Effect of cyclophosphamide on development of reticulum cell sarcoma in SJL/J mice

Summary Studies to determine the effect of cyclophosphamide (CY) on the development of reticulum cell sarcoma (RCS) in SJL/J mice indicated a dependence on the duration of the test period. Age also appeared a factor of importance. Thus, a comparison of tumor incidences at 52 weeks of age showed maximal inhibition when CY was administered at 40 weeks, minimal inhibition when the drug was given at 30 weeks, and intermediate inhibition when treatment was initiated at 10 and 20 weeks. Consistent with these findings, long-term treatment of 40-week-old SJL/J mice with low doses of CY resulted in an increase in the mean survival time and in a reduction in the incidence of RCS. Stem cells in the peripheral blood of rauscher leukemic mice

Zusammenfassung CBA Mäuse zeigen nach der Infektion mit dem Rauscher Virus einen starken Anstieg hömopoetischer Stammzellen («colony-forming units») im peripheren Blut. Nach 21 Tagen wird im Zustand der peripheren Erythroblastose eine 50-fache Zunahme gefunden.

Adenosine triphosphate and diphosphoglycerate levels in red blood cells from patients with Down's syndrome

Summary The levels of ATP and ATP plus DPG were significantly elevated in erythrocytes from Down's syndrome patients when compared to erythrocytes from age matched controls. The hemoglobin content and hematocrit values were significantly reduced. The resultant tendency towards anemia probably explains the elevation in metabolite levels.Acknowledgments. Guinea pig liver cytosol has an L-threonine deaminase activity distinct from L-serine deaminase]

Using sodium sulfate precipitation, "Sephadex G200" gel filtration and polyacrylamide gel electrophoresis, a L-threonine desaminase was demonstrated in the Guinea-Pig liver cytosol. This enzyme was separated from the guinea pig liver L-serine desaminase possessing an auxiliary activity on L-threonine substrate described by us in a previous work. The optimals for pH (7,1) and temperature (+ 55 degrees C) and the apparent molecular weight (134,000 + 20,000) were established.     

Interaction between peripheral blood leucocytes and X-irradiated cells from lymphoid cell lines

A variety of organoselenium compounds were originally described as antiinflammatory, antioxidant or glutathione-peroxidase-like agents, and as inhibitors of prostaglandin and leukotriene synthesis. Recently, the compounds have also been found to be inducers of interferon gamma and tumor necrosis factor in human peripheral blood leukocytes (PBL). We evaluated the effects of bis [2-(N-phenylcarboxamido)phenyl] diselenide and Ebselen; 2-phenyl-1,2-benzisoselenazol-3(2H)one, on the incorporation of tritiated thymidine into the DNA of PBL cultured in vitro. Both compounds were mitogenic and this effect was correlated with the expression of interleukin 2 receptor in T-lymphocytes. Therefore, we suggest that the selenoorganic compounds may induce mitogenic cytokines.

Comparison of protein synthesis profiles in chronic lymphocytic leukaemia cells and B-lymphocytes from peripheral blood,cord blood and tonsil

2D-gel electrophoresis was used to investigate protein synthesis in leukaemic cells from a series of 15 chronic lymphocytic leukaemia (CLL) patients, and in non-malignant B-cell populations from different sources. The protein synthesis profiles of CD5+ B-cells from umbilical cord blood and from tonsil were determined, and the levels of expression of their proteins were observed to be similar to the CLL cells. The CD5-cells from cord blood resembled peripheral blood B-lymphocytes, and the protein synthesis profile of CD5-cells from tonsils was very complex. One protein was also identified which consistently appeared to be synthesised at a low level in CD5+ B-cells from tonsil but which was always more prominant in CLL cells and other non-malignant B-lymphocytes. On the basis of these data it is possible that the closest non-malignant counterpart to CLL is the CD5+ B-lymphocyte from cord blood.

The sensory organs of skeletal muscles, the muscle spindles, were examined using electron microscopy in dy2J/dy2J dystrophic mice. Despite widespread damage to the extrafusal (skeletomotor) fibres the intrafusal (spindle) fibres appeared normal and seemed resistant to the aetiological factors for murine dystrophy.     

Inhibition of interleukin-1 beta release from cultured human peripheral blood mononuclear cells by prednisolone

Prednisolone, a water-soluble glucocorticoid hormone, suppressed the secretion of interleukin-1 beta from human peripheral blood mononuclear cells in culture. The prednisolone-induced suppression of the monokine release was dose-related and the half maximal response was observed at 0.1 nM.

Preparation of lymphocytes from small volumes of peripheral mouse blood

Summary A method was developed which allowed for the isolation of lymphocytes (95% of leukocytes in the final suspensions) from small volumes of peripheral mouse blood (38% recovery). The method proved of value for studies of murine lymphocytes which required biochemical analysis of samples from individual animals on a serial basis.This work was supported in part by a University College Faculty Research Grant.     

Stimulation of peripheral guinea pig blood lymphocytes by autologous epidermal cells]

Using a strictly autologous (and not syngeneic) mixed culture system in the guinea-pig, it was shown that Mitomycin C treated ear epidermal cells (E) were capable of stimulating peripheral blood lymphocytes (L) from the same animal. The proliferative response of the lymphocytes as measured by 3H thymidine incorporation, is optimal on the 5th day of culture, and when the ratio of the concentrations of the two cell populations (L : E) is 1 : 1. The autoreactivity of the lymphocytes varies with the animal.     

Histochemical demonstration of differences in AMP deaminase activity in rat skeletal muscle-fibres

Summary The histochemical method for AMP deaminase confirms that rat slow-twitch fibres contain less AMP deaminase activity than fast-twitch fibres. However, the enzyme appears similarly localized in all fibre types.This study was supported by NIH grant AM 21617.
