Female Commentry Rats 80--85 days old, were immunized by 5 intramuscular injections of Mouse alphafetoprotein (AFP) and then mated. After fertilization a supplementary injections was administered. The animals were bled at different times and killed immediately after the last bleeding on the 19 or 20th day of gestation. Titers of AFP and of autologous anti-AFP antibodies in the maternal blood were determined as well as the AFP concentration in the pooled amniotic fluids from live embryos of each litter. Compared to non-immunized control series, the total of live and dead embryos per litter in animals immunized with Mouse AFP showed no difference. However, the number of live embryos was on the average 50% lower than that in the control series. The serum titers of AFP and of antibodies to autologous AFP in immunized pregnant Rats bearing dead embryos decreased concomitantly with the number of live embryos. The results reported herein demonstrate that the presence of anti-autologous AFP antibodies in pregnant Rats correlates with the interruption of development in a significant proportions of embryos. This suggests that certain spontaneous abortions in the Rat and perhaps in other mammals can be explained by the rupture of immunological tolerance to autologous AFP.  相似文献

Quantitative data are presented on the fatty acid composition of rat alpha-fetoprotein (AFP) and serum albumin, (SA), and of brain extracts of suckling rats. In AFP and SA preparations, 40% and 13%, respectively, of total fatty acids present are polyenoic acids. Among them, docosahexaenoic acid is quantitatively the most important in AFP, while in SA, arachidonic acid is largely predominant. Both docosahexaenoic and arachidonic acids were the predominant polyenoic acids in brain extracts. The rate of accumulation of these acids in the brain of suckling rats and the rate of AFP secretion during the same period showed a maximum around 10--12 days after birth. These results suggest that AFP and SA play an important role in the transport and the incorporation of polyunsaturated fatty acids in the developing brain.  相似文献

Fluid obtained from rat fetuses was utilized to characterize the affinity, number of binding sites, and the association and dissociation rate kinetics of the binding of estradiol and estrone to AFP. Statistical analysis demonstrated no differences when the values for the AFP-estradiol interaction were compared with those obtained for the ATP-estrone interaction. The morphological localisation of alphafetoprotein (AFP), serumalbumin (SA), transferrin and immunoglobulins (IgG) has been studied in the developing central nervous system of the Rat by immunocytochemical methods. Evidence is presented of a highly selective staining for AFP and SA, both proteins exhibiting the same topographical distribution. Practically all the areas of the brain and the spinal cord are stained at a given moment of the developmental process. The labeling is cytoplasmic and in the neuronal elements extends to their axonic and dendritic prolongations. The localization of AFP and SA in the nervous system may be related to the well known binding properties of these proteins for varied substances (estrogen and/or fatty acids). The morphological data presented here suggest that both proteins may be actively involved in the uptake of such substances by the cellular structures of the nervous tissue.  相似文献

目的调查分析我院20ff6~2010年临床分离的革兰阴性菌对碳青霉烯类抗生素的耐药性。方法采用纸片扩散法进行抗菌药物敏感实验,采用WHONET5.4软件及SPSS13.0软件进行数据分析。结果大肠埃希菌和肺炎克雷伯菌对亚胺培南和美罗培南的耐药率呈上升趋势,但最高不超过4.5%。铜绿假单胞菌对亚胺培南的耐药率2006年和2007年监测显示为21.3%和41%。2008年达到44.8%,在2009年下降为12.4%,但2010年爽迅速增加到30.1%。鲍曼不动杆菌对亚胺培南耐药率的逐年上升更为显著,从2006年9.1%上升到2010年的68%,差异具有统计学意义(P〈0.01)。嗜麦芽窄食单胞菌对碳青霉烯类抗生素天然耐药,2006年对亚胺培南耐药率为30%,但自2007年起就增加到80.8%,最高达100%。结论肠杆菌科细菌对碳青霉烯类敏感性高,近年耐药率有上升趋势。铜绿假单胞菌和鲍曼不动杆菌对碳青霉烯类耐药率高,尤其是鲍曼不动杆菌的分离率逐年增加,耐药形势严峻。临床应合理使用抗生素,减少多重耐药株的产生。  相似文献

The aim of this study is twofold: to explore, first, the influence of the intellectual and social conditions on the transfer of thermodynamics to chemistry and thereby the making of chemical thermodynamics, and second, the way that this knowledge was transferred from Europe to America. Consequently, it is of interest to examine the methodological approaches used by physicists and chemists to transfer thermodynamics to chemistry, to evaluate the potential of this science to offer solutions to existing chemical problems, and to discuss the attitude of the scientific community towards these new ideas. The development of chemical thermodynamics in America followed a different route compared to the European experience. Although it was transferred from Europe, it had distinctive characteristics imposed by a different traditional, intellectual and social milieu. This study focuses on the content of the transferred knowledge to America and the direction that this knowledge assumed by the American scientists. As a paradigm, the chemical thermodynamics of Gilbert Newton Lewis will be considered.  相似文献

The first function attributed to lactoferrin (Lf), an iron binding protein belonging to the non-immune natural defences, was antimicrobial activity that depended on its capacity to sequester iron. Iron-independent microbicidal activities, requiring direct interaction between this cationic protein and microbial surface components, were later demonstrated. Many other anti-microbial and anti-viral functions have since been ascribed to Lf. In mucosal secretions, iron and Lf modulate the motility and aggregation of pathogenic bacteria. Lf inhibits bacterial adhesion on abiotic surfaces through ionic binding to biomaterials, or specific binding to bacterial structures or both. Lf inhibition of bacterial adhesion to host cells requires Lf binding to bacteria and/or host cells. Lf hinders microbial internalization by binding to both glycosaminoglycans and bacterial proteins which can be degraded by Lf-mediated proteolysis. Moreover, Lf internalisation and localisation to the host cell nuclei could modulate bacterial entry into cells through gene regulation. Finally, the capability of Lf to exert antiviral activity, through its binding to host cells and/or viral particles, strengthens the idea that it is an important brick in the mucosal wall, effective against both microbial and viral attacks.  相似文献

Developmental biology investigations have evolved from static studies of embryo anatomy and into dynamic studies of the genetic and cellular mechanisms responsible for shaping the embryo anatomy. With the advancement of fluorescent protein fusions, the ability to visualize and comprehend how thousands to millions of cells interact with one another to form tissues and organs in three dimensions (xyz) over time (t) is just beginning to be realized and exploited. In this review, we explore recent advances utilizing confocal and multi-photon time-lapse microscopy to capture gene expression, cell behavior, and embryo development. From choosing the appropriate fluorophore, to labeling strategy, to experimental set-up, and data pipeline handling, this review covers the various aspects related to acquiring and analyzing multi-dimensional data sets. These innovative techniques in multi-dimensional imaging and analysis can be applied across a number of fields in time and space including protein dynamics to cell biology to morphogenesis.  相似文献

Climate change is identified as a major threat to wetlands. Altered hydrology and rising temperature can change the biogeochemistry and function of a wetland to the degree that some important services might be turned into disservices. This means that they will, for example, no longer provide a water purification service and adversely they may start to decompose and release nutrients to the surface water. Moreover, a higher rate of decomposition than primary production (photosynthesis) may lead to a shift of their function from being a sink of carbon to a source. This review paper assesses the potential response of natural wetlands (peatlands) and constructed wetlands to climate change in terms of gas emission and nutrients release. In addition, the impact of key climatic factors such as temperature and water availability on wetlands has been reviewed. The authors identified the methodological gaps and weaknesses in the literature and then introduced a new framework for conducting a comprehensive mesocosm experiment to address the existing gaps in literature to support future climate change research on wetland ecosystems. In the future, higher temperatures resulting in drought might shift the role of both constructed wetland and peatland from a sink to a source of carbon. However, higher temperatures accompanied by more precipitation can promote photosynthesis to a degree that might exceed the respiration and maintain the carbon sink role of the wetland. There might be a critical water level at which the wetland can preserve most of its services. In order to find that level, a study of the key factors of climate change and their interactions using an appropriate experimental method is necessary. Some contradictory results of past experiments can be associated with different methodologies, designs, time periods, climates, and natural variability. Hence a long-term simulation of climate change for wetlands according to the proposed framework is recommended. This framework provides relatively more accurate and realistic simulations, valid comparative results, comprehensive understanding and supports coordination between researchers. This can help to find a sustainable management strategy for wetlands to be resilient to climate change.  相似文献

The technique of selective breeding has been employed to develop a number of mouse lines differing in genetic sensitivity to specific effects of ethanol. Genetic animal models for sensitivity to the hypnotic, thermoregulatory, excitatory, and dependence-producing effects of alcohol have been developed. These genetic animal models have been utilized in numerous studies to assess the bases for those genetic differences, and to determine the specific neurochemical and neurophysiological bases for ethanol's actions. Work with these lines has challenged some long-held beliefs about ethanol's mechanisms of action. For example, lines genetically sensitive to one effect of ethanol are not necessarily sensitive to others, which demonstrates that no single set of genes modulates all ethanol effects. LS mice, selected for sensitivity to ethanol anesthesia, are not similarly sensitive to all anesthetic drugs, which demonstrates that all such drugs cannot have a common mechanism of action. On the other hand, WSP mice, genetically susceptible to the development of severe ethanol withdrawal, show a similar predisposition to diazepam and phenobarbital withdrawal, which suggests that there may be a common set of genes underlying drug dependencies. Studies with these models have also revealed important new directions for future mechanism-oriented research. Several studies implicate brain gamma-aminobutyric acid and dopamine systems as potentially important mediators of susceptibility to alcohol intoxication. The stability of the genetic animal models across laboratories and generations will continue to increase their power as analytic tools.  相似文献

This paper proposes an Average Recession/Recovery Model (ARRM). It discusses the reasons why such a model is needed and how it can be developed. Major utilities of the model are (1) it is an extremely simple way to display complex forecasts to management graphically, (2) it can be applied to a single time series–with monthly or quarterly data–and is applicable to a large proportion of time series, both macro and micro, (3) it is easy to calculate, to understand, to program and to communicate with it and (4) it employs widely accepted statistical and economic methods.  相似文献

In contrast to antibodies, which recognize antigens in native form, αβ T cell receptors (TCRs) only recognize antigens as peptide fragments bound to MHC molecules, a feature known as MHC restriction. The mechanism by which MHC restriction is imposed on the TCR repertoire is an unsolved problem that has generated considerable debate. Two principal models have been advanced to explain TCR bias for MHC. According to the germline model, MHC restriction is intrinsic to TCR structure because TCR and MHC molecules have co-evolved to conserve germline-encoded TCR sequences with the ability to bind MHC, while eliminating TCR sequences lacking MHC reactivity. According to the selection model, MHC restriction is not intrinsic to TCR structure, but is imposed by the CD4 and CD8 co-receptors that promote signaling by delivering the Src tyrosine kinase Lck to TCR–MHC complexes through co-receptor binding to MHC during positive selection. Here, we review the evidence for and against each model and conclude that both contribute to determining TCR specificity, although their relative contributions remain to be defined. Thus, TCR bias for MHC reflects not only germline-encoded TCR–MHC interactions but also the requirement to form a ternary complex with the CD4 or CD8 co-receptor that is geometrically competent to deliver a maturation signal to double-positive thymocytes during T cell selection.  相似文献

Philosophers of science continue to elaborate our understanding of the roles that values play in scientific reasoning, practice, and institutions. This special issue focuses on the environmental sciences, a mosaic of fields ranging from restoration ecology to forestry to climatology, unified by its attention to the relationships between humans and their habitats. It is a field that revolves around ameliorating environmental problems, aiming to support the provision of social goods and provide guidance to policymakers about how to regulate individuals and industries. Values abound in such judgments as setting the boundaries of an ecosystem, integrating the human dimensions of social-ecological systems, and collaborating with stakeholders. Since few in the field are likely to insist that these judgments can be made without reference to social values, environmental science can serve as fertile ground for exploring the ethical, social, and political terrain at the frontier of the science and values discourse.  相似文献

Plants often encounter unfavorable environmental conditions because of their sessile lifestyle. These adverse factors greatly affect the geographic distribution of plants, as well as their growth and productivity. Drought stress is one of the premier limitations to global agricultural production due to the complexity of the water-limiting environment and changing climate. Plants have evolved a series of mechanisms at the morphological, physiological, biochemical, cellular, and molecular levels to overcome water deficit or drought stress conditions. The drought resistance of plants can be divided into four basic types-drought avoidance, drought tolerance, drought escape, and drought recovery. Various drought-related traits, including root traits, leaf traits, osmotic adjustment capabilities, water potential, ABA content, and stability of the cell membrane, have been used as indicators to evaluate the drought resistance of plants. In the last decade, scientists have investigated the genetic and molecular mechanisms of drought resistance to enhance the drought resistance of various crops, and significant progress has been made with regard to drought avoidance and drought tolerance. With increasing knowledge to comprehensively decipher the complicated mechanisms of drought resistance in model plants, it still remains an enormous challenge to develop water-saving and drought-resistant crops to cope with the water shortage and increasing demand for food production in the future.  相似文献
