抗蛋氨酸结构类似物突变株的筛选

以北京棒杆菌AS1.299经诱变后获得的高产蛋氨酸突变株N3-10作为出发菌株, 依次采用蛋氨酸结构类似物亮氨酸、 原亮氨酸和乙硫氨酸平板对
诱变后的突变株进行筛选, 得到蛋氨酸高产菌株依次为L3,LN7和E31. 结果表明, 亮氨酸对突变株N3-10的抑制质量浓度为8 g/L, 原亮氨酸对突变株L3的抑制质量浓度为3 g/L, 乙硫氨酸对LN7突变株的抑制质量浓度为3 g/L, 经紫外诱变和3种蛋氨酸结构类似物的筛选, 最终获得蛋氨酸产量最高突变株E31, 产量为1.479 g/L, 比出发菌株N3 10蛋氨酸产量高0.379 g/L. 经5次传代, 产量稳定.     

沼泽红假单胞菌高产辅酶Q10菌株的诱变选育研究

以沼泽红假单胞菌为出发菌株,通过紫外诱变和罗红霉素抗性筛选,以期获得辅酶Q10高产菌株.试验结果表明,紫外照射时间60-70s时可能达到最大正向突变概率.通过紫外诱变、罗红霉素初筛和发酵复筛,获得一株沼泽红假单胞菌突变株R-1,辅酶Q10产量达到0.021g/L,较出发菌株产量（0.018g/L）提高16％.经过6次传代培养,突变菌株产辅酶Q10产量稳定,可用于进一步研究.     

克拉维酸高产菌的选育

在克拉维酸高产菌的选育过程中,应用解除终产物结构类似物作为选择压力,能有效地除去低效价菌株,提高高产菌株的检出率,较传统筛选法优越.试验表明,以带棒链霉菌Strep tomycesclavuligerusCCRC11518(效价为382.4×10-6g/mL)为出发菌,经紫外线诱变,筛选到一株抗20×10-3g/mL的舒巴坦钠突变株my51,my51菌株产克拉维酸的效价达到834.7×10-6g/mL,比出发菌提高了1.18倍.该菌株连续传代7代,克拉维酸的效价保持稳定.     

鸟苷产生菌解淀粉芽孢杆菌的选育

出发菌GR001通过形态观察、生理生化实验、16S rDNA同源性序列分析及部分特异性基因序列分析鉴定为解淀粉芽孢杆菌.以GR001为出发菌,通过化学诱变选育出一株遗传标记为腺嘌呤营养缺陷、6–巯基嘌呤抗性以及蛋氨酸亚砜抗性(Ade-+MSOr+6-MPr)的突变株GR600.以GR600为受体菌,采用原生质体转化法获得转化子GR607(Ade-+MSOr+6-MPr+kmr),鸟苷产量高达20.82,g/L.     

动物源大肠杆菌染色体ampC基因突变与头孢菌素耐药关系研究

为检测动物源性大肠杆菌染色体ampC基因突变与头孢菌素类药物耐药性的关系,选取22株头孢菌素类药物耐药菌作为受试菌株,采用K-B纸片法测定了其对7种头孢菌素的敏感性,采用PCR测序的方法进行受试菌染色体ampC基因调控区和部分编码区碱基突变分析.结果表明:受试菌对7种头孢菌素的耐药率均在50%以上,20株受试菌ampC基因调控区和部分编码区碱基发生了突变,占90.9%;共有10种突变组合,突变频率最高的为+185,+58,+122,+126,-88,-82,-18,-1位.ampC基因调控区和部分编码区碱基突变与β-内酰胺类药物耐药性有关.     

雨生红球藻对紫外光处理的响应及高产藻株的选育

用紫外线连续两代诱变雨生红球藻出发株HP-IS,经筛选获得两个突变品种系HP-M1-1和HP-M3-1,与出发株相比,两个突变株的营养细胞个体较大,周质空间较宽,形成的大孢子较多。两个突变株生长速率较出发株的提高8.2%～11.8%。是青素累积能力明显增强,培养25天时的是青素累积量分别为35.28mg/L和38.05mg/L,比出发株的高75.5%～89.3%。     

2型猪链球菌RevS基因敲除突变株生物学特征及致病性研究

比较2型猪链球菌(S.suis 2)ΔRevS突变株和05ZY/H33野生株生物学特性及致病性的差异.首先,通过吸光值测定和革兰染色比较△RevS突变株和05ZY/H33野生株的生长速度和菌体形态.分别对Balb/c及ICR(CD1)小鼠腹腔注射109 CFU·mL~(-1)的05ZY/H33菌液,评估两种品系小鼠对S.suis 2的易感性.分别对Balb/c小鼠腹腔注射2.5×10~9,5.0×10~8,1.0×108,2.0×10~7,4.0×10~6 CFU·mL~(-1)的05ZY/H33菌液,采用Reed法计算半数致死量.对Balb/c小鼠腹腔注射5.0×108 CFU·mL~(-1)的ΔRevS突变株和05ZY/H33野生株菌液,观察野生株和突变株对小鼠的影响.对仔猪耳缘静脉注射10~9 CFU·mL~(-1)的05ZY/H33野生株和ΔRevS突变株菌液,观察野生株和突变株对仔猪的影响.体外生物学特征分析表明,与野生株相比,突变株的生长速率和形态染色并未发现显著变化;Balb/c小鼠对S.suis 2 05ZY/H33的敏感性高于ICR(CD1)小鼠,05ZY/H33对Balb/c小鼠的LD50为4.2×10~7 CFU·mL~(-1).Balb/c小鼠攻毒试验结果显示,ΔRevS对小鼠的致病性明显减弱.仔猪攻毒试验结果显示,野生株和突变株都引发仔猪全部死亡,但ΔRevS引发仔猪死亡时间较晚.△RevS突变株保持了菌株的基本生物学特征,但对小鼠和仔猪两种动物模型的致病作用差异较大.     

酿酒酵母的连续复合诱变及其突变株的快速筛选

为了获得高产的工业用酿酒酵母(Saccharomyces cerevisiae)菌株,利用己烯雌酚(DES)和UV对酿酒酵母菌QD进行连续复合诱变,采用改进的发酵小管集气法结合乙醇定量测定来进行突变株的筛选,并与常规复合诱变及筛选方法进行比较.结果表明:连续复合诱变的正向突变率最大值为18.9%,与常规诱变方法的最大正向突变率15.6%相比提高了21.2%,说明运用该诱变方法能获得较高的正向突变率;改进的筛选方法可以快速筛选出目的突变株,同时,获得了一株高产乙醇的突变株,在甘蔗汁发酵培养基中其乙醇产量达到9 720 mg/100 mL,比常规诱变方法筛选到的突变株乙醇产量(9 170 mg/100 mL)高出6.0%;与常规诱变方法相比,连续复合诱变方法不仅能够得到较高的正向突变率,而且能够获得更加高产的突变株.     

γ-亚麻酸产生菌的低能离子束诱变选育

为了得到γ-亚麻酸(GLA)高产菌株,以深黄被孢霉As3.3410为出发菌株,利用氮离子(N+)注入诱变,并采用马来酰肼抗性筛选和红四氮唑(TTC)法相结合对突变株进行筛选。试验结果表明:当能量为10 ke V时,最佳氮离子诱变剂量为1.56×1015cm-2;当马来酰肼的添加量为40 mg/L时,出发菌株的抑制率为100%;TTC法酶活测定的最佳条件为:温度35℃、p H为8.4、TTC浓度为8 g/L、反应时间为1 h,以此作为摇瓶初筛的条件。经过反复诱变筛选获得一株遗传稳定的高产突变株F312,其γ-亚麻酸产量为1 236 mg/L,比出发菌株(480 mg/L)提高了157.5%。     

GSH产生菌的选育

对酵母菌(Saccharomyces)2.558进行紫外照射、紫外照射 LiCl复合处理获得了1株突变株F400-1,GSH细胞含量19.25mg/g,比出发菌株提高了535.38%;发酵液含量达到106.14mg/L,比出发菌株提高了745.03%。经遗传稳定性分析,证明菌株F400-1是遗传稳定的。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
