In vitro development ofXenopus skin glands producing 5-hydroxytryptamine and caerulein

The granular glands of amphibian skin synthesize and store a large amount of bioactive amines and peptides which are structurally similar to mammalian brain-gut peptides. To investigate the development of peptide- and amine-producing cells in the granular glands, pieces of dorsal skin taken at various stages fromXenopus laevis tadpoles were cultured, and the contents of caerulein and 5-hydroxytryptamine (5-HT) were measured. When pieces of skin from tadpoles at stages 57 to 60 (Nieuwkoop and Faber stages) were cultured in a medium containing 10% fetal calf serum (FCS medium) or one containing FCS treated with charcoal (chFCS medium), the caerulein and 5-HT levels were increased for the six days of the incubation period. The caerulein content was lower in the chFCS medium than in the FCS medium. Addition of thyroxine to the chFCS medium had no significant effect on the caerulein content. These results show that the caerulein-and 5-HT-producing cells of the granular glands can develop in a culture system with FCS- or chFCS-containing media, and suggest that FCS contains substances which are absorbed by charcoal and stimulate development of the amine- and peptideproducing cells of the glands. In a preliminary search for correlation between caerulein and 5-HT synthesis, addition of 5-hydroxytryptophan (5-HTP), a precursor to 5-HT, to the FCS medium increased 5-HT content and, conversely, caused significant decrease in caerulein content, suggesting that accumulation of caerulein in the granular glands is influenced by the amount of 5-HT synthesis. These studies indicate that this culture system is a useful model for investigating the development of peptide- and amine-producing cells.     

Thein vitro inhibition of pentobarbital metabolism by chlorpromazine

Riassunto Si è osservato che il pentobarbital alla concentrazione di 2 × 10^–4 M può inibire per il 44% il metabolismoin vitro della cloropromazina alla medesima concentrazione.Si è discussa quindi la possibilità di inibire il metabolismoin vitro di alcuni farmaci propria di altre sostanze che vengono metabolizzate in maniera del tutto simile da sistemi enzimatici a livello microsomale epatico.     

Binding of 5-hydroxytryptamine and noradrenaline by rat lung

Résumé Après injection i.v. de 5-hydroxatryptamine-C¹⁴ et de noradrénaline-H³ à des rats, on a préparé et centrifugé des homogénates de leurs poumons dans un gradient linéaire de sucrose. Après sacrifice immédiat ces deux amines sont retrouvées dans une grande fraction de particules pulmonaires sédimentant dans 1M de sucrose. Après 30 min les poumons des rat offrent un pic additionel de noradrénaline-H³ correspondant à une densité de sucrose de 0.4 à 0.5.

Inhibition of brain enolases by acrylamide and its related compounds in vitro,and the structure-activity relationship

Summary Acrylamide and its related compounds inhibited brain enolases in vitro independently of their neurotoxicity. The inhibitory potency was a function of the binding constants of the compounds for phenylalanine. The binding constant for tryptophan was higher in neurotoxic compounds than in non-neurotoxic ones.

Human myeloperoxidase activity is inhibited in vitro by quercetin. Comparison with three related compounds

Quercetin is an effective inhibitor of human myeloperoxidase (MPO) activity, both with purified enzyme (IC50 = 3.5 microM) and in a system using stimulated human neutrophils. Quercetin is significantly more potent than three other related compounds (rutin, rutin sulfate and troxerutin) and than methimazole, a previously-known myeloperoxidase inhibitor. The inhibitory activity of quercetin is of the competitive type. Moreover, quercetin is directly able to scavenge hypochlorous acid (HOCl), a chlorinated species generated by the MPO/H2O2/Cl- system.

Prolongation of hypnosis by 5-hydroxytryptamine (Serotonin)

Zusammenfassung Die Angabe, dass 5-Oxytryptamin (Serotonin) die Chloralhydrat- oder Barbitursäure-Hypnose bei Mäusen verlängern kann, wurde bestätigt. Da aber Stoffe wie Adrenalin und Histamin den gleichen Effekt zeigten, erscheint es möglich, dass die hypnoseverlängernde Eigenschaft des 5-Oxytryptamin auf einen relativ unspezifischen, vaskulären Effekt zurückzuführen ist.     

5-hydroxytryptamine content of glioma

Résumé La teneur en 5-HT de tumeurs intra-crâniennes humaines, déterminée par une méthode biologique, est en général plus faible que celle du cortex cérébral adjacent. Cette observation n'est pas en faveur de l'hypothése que la 5-HT est localisée dans les cellules névroglies.     

Reserpine and human platelet 5-hydroxytryptamine

Zusammenfassung Nach einmaliger intramuskulärer Injektion von 1 mg Reserpin sinkt beim Menschen der 5-Oxytryptamingehalt der Blutplättchen zwei Tage lang ab, um bis zum neunten Tag auf unbestimmbarem Wert zu verharren. Während dieser Zeit ist die 5-OT-Aufnahmefähigkeit der Plättchen vermindert. Der Oxytryptamingehalt der Plättchen erreicht seinen Ausgangswert erst ungefähr 3 Wochen nach der Reserpingabe.Die Ansicht, dass die Plättchen freies 5-Oxytryptamin aus dem Kreislauf aufnehmen, wäre mit diesem Ergebnis zu vereinbaren. Die Dauer des Effektes könnte bedeuten, dass Reserpin entweder auf Plättchenvorstadien im Knochenmark wirkt oder aber dass die Lebensdauer der Plättchen in Wirklichkeit länger ist, als neuerdings angenommen wird.     

Occurrence of 5-hydroxytryptamine in chick retina

Summary 5-Hydroxytryptamine (5-HT) was found in chick retina. 5-HT level in chick retina was increased by the administration of pargyline and decreased by reserpine, but remained unchanged with tryptophan.     

Inhibition of insect nerve cord phosphorylase activity by 5-hydroxytryptamine

Zusammenfassung Die Phosphorylaseaktivität der in vitro mit 1×10^–4 M 5-Hydroxytryptamine inkubierten Insektennervenstränge war bedeutsam verringert. Der Effekt schien konzentrationsabhängig zu sein, da er sich mit steigender 5-HT-Konzentration verstärkte. 5-HT verminderte die aktive Form des Enzyms im Verhältnis zu dessen inaktiver Form. Epinephrin hatte keine Wirkung.     

Quantitative biological determination of 5-hydroxytryptamine

Zusammenfassung Die zwei meistgebrauchten Methoden der biologischen Auswertung von 5-Hydroxytryptamin, nämlich die am isolierten, atropinisierten Rattenuterus bzw. Rattencolon, wurden in bezug auf ihre Eignung für die quantitative Bestimmung verglichen. Für diesen Zweck bedienten wir uns der sogenannten 2 + 2-Dosen-Prozedur. Beide Methoden ergaben statistisch gleichwertige Resultate. Die Unregelmässigkeit der Uteruskontraktionen machen das Testieren öfters unmöglich, was beim Colon sehr selten der Fall ist. Ausserdem ist das Colon empfindlicher als der Uterus. Wegen dieser Vorteile ist das Colon für die quantitative biologische Auswertung von 5-Hydroxytryptamin dem Uterus vorzuziehen.     

Increase of free 5-hydroxytryptamine in blood plasma by reserpine and a benzoquinolizine derivative

Zusammenfassung Nach Vorbehandlung von Kaninchen mit einem Monoaminoxydase-Hemmer des Hydrazin-Typs wurde plättchenreiches Plasma mit Reserpin und einem Benzochinolizin-Derivat inkubiert. Im Plasma kam es dadurch zu einem Anstieg des nicht an Thrombozyten gebundenen 5HT, der dem Abfall des Amins in den Thrombozyten entsprach. Bei Kontrolltieren (ohne Monoaminoxydase-Hemmer) führte die Inkubation von plättchenreichem Plasma mit Reserpin zu keinem Anstieg des freien 5HT im Plasma, obgleich es in den Thrombozyten zu einer starken Senkung des Amins kam.
