抗体和抗原的相互作用面常常被描述成锁和钥匙的关系,说明一个抗体只能适合一种抗原。美国加州基因技术公司Germaine Fuh和同事分离出一种治疗用单克隆抗体Herceptin的异构体,其结合位点可以同时结合人表皮生长因子受体2（HER2）和血管内皮生长因子（VEGF）     

抗血管内皮生长因子药物治疗湿性年龄相关性黄斑变性的研究进展

湿性年龄相关性黄斑变性(ARMD)的主要病理改变是脉络膜新生血管膜(CNV)的形成,而血管内皮生长因子(VEGF)在脉络膜新生血管形成中起重要作用。目前,抗血管内皮生长因子(anti-VEGF)药物是治疗湿性ARMD的重要选择,主要有哌加他尼钠、贝伐单抗、雷珠单抗和阿柏西普等,目前临床上主要使用的是贝伐单抗和雷珠单抗。该文就临床上主要使用的anti-VEGF药物的疗效、安全性和经济性做一综述,为临床治疗提供依据。     

自身免疫性药物阿达木单抗在中国的发展现状

阿达木抗体是一种特异性靶向人肿瘤坏死因子TNF-α的全人源单克隆抗体类药物.近年来先后被美国和欧盟批准用于类风湿关节炎、克罗恩病和银屑病关节炎等8种自身免疫性疾病的治疗.自2010年阿达木单抗在中国上市,已获批类风湿关节炎和强制性脊柱炎2种适应症的应用.因阿达木单抗疗效好,副作用小,其在临床上受到越来越多的关注.本文就阿达木单抗在中国的发展现状和研究成果进行扼要论述,以期为后续阿达木单抗在中国更为有效和广泛的临床应用提供帮助.     

抗VEGF玻璃体腔内注射雷珠单抗围手术期的护理

目的:探讨抗VEGF玻璃体腔内注射雷珠单抗围手术期护理配合方法,积累工作经验,规范护理配合流程,提高护理配合水平.方法:对血管源性眼底病患者抗VEGF玻璃体腔内注射雷珠单抗256例手术/136例患者,在围手术期进行护理,分析总结护理措施实施情况和效果.结果:所有手术进行顺利,注射后除4例患者病情没有改善(与黄斑萎缩或瘢痕化有关)外,其余患者病情都有不同程度的改善.未发生眼内感染和出血.术后患者遵医行为和随访依从性的执行情况明显提高.结论:做好玻璃体腔内注射围手术期护理配合,有助于抗VEGF治疗的顺利进行,增强患者对所患疾病的正确认识,避免盲目治疗.提高患者日常预防保健和自我检测病情变化能力,医患关系也得到改善.     

单克隆抗体制剂的最新进展

杂交瘤技术建立以来,单抗研究走过了漫长而又曲折的历程,分子生物学的发展使单抗从诊断工具变为有效的治疗药物.近时期,单抗作为治疗制品有了迅速的发展,其在生物技术治疗药物的研究开发中占有重要地位.主要阐述了单克隆抗体人源化、核素耦联特异性抗肿瘤抗体、单克隆抗体导向药物、单克隆抗体生产技术、治疗用单抗制剂等5个方面的最新进展.     

重组人血管内皮生长因子(VEGF)单克隆抗体的制备

为制备人血管内皮生长因子(VEGF)单克隆抗体,制备并纯化了VEGF蛋白,通过免疫、细胞融合、筛选等方法获得了能够稳定分泌单克隆抗体的细胞株;采用小鼠腹腔接种法制备腹水,并用G蛋白(Protein G)亲和层析柱对抗体进行了纯化;采用间接ELISA及Western blot实验分别对纯化后的抗体进行了效价测定和特异性验证.结果表明:此方法可成功筛选出能稳定分泌VEGF单克隆抗体的杂交瘤细胞株,纯化后腹水抗体效价为1∶1 024;Western blot检验证实制备的单克隆抗体能特异性识别VEGF蛋白.     

罗氏公司血管内皮生长因子抗体技术专利分析

 检索了罗氏公司血管内皮生长因子(vascular endothelial growth factor,VEGF)抗体相关专利申请情况,分析了贝伐珠单抗和兰尼单抗的技术研发路线、罗氏公司VEGF抗体相关在华专利布局情况,对其未来技术研发趋势和方向进行了预测。     

表皮生长因子受体与胃肠道肿瘤

介绍表皮生长因子受体（EGFR）的生物学特征及其与细胞转化、胃肠道肿瘤的相互关系，以及胃肠道肿瘤治疗新途径。     

博安霉素及其单克隆抗体偶联物对裸鼠移植的人大肠癌的作用

大肠癌是我国常见的消化道恶性肿瘤之一,对大多数化疗药物均不敏感。本研究用裸鼠移植的人大肠癌动物模型,观察博安霉素及其与单克隆抗体R19偶联物对人大肠癌生长的抑制作用。在裸鼠可以耐受的剂量下,博安霉素对移植的人大肠癌(人结肠癌HT-29和人盲肠癌HCe-8693)均有明显的抑制作用。博安霉素15mg/kg和10mg/kg对人结肠癌HT-29的抑制率为92％和89％(P<0.01)对肓肠癌HCe-8693的抑制率分别为92％和190％(P<0.01)。博安霉素MMC和5-Fu在相当于l/9 LD50的剂量进行比较,博安霉素对HT-29的抑制率为82％;MMC为53％;而5-Fu仅为12％。博安霉素对人大肠癌的抑制作用明显较MMC(P<0.05)和5-Fu(P<0.01)为强。用人盲肠癌HCe-8693细胞免疫Lou/C大鼠,用大鼠杂交瘤技术获得抗人大肠癌单抗R19。用免疫组织化学的方法检测了R19单抗对肠癌的特异性。用ABC法检测,发现R19对裸鼠移植的人结肠癌HT-29和人结肠癌的外科手术切块呈阳性反应,对人胚脑、胸腺、淋巴结、肺、脾和血细胞等呈阴性,对肾小管上皮细胞显示弱阳性。用DextranT-40为中介体的方法连接大鼠R19和博安霉素形成的偶联物兼有二者生物学活性。实验共设5组,各治疗组的剂量以博安霉素计算为1mg/kg,每周二次,共8次,最后通过称量瘤重计算各组的抑制率,游离博安霉素为52％;R19与博安霉素混合物为34％;无关抗体M_3与博安霉素偶联物为48％;R19与博安博素偶联物为90％,显著地高于其它几组(P<0.01)。综上所述,博安博素及其与R19的偶联物对裸鼠移植的人大肠癌均有明显的抑制作用,尤其是博安霉素和单抗R19的偶联物在同等剂量下对肿瘤的抑制率显著地高于游离博安霉素。     

急性白血病患者血管内皮生长因子含量的测定及意义

目的研究血管内皮生长因子（VEGF）与急性白血病（AL）诊断及预后的关系.方法应用定量酶联免疫吸附实验测定25例不同病期AL患者（10例初诊未治、10例完全缓解期、5例复发患者）和9例正常对照者血清中VEGF的质量浓度.结果初诊未治、复发AL患者的VEGF含量高;完全缓解患者的VEGF质量浓度下降;VEGF高者完全缓解...     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. 低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
