Reduction of high affinity glutamate uptake in rat hippocampus by two polyamine-like toxins isolated from the venom of the predatory waspphilanthus triangulum F

Summary Two components of the venom of the predatory waspPhilanthus triangulum F. significantly reduce — to a greater or less extent — the high affinity uptake of glutamate in rat hippocampus. A concentration of 10 M -PTX caused a reduction of 74%, while the other component, -PTX, at the same concentration, caused a reduction of 18%. Hence the effect of -PTX on high affinity glutamate uptake in the hippocampus is comparable with its effect on high affinity glutamate uptake in insect neuromuscular junctions. Contrary to our previous findings that -PTX has no effect on high affinity glutamate uptake in insect glutamatergic terminal axons, however, -PTX significantly reduces high affinity glutamate uptake in the hippocampus, albeit less effectively than -PTX.

Exchange of tritium from randomly tritiated taurocholate by microbial bile salt oxidoreductases

Summary Tritium distribution on randomly labelled taurocholate (TC) was estimated at 28%, 4%, 1% and <0.5% on the hydrogens opposite the 3-, 7- and 12-OH groups and taurine moiety respectively. Anomalously,C. perfringens 3-hydroxysteroid dehydrogenase (3-HSDH) catalyzed tritium loss of 36% on formation of 7-, 12-dihydroxy-3-keto-5-cholanoate, implying additional losses of tritium at other sites by this enzyme.This work is supported by the National Cancer Institute.     

Enzymatic basis for protection of fish embryos by the fertilization envelope

The mechanism by which the fertilization envelope (FE) is able to protect the embryo of fish until hatching is almost unknown, except for its function as a physical barrier. FE extract from activated or fertilized eggs of the fishSalmo gairdneri was demonstrated to contain enzyme activities using an agar plate enzyme assay. The enzymes apparently active were carboxymethylcellulase (cellulase; EC 3.2.1.4), laminaranase (endo-1,3(4)--glucanase; EC 3.2.1.6), carboxymethylchitinase (chitinase; EC 3.2.1.14), xylanase (endo-1,4--xylanase; EC 3.2.1.8), mannanase (mannan 1,2-(1,3)--mannosidase; EC 3.2.1.77), dextranase (EC 3.2.1.11), a protease and lysozyme (EC 3.2.1.17). The FE extract exerted an antifungal or fungicidal action on the fungusSaprolegnia parasitica, whereas an extract from the vitelline envelopes (VE) has no apparent enzyme activity nor antifungal or fungicidal action. Enzymes acquired by the FE through the cortical reaction may have an important defensive role, protecting the embryo against invaders or pathogens.     

Storage of 5-hydroxytryptamine in human blood platelets

Zusammenfassung Menschliche Blutplättchen zeigten die höchste 5-Hydroxytryptamin (5HT)- und Adenosin-5-Triphosphat (ATP)-Konzentration in einer subzellulären Fraktion, die elektronenoptisch aus Vesikeln von unterschiedlicher Dichte bestand. Diese waren deutlich verschieden von anderen subzellulären Organellen, z.B. den -Granula. Inkubation der Plättchen mit 5 HT erhöhte die Elektronendichte und den 5 HT-Gehalt der Vesikel, veränderte aber die -Granula nicht, Es wird geschlossen, dass in menschlichen Blutplättchen ein Teil des 5 HT zusammen mit ATP in spezifischen subzellulären Organellen gespeichert ist.     

Defensive steroids from a carrion beetle (Silpha americana)

Summary The defensive anal effluent discharged bySilpha americana in response to disturbance contains a mixture of steroids stemming from a glandular annex of the rectum. The compounds have been characterized as 15-hydroxyprogesterone (1, principal component), 5-pregnan-15-ol-3, 20-dione (2), 5-pregnan-3, 15-diol-20-one (3), 5-pregnan-7, 15-diol-3,20-dione (4), 5-pregnan-3, 7, 15-triol-20-one (5), 5-pregnan-16-ol-3,20-dione (6), and 5-pregnan-3, 16-diol-20-one (7), none previously found in insects. Bioassays with jumping spiders showed compounds1 and6 to be feeding deterrents at the 1 g level.Paper No. 78 of the series: Defense Mechanisms of Arthropods. Summary -Asp¹-Angiotensin II had a more pronounced action on the blood pressure of nephrectomized rats than the corresponding-compound.-Asp¹-Angiotensins were more slowly destroyed by rat serum, rat kidney homogenate, and human plasma than-compounds, especially-Asp¹-Angiotensin II amide.     

Versuche zum enzymatischen Abbau der «gebundenen» gibberelline vonPharbitis purpurea

Summary Seeds and fruit walls ofPharbitis purpurea contain in addition to the free gibberellins A₈, A₃, A₅, more polar gibberellin-like substances, so-called bound gibberellins tentatively namedPharbitis and. After purification by thin layer chromatography, these 2 substances were treated with both Ficin and Emulsin in buffered solutions of pH 6.2. After treatment with Emulsin, the free gibberellins A₈ and A₅, after treatment with Ficin the gibberellins A₃ and A₅, were detectable in thin layer chromatograms.

Positive correlation of responsiveness to catecholamines of the rat liver glycogenolytic receptor with other α-receptor responses

Zusammenfassung Durch die vollständige Dosis-Wirkungskurve der Katecholamin-i.p.-induzierten Leber-Glykogenolyse an der Ratte konnte deren Einfluss auf die -Rezeptoren nachgewiesen werden. Da eine Wirkung auf die -Rezeptoren (Lipolyse) ausschied, konnte es sich nur um einen -Effekt handeln.     

Casein,a prohormone with an immunomodulating role for the newborn?

Summary Maternal colostrum and milk, the earliest food of the newborn, should not only be considered as supplying nutrients, but also as agents providing protection against aggressions from the new environment. Indeed by enzymatic digestion of the main milk proteins, the caseins, biologically active peptides are released; they may be implicated in the stimulation of the newborn's immune system. From this point of view a strategic active zone has been characterized in -casein. A possible role of casein as a prohormone for the newborn is suggested.     

Hydrolysis of amino acid β-naphthylamides by aminopeptidases in the parotid gland

Zusammenfassung Die Aktivität der Aminopeptidasen in Ohrspeicheldrüsen wurde gemessen. Glycyl-Prolin-naphthylamid, Alanin-naphthylamid, Leucin-naphthylamid, Methionin-naphthylamid, und Arginin-naphthylamid wurden von der Mikrosomenfraktion und der löslichen Fraktion schnell gespalten. Das Glycyl-Prolin-naphthylamid spaltende Enzym war in Ohrspeicheldrüsen in relativ grösserer Menge vorhanden. Die Aufspaltung von Glycyl-Prolin-naphthylamid in Glycyl-Prolin und-Naphthylamin wurde papierchromatographisch nachgewiesen.     

Diminution of 5-hydroxytryptamine in thrombocytesin vitro by chlorpromazine and related compounds

Riassunto Cloropromazina, imipramina, cloroprotixene e amitriptilinain vitro diminiscono la 5-idrossitriptamina (5HT) endogena nei trombociti di coniglio e, per quanto si è visto, anche in quelli umani. Nel plasma di coniglio pretrattato con isocarbossazide, inibitore della mono-amino-ossidasi, si osserva un corrispondente aumento di 5HT. Cocaina, -metil-dopa e triptamina hanno un effetto meno pronunciato sulla 5HT delle piastrine.

A possible pathway for the biosynthesis of adenochromines

Ein neues Verfahren zur Herstellung von 19-Norsteroiden

Summary The new methods for the production of 19-norsteroids described involve the conversion of 5-halogen-6-hydroxy-steroids into the corresponding 5-halogen-6:19-ethers either with lead tetraacetate or by the hypoiodite reaction^8,10. The 6:19-oxygen bridge is then opened reductively either directly or after oxidation of the 6:19-ethers to lactones or preferably after introduction of a ^4,6-3-oxo grouping. Acylolytic cleavage of the ⁴-3-oxo-6:19-ethers followed by alkaline hydrolysis gives ^4,6-3-oxo-19-hydroxy-steroids. The 19-hydroxycompounds formed are easily converted into 19-norsteroids by known methods.

Comparative analysis of human milk and human blood plasma by means of diffusion-in-gel methods

Zusammenfassung Mit Hilfe von Immunoelektrophorese und Doppeldiffusiontechnik wurden in Humanmilch an Blutplasmaproteinen serologisch identifiziert: Praealbumin, Albumin, Acid Seromucoid, ₁-Glykoprotein (3·5 S), Coeruloplasmin, Haptoglobin, ₂- oder-Lipoprotein, Transferrin, Fibrinogen, _2A-Globulin, _2M-Globulin und-Globulin.     

Potentiating effect of phenylephrine on isoproterenol activation of thyroxine type II deiodinase in the pineal gland of adult rats

In the present study we show, for the first time, that phenylephrine (PHE), an -adrenergic receptor agonist, potentiates the effect of isoproterenol (ISO), a -adrenergic agonist, in activating pineal type II5-deiodinase (5-D) activity. The potentiating effect of PHE was observed only at doses of ISO which induce submaximal activation of the enzyme. However, at doses which lead to maximal activation of the enzyme, PHE was ineffective. The results suggest that not only -, but also -adrenergic receptors, are involved in the sympathetic noradrenergic regulation of pineal 5-D activity in the adult rat.

Determinant of efficiency of a monomeric enzyme: Acceleration by site-specific molecules for trypsin

Summary The interaction of a specific ligand at substrate binding site was shown to be responsible for the catalytic efficiency of trypsin. Methemoglobin formation and reduction in relation to hemoglobin oxygen affinity

Summary It is suggested that although the high oxygen affinity hemoglobin (Hb LR) (143(H21)HisGln) with stabilization of R quaternary conformation is not more susceptible to the oxidizing effect of nitrites in vitro, in an in vivo situation where hemoglobin is partly deoxygenated, it might be more susceptible to methemoglobin formation due to higher oxidation and lower reduction rate.Supported by NIH grant No. 5 RO1 AM 20181-02 and VA grant No. 5455-002.Acknowledgments.
