丝素包埋SPA成膜的理化性能和生物效应研究   总被引:1,自引:0,他引:1  
研究将丝素溶液包埋金黄色葡萄球菌A蛋白(staphylococcal protein A)(简称A蛋白或SPA ),制成不溶性SPA丝素膜.用粘附生长因子RGD(序列为GLY-ARG-GLY-ASP-SER-PRO-L YS)的抗体IgG结合IgG到该丝素膜的表面,再将RGD结合到不溶性SPA丝素膜表面RGD抗体IgG 上,制备成RGD-IgG-SPA丝素膜.用酶联免疫吸附试验(简称ELISA)方法,证明RGD-IgG -SPA丝素膜是稳定的.种植培养血管内皮细胞(vascular endothelial cell,简称EC细胞) 于该改性丝素膜的表面,用四甲基偶氮唑盐比色方法(简称MTT法)检测细胞的生长,证明改性丝素膜能有效地促进EC细胞的生长.  相似文献

肝素化丝素/聚氨酯共混膜的制备及其缓释性能   总被引:1,自引:0,他引:1  
以自制的非水溶性丝素粉体为载体,将肝素与医用聚氨酯同溶液混合,制备出含有肝素的丝素/聚氨酯共混膜,解决了肝素与聚氨酯同溶液共混成膜时团聚的问题,并利用丝素蛋白良好的吸附和缓释功能,使肝素从共混膜中缓慢释放出来,达到相对长效的抗凝血效果.利用ATR-FTIR分析了肝素加入后丝素粉体、丝素/聚氨酯共混膜的红外光谱的变化,并对共混膜中肝素的缓释性能及共混膜在PBs溶液中浸泡48h前后的形貌变化进行了研究.  相似文献

丝素/水性聚氨酯膜的制备及表征   总被引:1,自引:0,他引:1  
首次采用丝素粉体和水性聚氨酯按不同比例混合,经热压法成功制备出丝素/聚氨酯共混薄膜.红外光谱、X-射线衍射、动态力学分析及拉伸实验结果表明:聚氨酯的加入,促进了丝素蛋白分子β-折叠结构的形成,导致丝素结晶度提高;丝素含量越大,共混膜中聚氨酯的玻璃化温度越低,共混膜的拉伸断裂强度和初始模量越高;当丝素含量从0到70%变化时,试片的断裂强度由0.6 MPa增大到10.4 MPa,断裂伸长率从1065%下降到47.5%.聚氨酯的加入改善了共混膜的弹性,韧性随聚氨酯含量的增大而增大.  相似文献

采用一种自制的非水溶性丝素粉体与医用聚氨酯共混制膜.一方面,利用丝素蛋白良好的生物相容性来提高聚氨酯材料的生物相容性,弥补其用于小口径人造血管材料时生物相容性的不足;另一方面,以这种非水溶性丝素粉体为药物载体,将阿司匹林加入到聚氨酯膜中,来提高聚氨酯膜的抗炎症性能,开发新的小口径人造血管材料.以空白组为阴性对照组,PTFE材料为阳性对照组,采用体内埋植实验来评价新材料的组织相容性.结果表明,PTFE材料的组织相容性最差,丝素/聚氨酯共混膜材料的组织相容性良好.共混膜中加入阿司匹林后,其炎性反应减轻.  相似文献

采用溶胶-凝胶法制备了大豆分离蛋白/二氧化硅杂化溶胶,并应用于水性聚氨酯的改性.采用流延法制备大豆分离蛋白/二氧化硅/水性聚氨酯共混膜,红外光谱研究表明大豆分离蛋白、二氧化硅和水性聚氨酯间发生了多层次的氢键作用,以膜的拉伸强度、断裂伸长率和溶失率为指标,研究了各组分混配比例对共混膜力学性能及耐溶剂性能的影响.通过接触角、热重分析、扫描电镜和原子力显微镜分析讨论了共混膜的亲水亲油性、耐热性能,以及膜表面的形态形貌.结果表明,大豆分离蛋白/二氧化硅添加量为6%(质量分数)、两者之间质量比为1∶1时,改性膜的力学性能最优,耐溶剂性随添加量增加而改善.  相似文献

研究了蚕丝丝素水溶液经不同方式干燥后,所得到的再生丝素固体的微细结构,以及存放过程中丝素的结构变化,指出经30℃热风干燥或经高于-20℃冷冻干燥后,所得到的丝素固体的结构是无定形和Silk I共存,在室内温度湿度环境中存放后,部分无定形结构转化为Silk I;蚕丝丝素水溶液经-80--20℃冷冻干燥后,所得到的丝素膜的结构主要是无定形,含少量Silk II,存放过程中部分无定形结构转化为SilkI;室温放所自然形成的丝素凝胶中,丝素的结构是无定形和Silk Ⅱ共存;丝素水溶液表面膜中甘氨酸和丙氨酸的含量较高,表面膜的结构主要是Silk I和Tilk Ⅱ结晶度高。  相似文献

采用冷冻干燥法制备明胶与家蚕丝素的共混多孔蛋白膜,探讨了交联剂用量,共混比例及冷冻温度对膜结构和性能的影响,指出戊二醛对共混多膜具有明显的交联作用,膜内丝素与明胶的相容性较好,蛋白质的结构以无定形为主,通过与明胶共混,能够改善多孔丝素膜的拉伸性能。  相似文献

文章采用共沉淀法制备Fe3O4纳米粒子,分别用油酸钠、聚乙二醇对纳米粒子改性.超声下用乙醇分散改性纳米粒子,再与丝素蛋白溶液形成凝胶,制备出丝素蛋白/磁性纳米粒子复合材料.采用红外光谱仪、透射电镜、振动样品磁强计等对产物进行表征.结果表明,复合产物的铁氧特征吸收峰出现在583 cm-1处,透射电镜显示改性后的纳米粒子较均匀地分布到丝素蛋白溶液中,并表现出较好的磁性.油酸钠较聚乙二醇(PEG)改性Fe3O4纳米粒子的效果更好.  相似文献

不同相对分子质量的聚乙二醇(PEG)以不同比例混入丝素可形成共混丝素膜,并具有良好的拉伸性、透水、透;气、透光性和一定的生物降解与溶解性。实验结果表明:虽PEG和丝素的可混合性稍差,但在一定的透水、透汽性和较好的柔韧性前提下,可达到创面膜的基本要求。  相似文献

Surface-modified poly(butadiene)urethane (PBTU) films with silk fibroin (SF) were prepared by simple chemical method under the normal temperature. The physical properties and biological behaviour of the SF-modified PBTU film were evaluated. The results showed that the SF-modified PBTU films kept the tenacity and pliability very well, and could overcome rigid and brittle weaks of silk fibroin films. The morphology of SF in the PBTU film was dendritic aggregations, and the water-contact angle measurement indicated that the surface hydrophilicity of modified films was apparently enhanced. The biocompatibility of PBTU films was improved due to the change of surface components. The degree of platelet adhesion and the cell viability of rat embryo dermal fibroblasts seeded on PBTU films, SF films, and SF-modified PBTU films were measured by counting platelets before and after they contacted the films and MTT assay, respectively. The results indicated that platelet adhesion resistance and cell viability on the modified film were greatly superior to those on the PBTU film and the compound interface had good stability in the air.  相似文献

After removal of the caries or diseased teeth,the alveolar ridge will undergo absorption and atrophy.When the amount of alveolar bone is insufficient,it will cause an inability to perform effective dental implant restoration.In order to control the absorption and promote the repair and regeneration of alveolar ridge,a method of implanting guided bone regeneration(GBR) membranes at the extraction site is often used.In this study,silk fibroin(SF)and poly-L-lactide lactone(PLCL) were used to prepare bilayered guided bone regeneration membranes,and its morphology,hydrophilicity,surface roughness and mechanical properties were studied.At the same time,the drug release behaviors and cell compatibility of the bilayered membranes were studied.The results showed that SF/PLCL bi-layered membranes had good mechanical properties and surface hydrophilicity,and the drug-loaded bi-layered membranes had good cell compatibility.The bilayered membranes fabricated in this study are of potential for applying in the oral health field to promote bone regeneration.  相似文献

研究丝素蛋白及丝素蛋白/二氧化硅纳米杂化材料的两种药膜对天然抗菌药——茶多酚的缓释性能,并应用到聚丙烯无纺布的功能性后整理中.用体外释药方法测定了含茶多酚的两种药膜对茶多酚的缓释性能,并使用扫描电镜观察整理前后聚丙烯无纺布表面形态变化.结果表明,两种丝素蛋白膜都可以作为茶多酚的控制释放材料,丝素蛋白/二氧化硅纳米杂化药膜的控制释放作用优于丝素蛋白药膜.整理到聚丙烯无纺布上的上述两种药膜仍具有一定的缓释作用.  相似文献

In this study, the outstanding biocompatibility of silk fibroin (SF) and the highly efficient anti-bacterial effect of nano silver (NS) were utilized to prepare SF/NS composite film with anti- bacterial property. The structure and property of the film were characterized. The results showed that the structure of SF in the film was mainly silk I. SF in the film was almost insoluble in water. The tensile strength of film with NS was significantly lower than that of films without NS. When the addition of NS was within the range of 0%-0.6%, the elongation at break had no significant difference. The antibacterial rate of the film on staphylococcus aurens and escherichia coil increased with the amount of NS. The minimum amount of NS in the fdm was O. 1% and the maximum amount was 0.5%.  相似文献

Vitamin E( VE) is an ideal antioxidant and a stabilizing agent in biological membranes. In this study,silk fibroin( SF) /hydroxybutyl chitosan( HBC) nanofibrous scaffolds are loaded with VE tocopherol polyethylene glycol 1000 succinate( VE TPGS) via electrospinning. SEM images show that the average nanofibrous diameter has no significant difference when the content of VE TPGS increases to 4. 0%( SF / HBC). However,the average nanofibrous diameter decreases largely to 200 nm when the VE TPGS content reaches 6. 0%. Furthermore,VE TPGS presents a sustained release behavior from the nanofibrous scaffolds. Cell viability studies of mouse skin fibroblasts( L929) demonstrate that VE TPGS loaded SF / HBC nanofibrous scaffolds present good cellular compatibility.Moreover,the incorporation of VE TPGS could strengthen the ability of SF / HBC nanofibrous scaffolds on protecting the cells against oxidation stress using the Tertbutyl hydroperoxide( t-BHP)-induced oxidative injury model. Therefore,VE TPGS-loaded SF /HBC nanofibrous scaffolds might be potential candidates for personal skin care,wound dressing and skin tissue engineering scaffolds.  相似文献

To improve the toughness of silk fibroin( SF) films,poly( ethylene glycol-glycerin)( PEGG) was synthesized with ethylene glycol and epichlorohydrin. The SF / PEGG blend films were prepared by casting aqueous solution and their structures were characterized. The PEGG was in liquid state at room temperature so it will not be a single phrase at blend film. It crosslinked with SF and made it insolubility in water. The results of X-ray diffraction( XRD) indicated that the crystallinity of the SF in the blend films decreased with the content of PEGG increasing. The tensile strength and elongation at break of blend films were measured using an instron tensile tester. The results showed that the tensile strength and elongation at break of blend films were high enough for application.After the blend films were stored at room temperature for 100 d,the crystallinity, the tensile strength and elongation at wet state increased. The blend films are superior to SF films in providing excellent flexibility and mechanical properties in both dry and wet states. Based on the fact that SF has good biocompatibility,the SF /PEGG blend film will offer new options in many different biomedical applications.  相似文献

采用2,2-二羟甲基丙酸(DMPA)和N-甲基二乙醇胺(MEDA)分别作为阴、阳离子型扩链剂,在聚氨酯(PU)预聚体结构上先后引入两种离子型的亲水基团,再将制得的聚氨酯预聚体与丝素蛋白(SF)水溶液混合交联,得到SF/PU两性水凝胶.通过衰减全反射(ATR)、扫描电子显微镜(SEM)表征其结构、微观形貌及多孔结构.结果表明:制备出的SF/PU两性水凝胶具有明显的孔洞结构,孔径分布在15μm左右.通过测试在不同条件下的吸水溶胀比(SR)和平衡溶胀含水量(EWC),分析了两性水凝胶的溶胀动力学机理.通过万能实验拉力机测试了两性水凝胶的机械性能,当SF/PU比例为65/35时,水凝胶材料的压缩强度达到最大值,为0.577 MPa;当SF/PU为50/50时水凝胶有着最大变形率,为85.8%.  相似文献

应用二种中性盐溴化红和氯化钙作为溶解蚕丝丝素的试剂,制成了二种固定化葡萄糖氧化酶丝素膜。经酶比色法和红外分光光度法分析,结果表明这二种丝素膜都是良好的固定化酶的生物材料。葡萄糖氧化酶经这些丝素膜固定以后对热和pH稳定性得到明显改良。这些酶膜性能稳定,具有高的活性得率,能长期存放。以这些固定化葡萄糖氧化酶丝素膜和氧电极为基础,研制的流动注射分析式电流型葡萄糖生物传感器性能较稳定,具有较宽的葡萄糖线性响应范围(0.5~15.0mmol/L),相关系数为0.999,可以反复测定葡萄糖一千次以上。  相似文献
