Further studies on gel filtration of urinary substances

Summary Urine specimens from healthy children were concentrated and filtered through Sephadex G-100 and G-25. The eluate was recovered in fractions of 5 ml each. Total hexoses, galactose, total proteins and total uronic acids were determined on each fraction. Total hexoses and galactose were eluted in 6 distinct peaks. The uronic acid containing GAG were eluted in 4 small peaks on G-25 and 1 peak on G-100. This peak is eluted always with high amounts of galactose containing glycoproteins.This work was partially supported by research grants from the CONICET, República Argentina.

Separation of serum proteins in different amphibian species by polyacrylamide gel electrophoresis

Zusammenfassung Mittels Polyacrylamid-Gel-Elektrophorese wurden die Serumproteine von 11 Amphibienarten in 15 bis 20 Fraktionen aufgetrennt. Das elektrophoretische Muster wurde mit demjenigen des menschlichen Serums verglichen und erwies sich als artspezifisch.

Further studies on bovine red cells having a different glycoprotein coat

Zusammenfassung Untersuchungen über den pronaselabilen Anteil der äusseren neuraminsäurehaltigen Glykoproteinschicht verschiedener Rindererythrozyten ergeben bei den einzelnen Tieren zwar signifikante Unterschiede, die jedoch in keiner direkten Beziehung zu den ebenfalls neuraminsäurehaltigen F-Antigenen dieser Zellen stehen.     

Cellular contaminants and structural proteins of Rous sarcoma virus (RSV), studied by polyacrylamide gel electrophoresis]

The number of polypeptides in highly purified preparations of RSV, of two different subgroups, produced in culture, has been compared to the polypeptides present in the supernatant of uninfected cultures and processed in identical manner. The analysis of PAGE-SDS shows that from 13 to 18 polypeptides present in viral preparations may be cellular contaminants. Fewer contaminating polypeptides are found in the myeloblastosis virus purified from plasma of Chicken.     

Two 2-dimensional electrophoreses: A combination of crossed immunoelectrophoresis and dodecyl sulfate polyacrylamide gel electrophoresis for protein analysis

Summary An improved method of two 2-dimensional electrophoreses that combined crossed immunoelectrophoresis and 2-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis was developed with a horizontal gel-electrophoretic apparatus.Acknowledgments. Free flow electrophoresis of isolated secretory granules from bovine neurohypophyses

Zusammenfassung Sekretorische Granula aus der Neurohypophyse von Rindern wurden durch Ultrzentrifugierung isoliert. Frei-Fluss Elektrophorese dieser Partikel zeigte, dass sie eine negative Ladung besitzen. Ca⁺⁺ und Na⁺ verminderten die Mobilität der Granula im elektrischen Feld.     

Acetylcholine content in brain white matter as determined after extraction with a new solvent mixture

Zusammenfassung Eine stark verbesserte Methode zur Extraktion und damit zur Bestimmung von Acetylcholin in der weissen Substanz von Ratten- und Meerschweinchengehirn, die bis zu 20–40% höhere Werte ergibt als die bisherigen Verfahren, wird beschrieben.     

Purification of solubilized fucosyltransferase from lamb brain using ethylagarose gel]

The glycoprotein: fucosyl-transferase of the cerebral hemispheres, assayed with desialylated fetuin as exogenous acceptor, was the most active of the protein: glycosyltransferases tested in the brain. The addition of Titron X-100 to the membrane suspension, followed by high speed centrifugation, led to a solubilization of the enzyme. The use of hydrophobic chromatography on ethylagarose gave a good purification of this solubilized fucosyl-transferase, whose homogeneity has been shown by Ultrogel AcA 22, DEAE-cellulose chromatography and disc electrophoresis.     

Fractionation of highly purified bovine growth hormone on sephadex G-25 gel

Zusammenfassung Hochgereinigtes Rinderwachstumshormon wurde durch Sephadex G-25 Gel-Chromatographie in zwei Komponenten zerlegt. Beide Fraktionen wie auch das unfraktionierte Präparat erwiesen sich in bezug auf Aminosäurenzusammensetzung, Elektrophorese und biologische Aktivität identisch.     

Disc electrophoresis of fractions of bovine central nervous system myelin

Zusammenfassung Fraktionen der weissen Hirnsubstanz, die offenbar zu den hauptsächlichsten Dichtelinien des Myelins respektive zur interperiodischen Linie gehören, wurden elektrophoretisch untersucht. Die Na-Fraktionen (interperiodische Linie; Membranseite zum extrazellulären Raum) enthalten meistens saure Proteine, während die CNS- und R-Fraktionen (Hauptlinie; Membranseite zum Zellinnern) zur Hauptsache alkalische Komponenten aufweisen.     

Butanol extracts from myelin fragments. III. SDS-urea gel electrophoretic studies

Summary The butanol extracts from myelin fragments showing 5-HT binding capacity were studied by SDS-urea gel electrophoresis. 5 main bands were observed and their molecular weights were determined by the method of Ferguson relationship, and revealed that these extracts contained proteolipid protein, DM-20 and basic proteins. Furthermore, the reconstituted fraction with crude basic proteins and lipids showed the saturable binding capacity for C¹⁴·5-HT.Acknowledgment. We thank Prof. T. A basic protein from bovine brain that co-precipitates with tubulin in vitro

A 193 kDa protein consisting of 58 kDa subunits, which has pI values of 8.50 and 8.65, was purified from bovine brain cytosol. It formed heavy precipitates with tubulin, and the molar ratio of tubulin dimer to this protein in the precipitate was 3.2. In contrast to microtubules containing ordinary microtubule-associated proteins, these complexes remained stable against cold and 1 mM CaCl₂.Acknowledgments.
