Summary It was found that some ciliates,Stentor, Spirostomum andBlepharisma, which can contract rapidly like the stalks of Vorticellidae, have Ca2+-binding proteins that are very similar to spasmins, in the immunological sense. The presence of spasmins in other Protozoa and in some Metazoa was also investigated.  相似文献

Protein-O-mannosyltransferases (Pmt proteins) catalyse the addition of mannose to serine or threonine residues of secretory proteins. This modification was described first for yeast and later for other fungi, mammals, insects and recently also for bacteria. O-mannosylation depends on specific isoforms of the three Pmt1, 2 and 4 subfamilies. In fungi, O-mannosylation determines the structure and integrity of cell walls, as well as cellular differentiation and virulence. O-mannosylation of specific secretory proteins of the human fungal pathogen Candida albicans and of the bacterial pathogen Mycobacterium tuberculosis contributes significantly to virulence. In mammals and insects, Pmt proteins are essential for cellular differentiation and development, while lack of Pmt activity causes Walker-Warburg syndrome (muscular dystrophy) in humans. The susceptibility of human cells to certain viruses may also depend on O-mannosyl chains. This review focuses on the various roles of Pmt proteins in cellular differentiation, development and virulence. Regulatory variation results from genetic changes with both cis and trans acting effects on gene expression. Here I describe the types of genetic variants that alter cis and trans regulation and discuss differences in the potential for cis and trans changes among different classes of genes. Summary In vivo microspectroscopy represents an effective and reliable technique to study pigment composition and distribution. In contrast to traditional extractive techniques, it preserves the integrity of biological specimens, without modifying the nature of the pigments. The spectroscopic apparatus we used is very simple and consists of a common microscope equipped with a monochromator, a photomultiplier, and two pinhole diaphragms. Absorption spectra obtained by means of this apparatus on different species of cyanobacteria are presented.  相似文献

Genetics of toxin production and resistance in phytopathogenic bacteria   总被引:2,自引:0,他引:2  
Genes for phytotoxin production have been identified and cloned from several phytopathogenic pseudomonads. These genes comprise physically linked clusters that have been located both on the chromosome and on endogenous plasmids. Contained within these genetic regions are resistance genes specific to those toxins that have a bactericidal component to their activity. DNA sequences required for toxin production are often conserved among bacteria with divergent host specificities, suggesting the ability of toxin genes to be transferred between bacteria. Toxins are usually modulators of plant pathogenicity, their production causing a significant increase in disease severity. In one case, however, toxin production appears to be a major contributor to the basic pathogenicity of a plant pathogenic bacterium.  相似文献

Summary The preferential destruction of chloroplast nucleoids in young zygotes in the coenocytic algaDictyosphaeria cavernosa and the giant unicellular algaAcetabularia calyculus was studied by high resolution epifluorescent microscopy. The proton-dependent synthesis of ATP was demonstrated in representative members of the generaHalobacterium, Haloarcula, andHaloferax. In all cases, synthesis was not inhibited by nitrate or N-ethylmaleimide, inhibitors of the vacuolar-like ATPase found in Archaea, but was affected by azide, an inhibitor of F0F1-ATP syntheses. These observations extend the earlier observations withHalobacterium saccharovorum and suggest that ATP synthesis in these organisms is brought about by an F0F1-APT synthase.  相似文献

Information concerning the specific nutritional requirements of malarial parasites developing in the mosquito host has been difficult to obtain, owing primarily to the complex nature of the blood meal that accompanies the parasites and the lack of success in culturing the complete invertebrate cycle ofPlasmodium in vitro. The present report describes a blood-free system for infecting mosquitoes with ookinetes ofPlasmodium berghei and for allowing the latter to develop into infective sporozoites. Ookinetes cultured in vitro were separated from blood proteins, suspended in defined medium, and fed toAnopheles stephensi mosquitoes through a membrane. The mosquitoes were then maintained on the same defined medium plus 5% sucrose. Infectivity of the parasites was demonstrated 17–19 days later by intracardial inoculation of the macerated mosquitoes into hamsters. This system makes it possible to evaluate nutritional factors that affect parasite development in the mosquito host under controlled conditions.This project was supported, in part, by the Public Health Service research grant AI-18345 from the National Institute of Allergy and Infectious Diseases to Prof. K. Summary The present report describes the changes in cyclic AMP level which occur upon parasitization of red cells byPlasmodium berghei. Parasitized erythrocytes were separated from the non-parasitized population by percoll density-gradient centrifugation. An increase in the cyclic AMP content of both non-parasitized and parasitized erythrocytes of infected animals compared with that of uninfected animals was observed. The patterns of physiological response to isoproterenol in normal, parasitized and non-parasitized erythrocytes were identical.  相似文献

Summary Using two species of caterpillars; larvae of the swallowtail butterfly, which produce large amounts of iso-butyric and 2-methylbutyric acids, and larvae of the puss moth, which produce no such detectable volatile compounds, it was shown that the insect which utilizes a chemical defense is better protected from its natural enemy, a praying mantis.  相似文献

Oviposition of four tortricid pests of vineyards (the European grapevine moth and the grape berry moth) and fruit orchards (the codling moth and the oriental fruit moth) is deterred by a blend of straight chain fatty acids and esters of fatty acids that have been identified in the eggs of one of them: the European grapevine moth (EGVM)Lobesia botrana. This is the first evidence of inter-specific recognition of an egg-like signal in moths. We demonstrate that oviposition site selection is influenced by population density, avoidance of deterrent being most important when females are isolated. Inter-specific egg recognition might be an important phenomenon, especially in species competing for a common food resource. We propose the term oviposition regulating synomone for molecules and blends that affect the inter-specific spacing of eggs.  相似文献

The metabolic relationship between the marine molluscHypselodoris orsini and the spongeCacospongia mollior has been reinvestigated. The predator-prey association has been confirmed even though the metabolic patterns of the two invertebrates are substantially different. Most probably the nudibranch converts the main sponge metabolite, the sesterterpenoid scalaradial (1), into a less oxygenated related metabolite, deoxoscalarin (4), followed by a second chemical transformation leading to a new sesterterpenoid, 6-keto-deoxoscalarin (5) which is selectively compartmentalized into some dorsal glands, mantle dermal formations (MDFs), strategically distributed near the gills. 6-keto-deoxoscalarin (5) has been characterized by 1D and 2D NMR methods. Finally, the unusual association of some Chromodorididae molluscs with sponges containing sesterterpenoids suggests a further analysis of their taxonomical collocation is required.  相似文献

Human bystin was identified as a cytoplasmic protein directly binding to trophinin, a cell adhesion molecule potentially involved in human embryo implantation. Although the trophinin gene is unique to mammals, the bystin gene (BYSL) is conserved across eukaryotes. Recent studies show that bystin plays a key role during the transition from silent trophectoderm to an active trophoblast upon trophinin-mediated cell adhesion. Bystin gene knockout and knockdown experiments demonstrate that bystin is essential for embryonic stem cell survival and trophectoderm development in the mouse. Furthermore, biochemical analysis of bystin in human cancer cells and mouse embryos indicates a function in ribosomal biogenesis, specifically in processing of 18S RNA in the 40S subunit. Strong evidence that BYSL is a target of c-MYC is consistent with a role for bystin in rapid protein synthesis, which is required for actively growing cells. Molecular determinants of antimalarial drug resistance are useful and informative tools that complement phenotypic assays for drug resistance. They also guide the design of strategies to circumvent such resistance once it has reached levels of clinical significance. Established resistance to arylaminoalcohols such as mefloquine and lumefantrine in SE Asia is mediated primarily by gene amplification of the P. falciparum drug transporter, pfmdr1. Single nucleotide polymorphisms in pfmdr1, whether assessed in field isolates or transfection experiments, are associated with changes in IC50 values (to arylaminoalcohols and chloroquine), but not of such magnitude as to influence clinical treatment outcomes. Recently described emerging in vitro resistance to artemisinins in certain areas correlates with mutations in the SERCA-like sequence PfATP6 and supports PfATP6 as a key target for artemisinins. The marine spongeTheonella swinhoei (lithistid Family Theonellidae, Order Astrophorida) has yielded many important, bioactive natural products, most of which share structural features with bacterial natural products. The presence of microbial symbionts inT. swinhoei has been reported, and it was originally suggested that the cytotoxic macrolide swinholide A and many of the bioactive cyclic peptides fromT. swinhoei were all produced by symbiotic cyanobacteria. By transmission electron microscopy, we found four distinct cell populations to be consistently present inT. swinhoei: eukaryotic sponge cells, unicellular heterotrophic bacteria, unicellular cyanobacteria and filamentous heterotrophic bacteria. Purification and chemical analyses of each cell type showed the macrolide swinholide A to be limited to the mixed population of unicellular heterotrophic bacteria, and an anti-fungal cyclic peptide occurred only in the filamentous heterotrophic bacteria. Contrary to prior speculation, no major metabolites were located in the cyanobacteria or sponge cells.  相似文献
