Immunological and radioimmunological studies of membrane antigen(s) from human breast carcinomas and non-tumoral breast tissues. II

Summary The authors found that breast carcinomas contain 2 groups of antigens not detectable in normal tissue. The groups were extracted and partially purified. One responded to absorbed anti-CEA and the other only to an antibody, produced by immunizing rabbits with human breast carcinoma extract which were unabsorbed and absorbed with control breast tissue.We express our appreaciation to Dr Carla Zanchetti for help in preparing the figures, and to Mr Riccardo Sarri for technical help.     

Estimation of the methylating capacity of the pineal gland. With special reference to indole metabolism

Summary In order to obtain more information on the methylating capacity of the pineal gland, a method determining the formation of different 5-methoxyindoles in the pineal gland was developed. The method depends on measuring the incorporation of labelled methyl groups into the various hydroxyindoles present in the pineal gland, after incorporation of pineal tissue with labelled S-adenosyl methionine. Hydroxyindoles were not added to the incubation medium. After incubation thin-layer chromatography was performed with pineal tissue together with the incubation medium; the spots were scraped and counted.The authors wish to express their gratitude to Prof. Dr. J.C. van de Kamer and Dr F.C.G. van de Veerdonk for their stimulating interest in these studies.     

Selective and reversible modification of essential thiol groups of D-glyceraldehyde-3-phosphate dehydrogenase by isothiocyanates

Summary Aralkyl and alkyl isothiocyanates, like aryl isothiocyanates, undergo a selective and reversible reaction with essential thiol groups of D-glyceraldehyde-3-phosphate dehydrogenase; the former 2 substances require for a reversible reaction course a more alkaline medium and presence of a thiol.The authors are indebted to Dr J. Radioimmunoassay of some hormones simultaneously measured in serum and breast cyst fluid

Summary Blood and breast cyst fluid were drawn simultaneously for hormonal determination. There was no difference between serum and cyst fluid values of PRL and TSH. A significant difference was noted for LH (p<0.01) and FSH (p<0.05), serum concentrations being higher than cyst fluid concentrations.The authors are indebted to Miss J. Meister, E. Dullaert and J. Effect of thiamine hydrochloride on the blood level of 2-formyl 1-methyl pyridinium oxime chloride (2-PAM.Cl) in rats

Summary The biological half-life of 2-PAM.Cl was found to increase in female rats pretreated with thiamine hydrochloride (10 mg/kg i.m.). No such effect was observed in the male rats.Dr B.L. Inverse relationship between melanogenesis and endogenous hydroquinone

Summary Inhibition or stimulation of melanogenesis have been found to occur as a result of the alteration of hydroquinone levels in the body. Substances which stimulate melanogenesis are found to lower the level of hydroquinone in amphibia, and evidence for the relationship is also given by mammalian experiments.Acknowledgments. Summary In the present paper, an extension is presented of an earlier described method, by which the methylating capacity of the pineal gland can be determined. A comparative study of tryptophan metabolism in skins and livers of black and albino rats

Summary The skin of black rats contains less tryptophan than the skin of albino rats, possibly because the activity of tryptophan pyrrolase is higher. Adenosine triphosphate and diphosphoglycerate levels in red blood cells from patients with Down's syndrome

Summary The levels of ATP and ATP plus DPG were significantly elevated in erythrocytes from Down's syndrome patients when compared to erythrocytes from age matched controls. The hemoglobin content and hematocrit values were significantly reduced. The resultant tendency towards anemia probably explains the elevation in metabolite levels.Acknowledgments. Effect of tryptophan on tyrosinase in relation to vitiligo

Summary Tryptophan can inhibit DOPA (3,4-dihydroxy phenylalanine) conversion to melanin both by the enzymatic and the nonenzymatic route. The effects of brain monoamine depletion on p-chlorophenyl-alanine-induced hypothermia

Summary I.p. administration of p-chlorophenylalanine produced a fall in rectal temperature in rats. The hypothermia was attenuated after pretreatment of the animals with 5,6-dihydroxytryptamine, but was unaffected after pretreatment of the animals with 6-hydroxydopamine.This work was supported by grants from the National Science Council (Republic of China) and J. Amnesic effects of intravenous diazepam and lorazepam

Summary The amnesic effects of 2 benzodiazepine drugs, diazepam (Valium) and lorazepam (Ativan), have been investigated. Some of the effects were similar to those of certain clinical amnesic syndromes. The effects were more extensive than previous work has indicated.Acknowledgments. The authors are grateful to Dr J. HLA-DRw7 in psoriasis vulgaris

40 unrelated patients with Psoriasis vulgaris were studied for their HLA-A, B and DRw antigen phenotypes. All underwent a skin biopsy to confirm diagnosis. Like most of the authors, we observed an increase in B13 and B17 antigens (17,50 and 25%) whereas in the healthy population the percentage was (5 and 6,02%) respectively. The strong increase in DRw7 suggests that psoriasis vulgaris was associated with DRw7 antigens, with an unbalanced linkage between the B13 and DRw7 antigens and between the B17 and DRw7 antigens.     

On fragmentation and elimination of ovarian oocytes

Summary Examination of numerous fragmented oocytes in the ovary revealed an asynchronous relationship between nuclear and cell divisions. The asynchrony was observed in non-fragmented oocytes as well and was considered to be one of the common processes leading towards oocyte elimination in the ovary. The present study additionally demonstrates the morphology of follicle cell degeneration observed on the surface of fragmented oocytes.The authors are very grateful to Mr T. Horii, Dr K. Genetic control of malate dehydrogenase inNicotiana suaveolens andN. glutinosa

Summary The study of malate dehydrogenase patterns in leaves ofNicotiana suaveolens, N. glutinosa and their interspecific hybrid has been carried out, in order to propose a model of its genetic control. Two possible explanations for the genetic control of malate dehydrogenase have been postulated forN. suaveolens, and at least two loci appear to be implicated in the control of this system inN. glutinosa.Acknowledgments. Some new aspects of a sheep pineal gonadotropic inhibiting activity in in vitro experiments

Résumé Par des méthodes simples et bien déterminées: gelfiltration sur Sephadex G-25, G-10 et l'ultrafiltration par des diaflomembranes UM-02 et UM-05, on a pu isoler à partir d'un extrait aqueux d'épiphyses de mouton, des fractions capables d'inhiber l'activité gonadotrope hypophysaire. Cette activité inhibitrice a pu être localisée par l'électrophorèse sur papier. Les fractions ainsi obtenues sont capables d'inhiber in vitro l'excrétion hypophysaire de FSH et de LH.

Effects of the optical isomers of 2-amino-3-fluoro-1-phenylpropane (monofluoroamphetamine) hydrochloride on uptake and release of dopamine in rat striatum in vitro

Summary The (+)- and (–)-isomers of the monofluorinated amphetamine 2-amino-3-fluoro-1-phenylpropane were prepared and compared for their ability to stimulate release and to inhibit reuptake of radiolabelled dopamine from rat striatal tissue in vitro. A new rapid method for collection and preparation of cell suspensions for electrom microscopy

Summary We describe a new rapid and simple method for collection and preparation of cell suspensions for electron microscopy; the cells are prefixed with glutaraldehyde in their culture medium, and are then compacted on a filter disc. Post-fixation in osmium, staining and dehydration are performed by transferring the filter disc and the cell pellet from one solution to the next. The pellet is easily separated from the filter disc just before treatment in propylene oxide. This method preserves the fine structure as well as the classical technique. Advantages are that numerous cells have the same orientation in the sections and that many samples can be taken in a very short time.Acknowledgments. The authors are indebted to Prof. H. Intensification of amphetamine-induced excitation by methysergide,a serotonergic receptor blocker

Summary Methysergide, a serotonergic receptor blocker, was studied to determine its effects against d-amphetamine-induced excitation as measured by convulsions elicited by handling in mice. Significant intensification (p<0.01) of the action of d-amphetamine was observed in mice. These results indicate that reduction in serotonergic activity in the central nervous system enhances excitation induced by d-amphetamine.This work was supported by the Pharmaceutical Manufacturer Association Starter Grant, No. 5 S01 RR 05654-06 and the Air Force Office of Scientific Research Grant. Responses of skeletal muscle fibres to lanthanide ions. Dependence of the twitch response on ionic radii

Summary A study of the effects of the lanthanide series of ions on toad skeletal muscle fibres reveals that they a) inhibit the twitch response, b) exhibit a marked dependence on ionic radius, and c) apparently exert their effects on the sarcolemma.This research was supported by grants from the National Heart Foundation of Australia and from the National Health and Medical Research Council.
