应用柱层析和凝胶过滤的方法,从中华眼镜蛇的粗毒中分离纯化出一个不含磷脂酶A2(PLA2)的膜毒素(Membranetoxin,MT)组分.MT经电泳鉴定为单一成分.将qdMT与鼠抗人鼻咽癌细胞(CNE2)的单克隆抗体(BAC5)偶联制备成免疫毒素(BAC5-MT),经间接免疫荧光检测,证实免疫毒素仍保留了很好的生物活性.体外抗癌实验证明,在无Ca2+培养基中,游离的MT对CNE2和S180细胞均有快速杀伤作用,并且对2种细胞的杀伤作用没有明显的区别,但BAC5-MT同样在无Ca2+条件下,仅对靶细胞CNE2有强烈的杀伤作用(杀伤率达78.6%),对S180细胞只有很弱的杀伤作用(杀伤率为21.3%).结果表明BAC5-MT对CNE2细胞有很好的选择性杀伤作用,提示蛇膜毒素在肿瘤的导向治疗中将是一条很有希望的途径.  相似文献

分离兔角膜上皮和皮肤表皮,提取水溶性蛋白质,采用SDS-PAGE以及IEF/SDS-PAGE技术对其组成进行分析研究,发现兔角膜上皮存在5种主要特异性水溶性蛋白质,其分子量和等电点分别为:Mr68200,pI7.5;Mr61700,pI5.1;Mr51300,pI7.6;Mr36300,pI8.9和Mr25700,pI5.7。  相似文献

采用喷射沉积坯轧制和RS/PM粉末挤压坯轧制的2种工艺制各了Al-8.5Fe-1.3V-1.7Si和Al-8.5Fe-1.3W-1.7Si合金的板材,并通过透射电镜,扫描电镜和力学拉伸实验研究了板材的组织性能.结果表明,综合性能AlFeWSi合金优于AlFeVSi合金的喷射沉积坯轧制板材.  相似文献

采用单克隆抗体作为试剂,建立了直接用于快速检测粪便中福氏痢疾杆菌的一种新方法即固相致敏红细胞吸附技术。检测210份疑以细菌性痢疾患者粪便标本的结果证明,用常规培养法检测阳性者,SPASE法检测阳阳性,两类方法的条例率为81.9%;常规培养法阳性率43.3%而SPASE法则为61.5%。SPASE法全过程仅需2.5-3小时,可检出10^3-10^4个/ml细菌,这快速免疫法不仅能检出完整的细菌,而且  相似文献

分别将编码牛肝微粒体细胞色素b5水溶性结构域片段A1a7-Arg88和Ser5-Ser97基因在大肠杆菌JM83中高效表达,分离纯化后得到Tb5和Lb52种重组野生型蛋白。用核磁共振、紫外可见光谱、圆二色散光谱、循环伏安等方法研究结果表明,Lb5肽链2端残基未能使蛋白质的血红素周围环境与Tb5蛋白质N末端的Ser5和Lys62个残基所贡献。  相似文献

论文提出了以2,3-二氨基萘(DAN)为荧光试剂,用高效液相色谱-荧光检测系统(HPLC—FLD)测定痕量硒(Se)的方法.对DAN-Se化合物的形成和萃取条件和HPLC—FLD分高测定条件进行了系统的研究.在实验条件下,DAN—Se的保留时间为1.7min,检测限为5.3ng,测定范围5~200ng.并应用该方法测定了几种常见的粮食中的含硒量,标准偏差为3.0~6.5ng,回收率为88.8~104.0%.  相似文献

Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
