The role of anti-tumor immune responses in oncolytic adenoviral therapy has not been well studied due to lack of efficacious tu- mor model in immunocompetent mice.Here,we evaluated the contributions of immune components to the therapeutic effects of oncolytic adenoviruse in an immunocompetent murine tumor model permissive for infection and replication of adenovirus.We found that CD8+T cells were critical mediator for antitumor efficacy by oncolytic adenovirus.Intratumoral viral therapy induced intensive infiltration of CD8+T cells in tumor,increased tumor-specific IFN-?(interferon-?)production and CTL(cytotoxic T lymphocyte)activity of lymphocytes,and generated a long-term tumor-specific immune memory.Boosting CD8+T cell responses by agonistic anti-4-1BB(cluster differentiation 137,CD137)antibody showed synergistic anticancer effects with oncolytic viro- therapy.Our results provide insight into antitumor mechanisms of oncolytic adenovirus in addition to their direct oncolytic effect. 相似文献

Export and remineralization of POM in the Southern Ocean and the South China Sea estimated from 210Po/210Pb disequilibria 总被引:1,自引:0,他引:1
Disequilibria between ^210Po and ^210Pb in the upper water and their potential applications as a proxy of particle export and remineralization were examined in the Southern Ocean (station IV3) and the South China Sea (NS44). ^210po was deficit in surface waters but excessive relative to ^210Pb in subsurface waters. Good positive correlation between ^210Po and particulate organic carbon (POC) indicated deficits and excess of ^210Po resulted from particulate organic matter (POM) export and remJneralization respectively, which was also supported by the decreased δ^13C and increased δ^15N downwards as a result of particle remineralization. On the basis of ^210Po/^210Pb box-model, POC export flux out of the surface waters were 1.2 mmol C. m^-2. d^-1 and 2.3 mmol C. m^-2. d^-1 for station NS44 and IV3, respectively. In the subsurface waters, remineralization fluxes of ^210Po were 0.062 Bq. m^-2.d^-1 and 0.566 Bq.m^-2.d^-1 for station NS44 and IV3 along with the recycle efficiency of 52±26% and 119±52%, respectively. Remineralized fluxes of POM derived from ^210Po and exported POC were 0.6 mmol C.m^-2.d^-1 and 2.7 mmol C. m^-2. d^-1 for NS44 and IV3. 为了探讨中药复方对小鼠脾脏淋巴细胞PD-1/PD-L1的表达情况,以及CD3~+CD4~+和CD3~+CD8~+T淋巴细胞比例的影响,采用传统熬制中药的方法得到中药复方提取液,按低、中、高3个浓度对小鼠灌胃给药(1次/d),2周后,采用脑脊髓脱臼法处死小鼠,利用Real-time PCR和流式细胞仪检测脾脏PD-1/PD-L1的表达情况以及淋巴细胞亚型的变化。结果表明,随着中药提取液浓度的增加,脾脏淋巴细胞PD-1的表达显著降低,而PD-L1的表达无显著变化;CD3~+CD4~+型和CD3~+CD8~+型T淋巴细胞的比例均显著增加。所研制的中药复方能抑制淋巴细胞PD-1的表达,激活免疫辅助细胞和效应细胞,有望应用于抗肿瘤的免疫治疗。 相似文献

The presence of several high affinity binding sites on human serum albumin (HSA) makes it a possible target for many drugs. This study is designed to examine the effect of aloe-emodin on HSA by fluo-rescence, CD spectroscopy and molecular modeling. The results of fluorescence measurements sug-gested that the hydrophobic interaction was the predominant intermolecular force stabilizing the AE-HSA complex, which was in good agreement with the result of molecular modeling study. And the enthalpy change ΔH0 and the entropy change ΔS0 were calculated to be -7.041 kJ·mol-1 and 76.619 J·mol-1·K-1 according to the Van't Hoff equation. The alterations of protein secondary structure in the presence of AE in aqueous solution were quantitatively calculated from CD spectra, and the content of α-helices obviously increased. 相似文献

Constitutively expressed Fas ligand (FasL) in several distinct epithelial cell types appears to protect tissues by inducing apoptosis of Fas immune cells during inflammatory reactions. To study the rela-tionship of FasL and inflammation process in cornea,we examined the effects of inflammatory cytokine IL-1β on the FasL production,expression and cytotoxic function in corneal endothelial cells. In this paper,we demonstrate that IL-1β inhibits the FasL production and expression in corneal endothelial cells. The promoter activities of FasL in these cells are reduced by IL-1β in a dose-dependent manner. Finally,we also find that IL-1β block the cytotoxic effects of FasL derived from corneal endothelial cells to the Fas target cells. These data support the view that FasL derived from corneal endothelial cells modulate inflammation within cornea. 相似文献

The purposes of this research were to study the stable expression of exogenous gene encoding therapeutic protein in attenuated Salmonella typhimurium, observe the metabolism of oral gene vaccine carried by attenuated Salmonella typhimurium in BALB/c mouse, and investigate the feasibility of prevention and treatment of tumors by the recombinant bacteria. Recombinant plasmid pcDNA3.1+ VEGFR2(n1-7) was transformed into competent attenuated Salmonella typhimuriurn SL3261 to develop oral DNA vaccine SL3261-pcDNA3.1+VEGFR2(n1-7). To observe whether the exogenous gene can be expressed in the recombinant bacteria, PCR was performed to amplify the CMV promoter of the eukaryotic expression vector as the proof of stable expression of exogenous protein; transmission elec- tron microscopy (TEM) was applied to observe the morphology of the recombinant bacteria to confirm that the exogenous gene has no impact on the growth of the bacteria, and then BALB/c mice were immunized with the gene vaccine. After inoculation of the gene vaccine, the recombinant bacteria SL3261 could be detected in the tissues such as small intestine, colon, liver and spleen. And then, mice in each group were challenged with tumor cells. The results of animal experiment showed that tumor growth of the mice in experimental group was inhibited and survival time of immunized mice was prolonged compared with control groups. A higher lymphocyte infiltration in tumors from animals treated with DNA vaccine was observed. Immunohistochemical analysis of tumor samples revealed an enhanced accumulation of CD8^+ cytotoxic T lymphocytes, as well as an increase in CD4^+ cells in the tumore of animals treated with the oral gene vaccine compared to tumors from control group mice. UI- trestructure of the tumor tissue showed that tumor cells in the samples of the immunized mice were well-differentiated. The nanoparticles (NPs) from polyvinyl butyrate (PVBu) which can work as an orally applicable donor of butyrate for intestine were prepared. Immunotolerance inducing molecules such as vitamin D3 and all-trans retinoic acid (ATRA) were incorporated into PVBu NPs. The alteration in populations and numbers of DC103+ dendritic cells (DC) and regulatory T (Treg) cells in intestinal immune tissues were examined after oral administration of NPs. It was found that NPs reduced the population of CD103+ DC and Treg cells in Peyer’s patched in lower part of the intestine and inversely increased the population of CD103+ DC in mesenteric lymph node (MLN), while the population of Treg cells in MLN was unchanged. These observations indicate that NPs may enhance the immunotolerance at MLN and lamina propria toward luminal antigens, indicating the promising features of PVBu NPs as therapeutics of allergy and autoimmune diseases. 相似文献

The family of cubic Thue equation which depend on two parameters | x^3 + mx^2 y-(m+3) xy^2+y^3|=k is studied. Using rational approximation, we give a smaller upper bound of the solution of the equation, that is quite better than the present result. Moreover, we study two inequalities | x^3 + mx^2y-(m + 3) xy^2+y^3 | =k≤2m+3 and |x^3 +mx^2y- (m+3)xy^2 + y^3| = k≤ (2m+3)^2 separately. A re-utilizable piezoelectric crystal (PC) immunosensor for thyroidal triiodothyronine (T3) has been developed. The crystal
was coated with protein A and then reacted with T3 antibody. T3 could be detected in the range of 0.5×10−6 g/L to 10.0×10−6g/L by the, system. Crystal was regenerated by saturating with T3 and subsequent binding of a new anti-T3 antibody layer.
The T3 assay could be repeated up to 5 times using each crystal.

The recombinant expression vector pGEMD-fhit which contains full encoding region offhit gene was constructed. The recombinant was introduced into the BL21 (DE3) strain ofE. coli and induced by 1 mmol/L IPTG to express a 29×103 polypeptide offhit fusion protein. The pollen flux in coniferous and deciduous broadleaved mixed forest in the Changbai Mountains is presented in one year's experiments. The results indicate that arboreal pollen percentages are more than 65% and pollen flux is higher than 5000 grain·cm-2·a-1,while less than 2% and lower than 1000 grain·cm-2·a-1 for shrubby pollen,and less than 20% and lower than 3000 grain·cm-1·a-1 for herbaceous pollen for most samples at different heights. The pollen assemblages are similar to those in the samples under 8 m height where Pinus and Ouercus are dominant and followed by the few non-local pollen types,and Fraxinus percentages are high with a few non-local pollen types at 16 m to 32 m height as well as non-local pollen clearly increased at 40 m height. Comparisons between pollen assemblages and vegetation composition suggest that similarities are higher for pollen trap samples than for surface moss samples. 相似文献

探讨4种镉盐对lewis肺癌的作用,以及锌与镉联合应用时锌的保护机制.建立lewis肺癌小鼠模型,随机分成不同的给药组.16 d后检测瘤重、胸腺指数、脾脏指数以及肝脏和肿瘤中的金属硫蛋白含量.氯化镉组对lewis肺癌抑制作用明显并可以增强荷瘤小鼠的免疫功能.1 mg·kg-1氯化镉组与联合应用组相比肿瘤抑制率无显著性差异,而且后者小鼠肝脏中MT表达量显著增加.结果表明氯化镉可有效抑制lewis肺癌细胞的增殖,最佳抑制剂量为1 mg·kg-1;在联合应用组中,氯化锌在不影响镉抗lewis肺癌效果的同时还能够进一步减轻镉对机体正常组织的毒性作用. 相似文献
