研究了云芝在PDY培养基中的漆酶活性;漆酶的产酶高峰在第9天出现,酶反应的最适温度为25℃,最适酸碱度为pH4.6,K^+,Zn^2+等离子可提高漆酶的活性,而Ag^+离子则以漆酶活性有明显的抑制作用。  相似文献

研究了多孔菌漆酶的产酶曲线及酶作用最适条件;结果表明在该培养条件下,多孔菌第9天达产酶高峰,峰值酶活为412u/ml,酶作用的最适pH值为4.2,最适酶解温度为25℃,Mg2 、Mn2 对漆酶有激活作用,而Ag 、Fe3 和Cl-则能明显抑止漆酶活性.  相似文献

【目的】重点研究秀珍菇废渣中的木质素降解酶系。【方法】对秀珍菇废渣中可能存在的3种木质素降解酶:锰过氧化物酶(Manganese peroxidase,MnP)、木质素过氧化物酶(Lignin peroxidase,LiP)及漆酶(Laccase,Lac)进行活力测定,并对漆酶的最适pH值和最适温度进行考察。【结果】在秀珍菇废渣中未检测到锰过氧化物酶活力,检测到漆酶和木质素过氧化物酶活力。漆酶的最适pH值为6.2,最适温度为30℃。【结论】较一般食用菌不同,秀珍菇废渣中同时具有木质素过氧化物酶和漆酶活力,并且木质素过氧化物酶活力还较高。  相似文献

产漆酶真菌的筛选、培养及对苯酚的降解   总被引:7,自引:0,他引:7  
提出了一种新的从土壤中筛选产漆酶真菌的方法.将51个土壤样品分别分散于无菌水中,接种至含有底物A的固体培养基上,培养3d后,当菌落周围的培养基变成褐色时表明该菌株可能产生漆酶.将筛选出的若干菌株与市购的几种真菌分别接种于LB液体培养基中培养7d后,测定培养液中漆酶的活性,表明从土壤样品中筛选得到的真菌L1产酶活性最高.研究了各种培养条件对真菌L1产漆酶能力的影响,并且对该漆酶的性质及其对苯酚的降解进行了研究.结果表明,从产漆酶真菌中筛选出的自选菌(L1),在苯酚作为诱导剂、30℃下PYGM培养基中培养7d,产酶活性最高.该漆酶的最适pH为8.0,最适温度为33℃,Cl^-对其活性抑制较大.该漆酶对浓度为(50-250mg/L)的苯酚有较好的催化降解效果,在24h内最大降解率可达60%以上.  相似文献

环氧基改性的丙烯酸系树脂含环氧基,环氧基可在温和条件下与蛋白质上的亲核基团氨基反应而实现其固定化.研究了环氧基改性的离子交换树脂固定化漆酶的最优条件和固定化漆酶的酶学性质,结果表明,固定化漆酶时,在硫酸铵浓度为1.5mol·L-1、加酶量0.2U·g-1、pH 6.5、温度35℃条件下固定3h,测得固定化酶活性最好,比活力为0.026U·g-1,活力回收率为3.67%;该固定化酶催化反应的的最适温度为45℃,最适pH 4.5,米氏常数(Km)为1.233×10-3 mmol·L-1.锰离子、铁离子和钙离子在4×10-3~4×10-4 mol·L-1浓度之间时对固定化漆酶活性有抑制作用,在4×10-5~4×10-6 mol·L-1浓度之间时有激活作用.  相似文献

采用壳聚糖铜固定化和交联酶聚集体的方法固定化漆酶.探索了固定化前后漆酶的最适温度、pH,热稳定性和金属离子对其活性的影响,交联漆酶聚集体制备的条件.结果为:游离漆酶的最适温度是20℃,最适pH是4.6;壳聚糖铜固定化漆酶的最适温度是25℃,最适pH是4.0;交联漆酶聚集体的最适温度是15℃,最适pH是3.6.固定化后漆酶的热稳定性提高,不同金属离子影响固定化漆酶的活性,其中K+的激活作用尤为明显.制备交联漆酶聚集体最适戊二醛浓度为1%,最适交联时间是2 h.  相似文献

以Cerrena sp. HYB07菌株所产漆酶为研究对象,制备磁性Fe_3O_4-壳聚糖固定漆酶.磁性壳聚糖微球固定化漆酶制备的最优条件为:磁性Fe_3O_4纳米颗粒0.4μg·mL~(-1)、戊二醛质量浓度4 mg·mL~(-1)、交联时间12 h、给酶量160 U·mL~(-1)、固定化时间8 h.在此条件下,漆酶固定化率为78.03%,固定化漆酶活力为97.19 U·g~(-1).酶学性质研究表明,固定化漆酶的最适反应pH值为3.0,最适反应温度为45℃.与游离酶相比,固定化漆酶的热稳定性有所提高.固定化漆酶用于蒽醌染料活性亮蓝脱色,具有良好的重复利用性,不仅染料脱色率优于游离酶,且在汞离子存在下也效果显著.  相似文献

作者对粗毛栓菌(Trametesgallica)在PAG培养基条件下摇瓶培养产生漆酶的情况进行了研究.15d左右达到产酶高峰,随后进入平台期.对15d产生的漆酶进行分离纯化.4L发酵液经硫酸铵沉淀,DEAE SepharoseFastFlow柱层析和SephadexG 100柱层析,漆酶被纯化了36.9倍.经SDS PAGE验证为一条带,表观分子量约为60,000Da.纯化漆酶是一种含糖量6%的糖蛋白,且在605nm处具有蓝色漆酶Ⅰ型铜离子典型的吸收峰.以ABTS,DMP和愈创木酚为底物,反应的最适pH值分别为2.2,3.0和4.0;Km值分别为0.172×10-2,0.51,0.48mmol/L.以ABTS为底物,最适反应温度为70℃.在pH7~9处理24h,纯化漆酶仍保持较高活性.EDTA,卤素离子,NaCN和NaN3对酶活有抑制作用,其中NaN3是最强的抑制剂.纯化的蓝色漆酶不具有氨酸酶活性,但能够氧化非传统底物酚红.  相似文献

对糙皮侧耳(Pleurotus ostreatus)Ax3菌的漆酶产生条件进行了研究,该菌产漆酶较高的条件是:培养温度25℃,培养基pH5.5,每升C/N含量0.252g/0.021g;加入0.05%~0.1%的吐温可提高漆酶酶活12倍。对漆酶的部分生化性质研究表明:最适反应温度25℃;最适反应pH2.8;琥珀酸缓冲液效果最好;金属离子:Mn^2 ,Cu^2 ,Zn^2 对漆酶有较强的激活作用,其中Mn^2 相对活性最高,Ag^ ,K^ ,Ca^2 ,Fe^2 对漆酶活性有抑制能力,其中Ag^ 的抑制作用明显。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
