New contribution to the study of chromosomes of the European Cryptocephalinae (Coleoptera,Chrysomelidae)

Summary The chromosomes of three species ofPachybrachis and nine ofCryptocephalus chrysomelids were analyzed. The male meiotic bivalent formula ofP. azureus Suffr.,P. catalonicus Burl. andP. petitpierrei Daccordi is 7^II+Xy_r.Cryptocephalus sexmaculatus Ol. andC. vittula Suffr. have 13^II+Xy_p,C. bipunctatus L. 14^II+Xy_r,C. ochroleucus Steph. andC. ocellatus Drap. 14^II+Xy_p,C. crassus 01. 15^II+Xy_r,C. sulphureus 01. 15^II+Xy_p, the same number as inC. fulvus Goeze with 2n=32 chromosomes, whileC. primarius Har. has 19^II+Xy_p. The modal chromosome number inCryptocephalus is 2n=30 (about 60% of spp.), and most species are characterized their small chromosomes. The low variation found in the karyotypes of Cryptocephalinae along with their possible interrelationships with allied chrysomelid subfamilies are also discussed.     

Targeting O 6-methylguanine-DNA methyltransferase with specific inhibitors as a strategy in cancer therapy

O ⁶-methylguanine-DNA methyltransferase (MGMT) repairs the cancer chemotherapy-relevant DNA adducts, O ⁶-methylguanine and O ⁶-chloroethylguanine, induced by methylating and chloroethylating anticancer drugs, respectively. These adducts are cytotoxic, and given the overwhelming evidence that MGMT is a key factor in resistance, strategies for inactivating MGMT have been pursued. A number of drugs have been shown to inactivate MGMT in cells, human tumour models and cancer patients, and O ⁶-benzylguanine and O ⁶-[4-bromothenyl]guanine have been used in clinical trials. While these agents show no side effects per se, they also inactivate MGMT in normal tissues and hence exacerbate the toxic side effects of the alkylating drugs, requiring dose reduction. This might explain why, in any of the reported trials, the outcome has not been improved by their inclusion. It is, however, anticipated that, with the availability of tumour targeting strategies and hematopoetic stem cell protection, MGMT inactivators hold promise for enhancing the effectiveness of alkylating agent chemotherapy.     

Araplysillins-I and-II: Biologically active dibromotyrosine derivatives from the spongePsammaplysilla arabica

Summary Araplysillins-I and-II, two novel dibromotyrosine derivatives, were isolated fromPsammaplysilla arabica and their structure was elucidated by spectroscopic methods. They proved to be inhibitors of Na⁺/K⁺ ATPase and to have antimicrobial activity.     

On the synthesis of serine and homoserine samples asymmetrically labelled with tritium and deuterium in the hydroxymethylene group

Summary (1 R) [1-³H,²H₁] 3-Phenylpropanol, the key intermediate in the synthesis of (4 R) [4-³H,²H₁] D, L-homoserine and of the (4 S)-isomer, is obtained from (1 S) [1-²H₁] 3-phenylpropanol and (1 RS) [1-³H] ethanol upon incubation with yeast alcohol dehydrogenase and NAD⁺; under similar conditions 2-phenylethanol undergoes very small exchange with [1-²H₂] ethanol.     

Effect of strychnine on the electroretinogram of the isolated rabbit retina

Zusammenfassung Strychnin in einer Konzentration von 3×10^–6 bis 3,5×10^–5 M/l verursacht eine vorübergehende Vergrösserung derb-Welle des ERG der isolierten Kaninchennetzhaut, in einer Konzentration von 7×10^–5 bis 7×10^–4 M/l verkleinert es dieb-Welle. Aus den Ergebnissen wird geschlossen, dass die Substanz zumindest teilweise schon auf die Schicht des zweiten Neurons wirkt.     

Functionality of the TRPV subfamily of TRP ion channels: add mechano-TRP and osmo-TRP to the lexicon!

Transient receptor potential (TRP) ion channels have been identified as cellular sensors responding to diverse external and internal stimuli. This review will cover the TRPV subfamily that comprises vertebrate and invertebrate members. The six mammalian TRPV channels were demonstrated to function in thermosensation, mechanosensation, osmosensation and Ca²⁺ uptake. Invertebrate TRPV channels, five in Caenorhabditis elegans and two in Drosophila, have been shown to play a role in mechanosensation, such as hearing and proprioception in Drosophila and nose touch in C. elegans, and in the response to osmotic and chemical stimuli in C. elegans. We will focus here on the role that TRPV ion channels play in mechanosensation and a related sensory (sub-)modality, osmosensation. The effect of X-rays and EMS on the behavior of the transposing element,TE98, inDrosophila melanogaster

Summary The sensitivity of TE98 (carryingw ^+R andrst ⁺) to X-rays does not differ significantly from the mutability ofcurled andkarmoisin loci. In addition no spontaneous mutants of TE98 were recovered, indicating its extreme stability. On the effect of EMS nowhite mutants were found supporting the view that thew ⁺ gene of TE98 is duplicated.     

Redundant and unique roles of coronin proteins in <Emphasis Type="Italic">Dictyostelium</Emphasis>

Dictyostelium discoideum harbors a short (CRN12) and a long coronin (CRN7) composed of one and two beta-propellers, respectively. They are primarily present in the cell cortex and cells lacking CRN12 (corA ⁻) or CRN7 (corB ⁻) have defects in actin driven processes. We compared the characteristics of a mutant cell line (corA ⁻ /corB ⁻) lacking CRN12 and CRN7 with the single mutants focusing on cytokinesis, phagocytosis, chemotaxis and development. Cytokinesis, uptake of small particles, and developmental defects were not enhanced in the corA ⁻ /corB ⁻ strain as compared to the single mutants, whereas motility and phagocytosis of yeast particles were more severely impaired. It appears that although both proteins affect the same processes they do not act in a redundant manner. Rather, they often act antagonistically, which is in accordance with their proposed roles in the actin cytoskeleton where CRN12 acts in actin disassembly whereas CRN7 stabilizes actin filaments and protects them from disassembly.     

Interaction oft alleles at theT locus in the house mouse

Zusammenfassung Mehrfach wurde gezeigt, dass zwei rezessive letale Allele in der Hausmaus (t ^o undt ¹) lebensfähige normale Tiere in heterozygoter Kombinationt ⁰/t ¹ produzieren. Neuere Daten machen wahrscheinlich, dass dast ¹-Allel mutativ abänderte, so dass sowohl seine homozygote Wirkung als auch die Zusammenwirkung mitt ⁰ beeinflusst worden ist. Hingegen sind Kombinationen vont ⁰ und dem neuen Allel nicht mehr lebensfähig, und Homozygote sterben erst nach der Implantation.

A reappraisal of the hormonal regulation of larval fat body histolysis in femaleDrosophila melanogaster

The histolysis of larval fat body cells in adult femaleDrosophila melanogaster was examined in wild type and mutant animals. The fat body cells of wild type (Canton-S),apterous ^56f homozygotes,apterous ^78jts homozygotes and heterozygotes,apterous ⁴/+, ecdysoneless¹ homozygotes and heterozygotes all underwent histolysis normally during the 72 h following adult eclosion. Only in the case ofap ⁴/ap⁴ adults did the cells fail to histolyze normally. The fat body cells of both diapausing and non-diapausing wild type females underwent histolysis at the same rate. Attempts to demonstrate histolysis in vitro were unsuccessful, even in the presence of juvenile hormones (JHs), larval ring glands, or adult ovaries. In all strains other than theap ⁴ homozygotes, a significant proportion of larval fat body cells were dead at any time while theap ⁴/ap⁴ animals, almost all cells remained viable. It is postulated that fat body cell lysis following eclosion is not a JH-mediated event, but is elicited by an as yet unidentified factor(s), possibly originating in the ovary.     

Influence of turbulence on electrokinetic phenomena; experimental determination of the thickness of the diffuse part of the double layer

Zusammenfassung Durch Messungen in 10^–5, 10^–4 und 10^–3 N benzolischen Lösungen von Zn-di-i-propylsalizylat haben wir einen beträchtlichen Einfluss von Turbulenz auf Strömungsströme feststellen können. Dieser Einfluss kann ausgedrückt werden durch einen Faktorf, der in einem unserer Experimente einen Wert vonf=66 erreichte; aus den Werten vonf kann die Dicke der diffusen Doppelschicht in der Flüssigkeit berechnet werden.     

Changes induced by the acute administration of iodide on secretion of iodinated components by the rat thyroid gland perfused in situ

Résumé Nous avons étudié l'effet de l'iodure 127 (10^–3 et 10^–4 M) sur la sécrétion de T₄ ^*, T₃ ^* et –I^* par la thyroïde de rat, prémarquée à l'iode 131 et perfusée in situ avec du sang non radioactif contenant 12 mU de TSH/ml. Il n'a pas été noté de changement significatif dans la sécrétion de T₄ ^* ou T₃ ^* jusqu'à 45 min après le début de l'administration d'iodure 127, qu'il y ait ou non du méthimazole (10^–3 M) dans le sang perfusé. Il y avait par contre une augmentation de la sécrétion d'-I^* pendant la perfusion de 10^–3 M d'ïodure, indiquant une décharge d'-I^* pendant la perfusion de 10^–3 M d'ïodure, indiquant une décharge d'-I^* de provenance intrathyroïdienne. Cet effet est similaire à celui noté auparavant pendant le perfusion de 10^–3 M de ClO ₄ ^– .     

Chemical defense in the three European species ofCrematogaster ants

The composition of the Dufour gland of the antC. scutellaris has been reinvestigated by gas chromatography/mass spectrometry. The major components of the gland are (2E,5E,12Z)-4-oxoheneicosa-2,5,12-trien-1-ol acetate (1a) its ¹⁴ and ¹⁶ double bond isomers (1b and1c), and the corresponding (Z,Z)-dienes5a and5b, all containing an acetylated C₂₁ chain. The previously proposed structures1d, 1e, and5c, which are based on an homologous acetylated C₂₃ chain, correspond to minor derivatives present in the gland. Traces of acetylated C₁₉ homologs, tentatively identified as1g-1i, have also been found. The Dufour gland contents of the two other EuropeanCrematogaster species have also been studied.C. auberti is very similar toC. scutellaris in producing mainly1a, 1b and1c, together with the same higher and lower homologs, but it lacks the dienic derivatives5, whereasC. sordidula contains essentially the acetylated C₁₉ compounds1g, 1h, and1i, accompanied by acetylated C₁₇ homologs.     

A reinvestigation of the chemistry of the Dufours gland of the formicine ant,Anoplolepis custodiens

The components of individual Dufours glands excised fromAnoplolepis custodiens workers were analysed by GC-MS. In addition to then-alkanes andn-alkenes previously reported² in these glands, primary alcohols (C₁₉-C₂₂), secondary alcohols (C₂₀-C₂₃), 2-ketones (C₂₀-C₂₃) and possibly carboxylate ethyl esters (C₁₉ and C₂₁) were identified as components of these glands. It seems possible that these high-boiling compounds are used by the workers in laying trails on the hot sandy surfaces of their characteristic habitat and in lining of the inner walls of nests, but no standard compounds have been available to us for any behavioral studies.     

Intraspezifische Unterschiede der relativen Hirngrösse beim Löwen (Panthera leo L.)

Summary Different stages of brain evolution expressed by the allometric relation of³ brain capacity and basal length of the skull are shown to be existent in the speciesPanthera leo. Whereas Asiatic lions obviously have the same level of brain size as leopards (Panthera pardus), African lions have higher brain capacities. A third level seems to be represented by the upper pleistocene American lion,Panthera leo atrox. These results permit us to reject some conceptions ofHerre andRöhrs ¹³ concerning the quantitative expression of mammalian brain evolution.     

High K+-induced release of somatostatin from the cortical preparation of rat brain

Summary Release of endogenous somatostatin (SRIF) from the rat cerebral cortical slices incubated in Krebs-bicarbonate buffer was increased from the basal rate of 3.4±0.6% of the total SRIF content in 15 min at [K⁺]_o=5.6 mM, to 13.1±1.6% upon raising the [K⁺]_o to 56.6 mM. The high-K⁺ evoked SRIF release was absent when Ca⁺⁺ in the medium was replaced by Mn⁺⁺. The isolated synaptosomes from rat cerebral cortex contain 13.2±3.1 ng SRIF/mg protein compared to 0.33±0.01 ng/mg protein in the cortical tissue as a whole, suggesting that nerve terminals are the main source of the peptide released upon membrane depolarization.The study was supported by a grant from the Medical Research Council of Canada. The effect of perchlorate ion and some pharmacological agents on the sensitivity of the frog (Rana pipiens) rectus abdominis to acetylcholine

Zusammenfassung Die Reaktion des mit Eserin vorbehandelten musculus rectus abdominis des Frosches auf Acetylcholin liess sich durch Oxotremorin (2.0·10^–5 M), Arecolin (2.0·10^–5M), Aceclidin (1.0·10^–4 M) und Carbachol (2.0·10^–7 M) verstärken, während Atropin (1.0·10^–5 M) hemmend wirkte. Perchlorationen in Konzentrationen über 2 · 10^–4 M sensibilisierten den Froschmuskel erheblich und reduzierten die Grenzdosis, bei der eine Potenzierung durch Substanzen mit muskarinähnlicher Wirkung beobachtet wird. Alle beschriebenen Reaktionen liessen sich durch Gehirnextrakte verstärken, in denen zuvor Acetylcholin durch Kochen in alkalischer Lösung zerstört worden war.

Forecasting for the generation of trading signals in financial markets

In this paper we show that optimal trading results can be achieved if we can forecast a key summary statistic of future prices. Consider the following optimization problem. Let the return r_i (over time i=1, 2, ..., n) for the ith day be given and the investor has to make investment decision d_i on the ith day with d_i=1 representing a ‘long' position and d_i=0 a ‘neutral' position. The investment return is given by r=Σⁿ_i=1r_id_i−cΣⁿ⁺¹_i=1∣d_i−d_i−1∣, where c is the transaction cost. The mathematical programming problem of choosing d₁, ..., d_n to maximize r under a given transaction cost c is shown to have an analytic solution, which is a function of a key summary statistic called the largest change before reversal. The largest change before reversal is recommended to be used as an output in a neural network for the generation of trading signals. When neural network forecasting is applied to a dataset of Hang Seng Index Futures Contract traded in Hong Kong, it is shown that forecasting the largest change before reversal outperforms the k‐step‐ahead forecast in achieving higher trading profits. Multiple roles of the DSCR1 (Adapt78 or RCAN1) gene and its protein product Calcipressin 1 (or RCAN1) in disease

The DSCR1 (Adapt78) gene¹ is transiently induced by stresses to temporarily protect cells against further potentially lethal challenges. However, chronic expression of the DSCR1 (Adapt78) gene has now been implicated in several pathological conditions including Alzheimer’s disease, Down syndrome and cardiac hypertrophy. Calcipressin 1 has been shown to function through direct binding and inhibition of the serine threonine protein phosphatase Calcineurin. Pharmacological inhibition of calcineurin, by the immunosuppressive drugs cyclosporin A and FK506, affects a wide variety of diseases. It is, therefore, likely that this endogenous calcineurin inhibitor, calcipressin 1, may also play a role in a variety of human diseases. ¹Please note that the mammalian DSCR1 gene is also called Adapt78 or RCAN1, and its protein products have been named Calcipressin1, MCIP1 and RCAN1. A proposal to adopt a single gene name of RCAN1 and a protein name RCAN1 (for Regulator of Calcineurin) has been endorsed by the HUGO Gene Nomenclature Committee, but final approval must await agreement from a majority of researchers in the field. Microbiological reduction of steroidal ketones using the thermophilic bacteriumCaldariella acidophila

Summary Twelve steroidal ketones have been subjected to reduction withC. acidophila resting cells, regio- and stereospecific reduction of the 3-keto groups being observed, as well as reduction of the ⁴-double bond. The presence of oxo groups at C-11 or C-12 and the presence of hydrophobic side chains on the steroidal molecules inhibit the reduction.
