner will restore to normal the functioning of this type of blood cell. The effect of parabiotic union is evident for several weeks following the separation of the parabiotic partners. It would be logical to conclude that the normal partner supplies a factor absent in the anemic animal needed for normal functioning of the red cell.     

The role of the thymus in antinuclear autoimmunization

Résumé La thymectomie chez des souris CF1 âgées de 1 mois est suivie comme dans le cas de la thymectomie néonatale de l'apparition d'anticorps antinucléaires. Le transfert adoptif de l'autoimmunité antinucléaire apparu chez des animaux thymectomisés à la naissance n'est possible que si les animaux receveurs sont eux-mêmes thymectomisés. Ces constatations mettent en évidence un rôle possible de contrôle des réactions autoimmunes par le thymus.

Evidence for the extranuclear localization of thymosins in thymus

A new radioimmunoassay has been developed for thymosin ₄ by generating rabbit polyclonal antibodies against the synthetic N-terminal peptide fragment 1–15 coupled to KLH. The synthetic analogue [Tyr¹²]-thymosin ₄ (1–15) was used as tracer. This radioimmunoassay, with a useful range of 10–1000 pmoles, showed cross-reactivity with the second homologous -thymosin of man and rat (thymosin ₁₀) but not of calf (thymosin ₉). This radioimmunoassay, together with an improved radioimmunoassay for the N-terminus of parathymosin , was employed for the measurement of the levels of thymosin ₄ and parathymosin in nuclear and extranuclear extracts of calf thymus. The bulk of these polypeptides was found in the extranuclear material whereas only traces were observed in the nuclear environment, which indicates the extranuclear localisation of - and -thymosins.

The granules of Langerhans cells in the thymus of the rat were subjected to X-ray microanalysis. It was established that gold is present in these morphological structures.
