Summary Platelets impregnated with heavy metals appeared as 3 distinct morphological types: reticular cells with a polygonal dense tubular network and stained granules, dark metallophilic cells, and pale metallophobic cells with microvesicles and non-staining granules. On stimulation, type 1 cells decreased while type 3 cells increased, suggesting that with activation dense tubules break up into microvesicles and granules become metallophobic. In the type 2 cells a different functional mechanism may exist.  相似文献

The electron dense granules of the matrix were not seen in the micrographs of cartilage treated with collagenase before fixation. The nucleus preopticus (NPO) and the pars lateralis of the nucleus lateralis tuberis (NLT) have been examined at the ultrastructural level. In the NPO two types of large neurons occur, both containing 140-150 nm secretory granules. The pars lateralis of the NLT contains only one cell type with similar granules. Both of these nuclei may be the source of type A fibres innervating the gonadotropic cells of Gambusia.  相似文献

Summary By electron microscopic examination of a solid, chiefly scirrhous mammary cancer, 3 kinds of unusual nuclear inclusions were found: (a) Electron dense particles of a 54–80 nm diameter, whose outlines appear mainly hexagonal, which points at an icosahedral structure, (b) Clusters of granules with a diameter of 200–300å, respectively 300–400å, at whose circumference the larger particles appear, (c) Bundles of filaments in close association and continuous with the granules. In all 3 nuclear inclusions there are subunits of 40–45å. The comparison between these results and experiments published suggest that these nuclear inclusions are (a) virus particles, (b) virus at an early stage of development and (c) virus protein subunits.  相似文献

Summary Secretory granules of extraordinary size, some of them bigger than the cell nucleus, abound in the adrenaline cells of lizard adrenals after metyrapone injections during 7 days. In these granules, the bounding membrane is studded with ribosomes, and the core is formed by rounded small subunits. Some granules of this type are also found in noradrenaline cells. They may represent an exceptionally increased elaboration and storage of adrenaline, induced by metyrapone probably through its action on steroidogenic tissue.This research forms part of project No. 31.26.S1-1154 supported by the Consejo Nacional de Investigaciones Científícas y Tecnológicas.  相似文献

Using the sensitive protein A-gold technique for the demonstration of antigenic sites in thin section for electron microscopy it was found that in the alpha granules of human pancreas, glucagon immunoreactivity (specific, C-terminal) is restricted to the dense granule core while glicentin immunoreactivity predominates on the peripheral halo surrounding the dense core.  相似文献

Serglycin is a proteoglycan found in hematopoietic cells and endothelial cells. It has important functions related to formation of several types of storage granules. In connective tissue mast cells the covalently attached glycosaminoglycan is heparin, whereas mucosal mast cells and activated macrophages contain oversulfated chondroitin sulfate (type E). In mast cells, serglycin interact with histamine, chymase, tryptase and carboxypeptidase, in neutrophils with elastase, in cytotoxic T cells with granzyme B, in endothelial cells with tissue-type plasminogen activator and in macrophages with tumor necrosis factor-α. Serglycin is important for the retention of key inflammatory mediators inside storage granules and secretory vesicles. Serglycin can further modulate the activities of partner molecules in different ways after secretion from activated immune cells, through protection, transport, activation and interactions with substrates or target cells. Summary The lumbar motor columns of a cynomolgus monkey which had become tetraplegic after infection with a highly virulent strain of type 3 poliovirus were examined by electron microscopy. Within the cytoplasm of many endothelial cells of intraspinal blood vessels, as well as within the perikarya of numerous mononuclear inflammatory cells, there occurred regular arrays of dense particles which, on the basis of morphological evidence, could be identified as poliovirus crystals. The possible significance of virual replication by endothelial and mononuclear cells for poliovirus spread within the central nervous system is briefly discussed.  相似文献

Summary The secretory activity of the SCO cells ofLacerta s.sicula Raf. is strongly reduced during the winter. Such reduction is documented by the decrease of the number of secretory granules type A and B described in previous papers in the summer SCO cells. Also the sacks of RER filled with electron-dense material (type C secretion) are very few; in their place there are, in the basal region of the cells, large vacuoles. In the distal region of the cells, at the free cell surface, a pronounced increase in the number of microvilli is noticed.  相似文献

Cytotoxic T lymphocytes patrol our body in search for infected cells which they kill through the release of cytotoxic substances contained in cytotoxic granules. The fusion of cytotoxic granules occurs at a specially formed contact site, the immunological synapse, and is tightly controlled to ensure specificity. In this review, we discuss the contribution of two intracellular compartments, endosomes and cytotoxic granules, to the formation, function and disassembly of the immunological synapse. We highlight a recently proposed sequential process of fusion events at the IS upon target cell recognition. First, recycling endosomes fuse with the plasma membrane to deliver cargo required for the docking of cytotoxic granules. Second, cytotoxic granules arrive and fuse upon docking in a SNARE-dependent manner. Following fusion, membrane components of the cytotoxic granule are retrieved through endocytosis to ensure the fast, efficient serial killing of target cells that is characteristic of cytotoxic T lymphocytes.  相似文献

Human myocardial cells from fresh autopsy material contained granules which possessed hemolytic activity against guinea pig and rabbit erythrocytes. The hemolytic granules, which had a density of 1.02 and a diameter of 200-300 nm, were recovered as a microsome fraction from subcellular homogenates of human myocardial cells by differential centrifugation in 300 mM sucrose containing 0.1 mM PMSF and 10 mM EDTA. The membrane lesions caused by the granules were ring-like structures with an internal diameter of about 10-17 nm, analogous to that caused by perforin- and complement-induced lysis. However, the requirement for divalent cation differed from that for perforin-induced lysis, since the microsome-mediated lysis occurred in the presence of EDTA.  相似文献

Dictyostelium discoideum cells are highly resis tant to xenobiotics. We previously observed that these primitive eukaryotic cells contain a 170-kDa P-glycoprotein, mediating multidrug resistance in mammalian cells, but nonfunctional in Dictyostelium cells. We show here that D. discoideum cells vitally stained with the DNA-specific dye, Hoechst 33342, release fluorescent material in their culture medium. Electron microscopy and lipid analysis demonstrate the vesicular nature of this material. Moreover, nucleic acids associate with these extracellular vesicles independently of Hoechst vital staining. The main vesicular DNA component exhibits a size >21 kb. Shedding of microvesicles during cell growth is not concomitant with programmed cell death. We propose that these extracellular vesicles are involved in a new cellular resistance mechanism against xenobiotics. Furthermore, since the association of DNA with vesicles occurs in physiological growth conditions and independently of vital staining, the new shedding process might be involved in a more general intercellular mechanism.
