基于网络药理学探讨连翘脂素减肥靶标及作用机制

文章利用网络药理学研究连翘脂素潜在减肥靶标及作用机制。首先通过PharmMapper筛选连翘脂素相关靶标，GnenCards筛选肥胖相关靶标，绘制venn图，共筛选到219个连翘脂素减肥相关靶标。其次通过String平台和Cytoscape 3.6.0构建靶标间相互作用网络图、进行可视化分析并筛选核心靶标，包括ALB、AKT1、MAPK1、EGFR、HRAS、CASP3、SRC、IGF-1、MAPK8、ESR1，其它靶标还包括JAK2、CDC42、NQO2、DHFR等。然后利用DAVID6.8进行GO功能富集分析和KEGG通路分析，GO功能富集涉及811个条目，KEGG通路分析得到111条信号通路，主要包括癌症通路、胰岛素信号通路、MAPK信号通路、ErbB信号通路、黏附斑等。最后利用分子模拟技术对连翘脂素减肥相关靶标进行验证，证明连翘脂素与ALB、AKT1、EGFR、HRAS、CASP3、SRC、IGF-1、MAPK8等都有较好的结合活性（结合能小于-5 kcal/mol）。研究通过对成分-靶标-疾病网络分析，从多靶标、多通路阐释了连翘脂素潜在减肥作用机制，为后续深入研究提供了理论依...     

基于网络药理学研究连翘抗炎作用机制

文章基于网络药理学研究连翘抗炎作用机制。通过TCMSP数据库收集连翘化学成分，并以OB≥30%,DL≥0.18筛选连翘化学成分。采用PharmMapper数据库预测连翘作用靶点。以“inflammation”为关键词使用OMIM数据库和TTD数据库检索炎症相关靶点，并用Cytoscape 3.8.1软件构建“活性成分-靶点”网络。通过String数据库进行蛋白质相互作用分析，并使用Cytoscape 3.8.1软件构建蛋白相互作用网络。最后使用DAVID数据库对连翘抗炎作用靶点进行通路富集分析，以探究连翘抗炎的作用机制。结果为经筛选得到良好的连翘活性成分21个，预测到45个潜在的抗炎靶点。其中连翘抗炎的主要靶点为丝裂原激活蛋白激酶14(MAPK14)、 SRC蛋白激酶（SRC）、胰岛素样生长因子1(IGF1)、丝裂原激活蛋白激酶1(MAPK1)及表皮生长因子受体（EGFR）等核心靶点。经GO分析和KEGG通路分析，连翘参与的抗炎信号通路主要有癌症通路、催乳激素信号通路、结核病、破骨细胞分化、丙型肝炎、PI3K-Akt信号通路等。本研究得连翘的抗炎作用潜在机制可能是槲皮素，连翘苷，木犀草...     

老翘HPLC特征图谱研究

通过对12批老翘药材进行研究,建立老翘的HPLC特征图谱。采用HPLC法测定,对12批老翘进行分析,以连翘酯苷A为参照峰建立了老翘的特征图谱,并指认了老翘中5个特征成分:梾木苷甲基醚、芦丁、连翘酯苷A、连翘苷和连翘脂素,各特征峰相对保留时间均在规定值的±5%之内。采用“中药色谱指纹图谱评价系统”进行相似度分析,12批老翘特征图谱相似度均达到0.900以上。本文所建立的特征图谱方法简便准确,重复性好,可用于老翘的质量控制。     

市售连翘饮片质量评价研究

对市售连翘饮片进行质量评价,分析连翘质量差异的原因。按《中国药典》2015版的相关测定方法,测定60批连翘中水分、总灰分、醇溶性浸出物、连翘苷和连翘酯苷A的含量,对测定结果进行数理统计和聚类分析,评价连翘饮片的质量差异。市售连翘饮片不合格率为5.0%,水分、总灰分、浸出物、连翘苷、连翘酯苷A测定结果符合正态分布,在95%的置信度下可接受的取值范围分别为(4.86%～4.98%)、(2.88%～3.05%)、(33.37%～36.31%)、(0.41%～0.50%)、(6.19%～7.78%);建立的PLS-DA模型可靠,预测能力良好,浸出物、连翘酯苷A、连翘苷是影响连翘质量的主要指标。本研究客观、有效地评价了市售连翘饮片的质量。     

高速公路护坡植被水保效应研究

公路建设引出了很多环境问题，如何进行边坡的防护是迫切需要解决的问题。本文主要就高速公路护坡植物水保效应进行浅要的分析，以期对相关工程建谩有借鉴意义。     

不同连翘叶炮制品抑制胰脂肪酶活性比较

为了比较不同连翘叶炮制品抑制胰脂肪酶活性,首先采集连翘叶并分成四等份分别进行自然阴干、蒸后阴干、煮后阴干、烘干处理;其次,对四种炮制品进行40%乙醇回流提取,并测定其粗提物抑制胰脂肪酶活性;第三,经HPLC检测、混合标品比对、绘制标准曲线,对各炮制品粗提物进行定性和定量分析,并测定各主要成分对胰脂肪酶的半抑制率;第四,进行相关性分析,推测连翘叶炮制品中影响抑酶活性的主要成分;最后,综合上述实验数据,浅析不同连翘叶炮制品抑酶活性差异原因。结果表明:影响连翘叶抑制胰脂肪酶活性的炮制方式依次为烘干煮后阴干蒸后阴干自然阴干;各炮制品主要成分为咖啡酸、连翘酯苷A、芦丁、橙皮苷、山柰酚-3-O-芸香糖苷、连翘苷和连翘脂素;各成分对胰脂肪酶抑制作用为山柰酚-3-O-芸香糖苷橙皮苷芦丁咖啡酸连翘酯苷A,连翘苷为促进作用,连翘脂素几乎无作用;相关性分析表明,抑酶活性最主要影响成分为咖啡酸、芦丁和连翘苷,其中咖啡酸和芦丁与粗提物抑酶活性呈正相关,而连翘苷为负相关。     

山西连翘子药材质量标准研究

为了建立山西省连翘子地方药材质量标准,文章采用薄层色谱法对连翘子药材进行定性鉴别,气相色谱法建立连翘子挥发性成分的指纹图谱,并用HPLC方法检测其中连翘酯苷A的含量。结果显示薄层色谱检出连翘酯苷A,特征斑点分离清晰;对10个产地的连翘子挥发性成分进行分析,建立了连翘子挥发性成分的气相色谱指纹图谱,相似度均在0.994以上;HPLC分析表明连翘酯苷A在0.188 0μg~2.256 0μg呈良好的线性关系,相关系数为1.000 0,平均回收率为98.72%,RSD为1.6%,10个产地的连翘酯苷A百分含量在2.331%~3.845%之间。     

连翘叶绿茶制备及活性成分分析

为了进一步提升连翘叶的开发，将一芽二叶至一芽三叶的连翘嫩叶加工成连翘叶绿茶。对连翘叶绿茶、桑叶绿茶和信阳毛尖的活性成分进行分析。分析结果表明：连翘叶绿茶含水量为36 mg ／g。微量元素含量较高，茶多酚为172 mg ／g，氨基酸为13．23 mg ／g，咖啡碱29．3 mg ／g，总黄酮26．48 mg ／g，总木脂素19．76 mg ／g，以及总三萜酸6．15 mg ／g。具有较强的清除 DPPH 自由基的能力，这表明连翘叶绿茶含有丰富的对人体具有保健作用的活性成分。     

贯叶连翘的研究进展与可持续发展对策

贯叶连翘除作为传统的中药材入药外，它的活性成分在治疗抑郁症，肝炎和艾滋病以及抗癌等方面具有重要的利用前景而被广泛关注。本文综述了贯叶连翘的生药显微鉴定特征，有效成分的研究，药理和临床试验等方面的研究进展。同时探讨了可持续发展的对策。     

中药材连翘道地性的分子物学探讨

运用随机扩增多态DNA（RAPD）方法研究了来自山西黎城、长治、平顺、壶关、屯留及安泽6个地区11个连翘地方栽培品系的亲缘关系，并用HPLC方法测定连翘主要指标成分-连翘甙含量的变异情况，RAPD分析结果表明，可分为A、B、C3个不同分支，连翘甙含量分析结果显示，2个遗传关系相近分支B和C的连翘甙含量差异不显著（t=0.44,p=0.67；平均连翘甙含量分别为0.53mg/g和0.67mg/g），这2个类群与第3个类群遗传差异大，连翘甙含量差异也很大（8.7mg/g），这种DNA分子多样性差异与化学指标成分含量差异的相关性说明连翘的遗传基础可能对化学指标成分的形成和积累有显著的影响，研究结果显示运用RAPD分析手段进行药材道地性研究具有可行性，研究结果不仅对于连翘的道地性研究和中药材GAP（Good Agricultural Practice)生产有一定的指导意义，而且对于其他道地中药材的研究也具有一定参考价值。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
