Summary The development of Reissner's fibre (RF) by secretory activity of the flexural and subcommissural organ was studied in early ontogenetical stages ofTriturus alpestris. In young stages RF arises from the flexural organ, which gradually reduces its secretory activity in the following stages. Now the secretion of the subcommissural organ becomes predominant and begins to form the RF. After the hatching of larvae, only the subcommissural organ produces the RF.     

Lack of effect of neurotransmitters on cyclic AMP phosphodiesterase activity in an insect CNS

Summary The inhibition of cyclic nucleotide phosphodiesterase from the nerve cord ofManduca sexta was studied using theophylline as a model compound. 11 putative neurotransmitters had no effect on enzyme activity.Supported in part by NIH grant NS-09161-20 to R. W. Newburgh     

Increased urinary excretion of cyclic nucleotides in X-linked hypophosphatemic (Hyp) mice

Summary Hyp mice, a model for human X-linked hypophosphatemia, had elevated urinary cyclic AMP, cyclic GMP, and magnesium excretion compared to normal mice. The data suggest a renal origin of the urinary cyclic nucleotides. No significant differences in plasma cyclic AMP and cyclic GMP were observed between genotypes.This work was supported in part by research grants from The Kroc Foundation and the NIH, RR-09016.     

Radiochemical assays of corpus allatum activity in adult female cockroaches following ovariectomy in the last nymphal instar

Summary Surgical removal of the ovaries from nymphalPeriplaneta americana results in lower than normal corpus allatum activity in the adult insect and an apparent absence of the manifestly cyclic pattern of juvenile hormone biosynthesis found in intact mated females. The results suggest that the presence of synchronously developing ovaries is necessary for the attainment of normal synthetic activity in the corpus allatum of this species.Acknowledgment. Hormone and forskolin-stimulated cyclic AMP accumulation in human lymphocytes: reliability of longitudinal time measurements

Summary Reliability of measurement of lymphocyte cyclic AMP synthesis in intact cells was estimated by taking 3 successive blood samples during a one-month period from 11 healthy volunteers. Isoproterenol and prostaglandin E₁-stimulated cyclic AMP accumulation were used to evaluate the activity of these two receptor activities in human lymphocytes. Forskolin-stimulated cyclic AMP accumulation was used to evaluate the activity of the Ns/catalytic subunit. Only for forskolin was significant reliability observed. For isoproterenol and prostaglandin E₁ significant reliability was observed only for male subjects.Acknowledgment. Studies on the cytochemical localization of adenylate-cyclase activity inDugesia lugubris s.l.

Summary The localization of adenylate-cyclase activity inDugesia lugubris s.l. has been investigated cytochemically using 5-adenylyl-imidodiphosphate as substrate. The enzyme was localized in mucous gland cells, in rhabdite cells, in intercellular spaces and also in nerve endings of this planarian. The presence of adenylate-cyclase on the membrane suggests that it might mediate different stimulus-secretion coupling by increasing cyclic AMP synthesis in specialized areas of the planarian.Supported in part by a grant from Consiglio Nazionale delle Ricerche, Rome.Acknowledgments. Cyclic AMP level in red blood cells ofPlasmodium berghei-infectedMastomys natalensis

Summary The present report describes the changes in cyclic AMP level which occur upon parasitization of red cells byPlasmodium berghei. Parasitized erythrocytes were separated from the non-parasitized population by percoll density-gradient centrifugation. An increase in the cyclic AMP content of both non-parasitized and parasitized erythrocytes of infected animals compared with that of uninfected animals was observed. The patterns of physiological response to isoproterenol in normal, parasitized and non-parasitized erythrocytes were identical.     

Inhibition of virus multiplication and alteration of cyclic AMP level in cell cultures by flavonoids

Summary The inhibitory effect of four flavonoid compounds on virus multiplication and their influence on the intracellular cyclic AMP (cAMP) level were studied in cell cultures. Quercetin and quercitrin reduced the yields ofHuman (alpha) herpesvirus 1 (HSV-1) andSuid (alpha) herpesvirus 1 (pseudorabies virus), but hesperidin and rutin had no effect. Further, quercetin and quercitrin elevated the intracellular level of cAMP, whereas hesperidin and rutin did not alter the cAMP level. Both antiviral activity and cAMP-enhancing effect were dependent on the concentrations of the flavonoids, and these effects turned out to be parallel.This study suggests that a relation exists between the antiviral effect and the cAMP-enhancing activity of flavonoids.     

Effect of hormones and cyclic AMP on γ-glutamyltranspeptidase activity of rat mammary gland explants

Summary -Glutamyl transpeptidase activity was assayed in midpregnant rat mammary gland explants at 14, 22 and 38 h, in the presence and absence of insulin, prolactin and corticosterone. With these 3 hormones the explants attained the characteristics of a secretory gland after 22 h of tissue culture, at which time the enzyme exhibited its maximal activity. The addition of dibutyryl cyclic AMP in the presence of the 3 hormones produced a significant increase in enzyme activity, which was maximal with a 1 mM concentration of the cyclic nucleotide. A similar effect was observed when theophylline or theophylline plus dibutyryl cyclic AMP were added to the culture medium.This work was supported by grant B1138-8333 from the Departamento de Desarrollo de la Investigación, Universidad de Chile.     

The relationship of phosphodiesterase and cyclic AMP to the process of fluid secretion in the salivary glands of the ixodid tickAmblyomma americanum

Summary Phosphodiesterase (PDE) activity in the salivary glands of the female.Amblyomma americanum decreased as the tick progressed from a slow to a rapid phase of feeding, while the rate of fluid secretion increased, when glands were stimulated with cyclic AMP and theophylline Dopamine stimulated PDE activity and an inhibitory factor was found in glands obtained from rapidly engorging ticks which decreased PDE activity. Two classes of metabolites fromTheonella swinhoei are localized in distinct populations of bacterial symbionts

The marine spongeTheonella swinhoei (lithistid Family Theonellidae, Order Astrophorida) has yielded many important, bioactive natural products, most of which share structural features with bacterial natural products. The presence of microbial symbionts inT. swinhoei has been reported, and it was originally suggested that the cytotoxic macrolide swinholide A and many of the bioactive cyclic peptides fromT. swinhoei were all produced by symbiotic cyanobacteria. By transmission electron microscopy, we found four distinct cell populations to be consistently present inT. swinhoei: eukaryotic sponge cells, unicellular heterotrophic bacteria, unicellular cyanobacteria and filamentous heterotrophic bacteria. Purification and chemical analyses of each cell type showed the macrolide swinholide A to be limited to the mixed population of unicellular heterotrophic bacteria, and an anti-fungal cyclic peptide occurred only in the filamentous heterotrophic bacteria. Contrary to prior speculation, no major metabolites were located in the cyanobacteria or sponge cells.

Influence of cyclic nucleotides on protein synthesis in vascular smooth muscle

Summary The incorporation of leucice-¹⁴C into protein in bovine mesenteric arteries was augmented by cyclic GMP (10^–3 M) and decreased by cyclic AMP (10^–3 M). There was no effect of 5 AMP (10^–3 M). The phosphodiesterase inhibiting drugs theophylline (10^–3 M) and papaverine (5×10^–5 g/ml) both decreased the leucine-¹⁴C incorporation.We are indebted to Mrs.Lena Burlin for hear assistance. Inverse regulation of spore germination and growth by cyclic AMP inStreptomyces hygroscopicus

Summary The cyclic AMP level in germinating spores ofStreptomyces hygroscopicus rises to a maximum at outgrowth of germ tubes. Exogenous cyclic AMP results in an inverse effect on germination speed and growth.     

Suppression of chaos by periodic oscillations in a model for cyclic AMP signalling inDictyostelium cells

We investigate how the introduction of cells oscillating periodically affects the behaviour of a suspension ofDictyostelium discoideum amoebae undergoing chaotic oscillations of cyclic AMP. The analysis of a model indicates that a tiny proportion of periodic cells suffices to transform chaos into periodic oscillations in such suspensions. A similar result is obtained by forcing the aperiodic oscillations by a small-amplitude, periodic input of cyclic AMP. The results provide an explanation for the observation of regular oscillations in suspensions of a putatively chaotic mutant ofDictyostelium discoideum ¹². More generally, the results show how chaos in biological systems may disappear through the coupling with periodic oscillations.

In rat cecal mucosa, Na-K-ATPase specific activity and sodium and fluid absorption were increased by giving polyethylene glycol administration with the drinking water. Whereas cyclic AMP levels were unchanged, cyclic GMP was reduced by about 50%. This finding suggests a regulatory role of cyclic GMP in intestinal sodium and fluid absorption.     

Cortinarius speciosissimus toxins — a preliminary report

Summary Methanolic extracts ofCortinarius speciosissimus yielded a fluorescent compound which was crystallized and shown to be a cyclic polypeptide. The compound, or an analogue, has been found in most members of the genusCortinarius.The authors acknowledge the help of Dr M. Cortinarins inCortinarius speciosissimus? A critical revision

Summary Fluorescent cyclic decapeptides, the so-called cortinarins, have been claimed to cause the toxicity of the deadly poisonous mushroomCortinarius speciosissimus. As the structure elucidation described in the literature is doubtful in decisive parts, the proposed structures must be disputed. The present study demonstrates that the fluorescence is mainly due to ergosta-4,6,8 (14),22-tetraen-3-one (8) and to photodecomposition products of the nephrotoxin orellanine (1). Moreover, we were not able to reproduce the isolation of fluorescent or any other cyclic peptides related to cortinarins fromC. speciosissimus, by various methods.
