A simple staining technique for electron microscopy with lead-uranyl acetate

Resumen Mediante el empleo de una solución saturada de acetato de uranilo-plomo en acetona de 70%, durante el proceso convencional de deshidratación, se ha conseguido incrementar marcadamente el contraste electrónico en ápices radicales dePhalaris canariensis, sin riesgo alguno de contaminación o deformación de las estructuras.     

Immunohistochemical localization by electron microscopy of GnRH in the median eminence of Xenopus laevis daud]

Using antibodies against mammalian LHRH with the unlabeled antibody enzyme method immunoreactive nerve fibres were localized in the median eminence of Xenopus laevis at the electron microscopic level. These fibres contain irregularly shaped neurosecretory granules with an average diameter of 890 A.     

A reliable method for obtaining longitudinal sections of sperms for electron microscopy

Summary A method is presented by which it is possible to obtain a high percentage of longitudinal sections through sperms for electron microscopy. The procedure is described in detail in order to permit easy reproduction.The technical help of Miss A. K. Baer is gratefully acknowledged.     

In vitro and in vivo interaction of nuclear antibodies with corresponding antigens

Zusammenfassung Es wird mit indirekter Immunofluoreszenz gezeigt, dass Leberzellkerne mit Antikörpern bedeckt werden können, wenn sie systemischemLupus erythromatosus-Serum (SLE) für weniger als 2 sec ausgesetzt werden, und dass die Färbungsintensität vom Titer des Antikörpers abhängt.Diese Ergebnisse deuten darauf hin, dass das in SLE-Geweben durch direkte Immunofluoreszenz nachgewiesene nukleare-Globulin hauptsächlich nukleare post-Biopsiereaktionen wiederspiegelt und weniger in vivo-Reaktionen, und dass in beiden Fällen nur abgestorbene oder nicht intakte Zellen vom Antikörper durchdrungen werden.

Localization of concansvalin-A binding sites in Tetrahymena by scanning electron microscopy.

Binding sites of concanavalin-A were detected in Tetrahymena on the body ciliature, while the cell membrane itself and the oral ciliature failed to bind the lectine.     

Fixation for electron microscopy and the retention of3H-noradrenaline by tissues

Résumé Le dégagement de la radioactivité des tissus qui contiennent³H-NA et fixés pour la microscopie électronique est influencé par le genre des tissus (cur, rate, mésentère, canal déférent) et par le fixatif utilisé (osmium tétroxyde, glutaraldéhyde-formaldéhyde, et permanganate de potassium). Les meilleurs résultats sont obtenus par la combinaison glutaraldéhyde-formaldéhyde, car les proportions de la radioactivité conservées dans ces tissus fixés sont similaires aux proportions de³H-NA dans les mêmes tissus non-fixés.
