Actinomycin-resistant antiviral activity associated with preparations containing chicken interferon.

Crude and purified preparations containing chicken interferon show a 2fold antiviral activity. One is inhibited by actinomycin and leads to total inhibition if interferon is added to cells before actinomycin, the other is insensitive to actinomycin and leads to partial inhibition if interferon is added simultaneously with or after actinomycin.     

Determination of acetaldehyde in human blood

Summary A method for the determination of acetaldehyde in human plasma by headspace gas chromatography is described. Chloralhydrate, an inhibitor of aldehyde dehydrogenase, is immediately added to the blood sample to prevent a rapid disappearance of acetaldehyde in the erythrocytes.This work was supported by the Swiss National Science Foundation.The authors thank Miss Ch. Ribosomal gene numbers in Microseridinae (Compositae: Cichorieae)

Summary 8 species of the subtribe Microseridinae contain between 1100 and 3400 genes for 25 and 18 S ribosomal RNA. The gene numbers seem to evolve by discrete steps. Their trend follows a general reduction in genome size during the evolution of the annual species ofMicroseris, but numbers remain high in one of them and inAgoseris grandiflora. 2 species ofPyrrhopappus differ by a duplication of the ribosomal gene numbers; 5 S ribosomal RNA genes in 4 species are repeated roughly 10,000 times.We thank Miss S. The lymphatic route. 1) Albumin and hyaluronidase modify the normal distribution of interferon in lymph and plasma

When human recombinant interferon-alpha 2 diluted in saline was injected s.c. into rabbits, the total amount recovered in thoracic lymph was less than 0.4%. Recoveries increased from 2- to 8-fold if interferon was injected in 4% albumin or with hyaluronidase, respectively. Albumin added to interferon acts as an interstitial fluid expander, thus favoring interferon absorption through lymphatics rather than blood capillaries. This strategy may increase the therapeutic index of interferon.     

Inhibition in the rat of gastric acid secretion and cyclic AMP analogs accumulation in vitro by somatostatin

Summary Various somatostatin (S) analogs exhibited similar degree and similar, or shorter, duration of inhibition of basal gastric acid secretion as S in the unanesthetized rat and similar, or less, inhibition of the cyclic AMP accumulation induced by prostaglandin E₂ in the rat anterior pituitary in vitro. With the analogs examined, the gastrointestinal and pituitary receptors appear to exhibit generally similar recognition specificity with the differences within the gastrointestinal activities reflecting duration of availability rather than receptor affinity.The author acknowledges the technical assistance of Mr L. Vitamin C and the immune response

Summary The inclusion of vitamin C in the drinking water of BALB/c mice was without effect on the humoral antibody response to sheep red blood cells and bacterial lipopolysaccharide. However, there was a significantly increased cell-mediated immune response as determined by increased T-lymphocyte responses to concanavalin A. This might suggest a mechanism, along with interferon enhancement, for the possible protection by vitamin C against some viral infections.We thank Miss Wendy Treat for excellent technical assistance.     

The effect of Valium® anaesthesia on the radiosensitivity of the skin of the mouse foot

Summary Valium^® anaesthesia significantly reduces the effect of large single doses of X-rays on the mouse foot skin as compared with Nembutal^® anaesthesia. The effect of cyclic AMP on dog renal function

Summary The infusion of dibutyryl-cyclic AMP into the dog renal artery in vivo leads to diuresis, natriuresis and glucosuria. Addition of the nucleotide to the incubation medium bathing dog renal cortex slices in vitro causes inhibition of p-amino-hippurate accumulation and stimulation of glycine and -methyl-glucoside transport. The results are interpreted in terms of the development of a blood-lumen flux of sodium and water in the renal proximal tubule, analogous to that seen in the intestine.We are grateful to Mrs P. Renal denervation delays blood pressure increase in the spontaneously hypertensive rat

Summary Bilateral surgical and chemical denervation of the kidneys in spontaneously hypertensive rats, performed 5 weeks after birth, delayed blood pressure increase by 2–3 weeks without affecting the rate of development or the final level of hypertension.Acknowledgments. Immunological studies on the key enzyme of arginine biosynthesis in Pseudomonas aeruginosa

Summary A method to manufacture specific antisera with a minute amount of pure enzyme is presented. The effect of actinomycin D on the production of viruses by the protoplasts of Chinese cabbage infected in vitro by the yellow mosaic virus of turnips]

Chinese Cabbage (Brassica sinensis L. var. Cantonner) protoplasts were infected by Turnip yellow mosaic virus (TYMV) and inoculated in the presence or absence of actinomycin D. Virus production was determined 40 hrs. after inoculation, the time required for the virus replication cycle to be terminated. While actinomycin D had no effect on TYMV production when present at a concentration of 1 microng/ml, a 50 to 80% inhibition of virus production was noticed at concentrations of the order of 5 to 10 microng/ml, and the inhibition reached 90% with 25 microng/ml.

Evidence against cyclic GMP acting as a direct modulator of active sodium absorption in rat cecum

Summary Cyclic GMP concentrations were measured in rat cecum mucosa in vivo when the net absorption of sodium, chloride and fluid was stimulated by methylprednisolone (MP). Whereas Na–K-ATPase specific activity was increased by MP, suggesting enhanced active sodium transport, cyclic GMP levels remained unaffected.Acknowledgments. We thank Miss U. Sensitivity of human lymphocytes to bleomycin increases with age

Summary The sensitivity of human peripheral blood lymphocytes to bleomycin and mitomycin-C was assessed by measuring the inhibition of phytohemagglutinin stimulated proliferation. The sensitivity to bleomycin, and not to mitomycin-C, increased with the age.We thank Miss L. Bailey for excellent technical assistance and the Bristol-Myers Company for supply of bleomycin. Pharmacological studies on a plant lectin,Aloctin A. I. Growth inhibition of mouse methylcholanthrene-induced fibrosarcoma (Meth A) in ascites form by Aloctin A

Summary A glycoprotein isolated fromAloe arborescens Mill markedly inhibited the growth of a syngeneic transplantable fibrosarcoma of mice, Meth A, in ascites form. There is evidence that the inhibition mechanism is host-mediated and not a direct toxic effect on the tumor cell.Acknowledgment. We thank Miss M. Dose-effects of 8-methoxypsoralen and long wave UV-light in 3T3 cells: evaluation of a phototoxic index

Summary 3T3 cells were cultured until confluency and treated with various doses of 8-methoxypsoralen followed by long wave UV light irradiation. The inhibition of³H-thymidine incorporation was dose-dependent for both, psoralen and light. A phototoxic index (PTI) was established demonstrating that a constant correlation between psoralen and UVA light exists for the photoinactivation in living cells.Supported by Deutsche Forschungsgemeinschaft.We thank Miss Anne Schröder for her skillful assistance.     

The effect of pinealectomy on serum progesterone and oestradiol levels during the last stage of pregnancy in rats

Summary Pinealectomy leads to significantly higher levels of progesterone (on days 19 and 20 of gestation) and oestradiol (on days 21 and 22) in the serum of pregnant rats. These results indicate that during the last phase of pregnancy the pineal gland may be modifying the levels of gonadal hormones, although not affecting the sequence of the physiological events.Acknowledgment. Inhibition of Ca2+-induced noradrenaline release from central noradrenergic neurons by morphine

Summary Morphine inhibited the noradrenaline release from slices of rat brain cortex induced by introduction of Ca²⁺ ions after superfusion with Ca²⁺-free, K⁺-rich solution.
