Enantiomeric cannabinoids: stereospecificity of psychotropic activity   总被引:1,自引:0,他引:1  
Summary The 1,1-dimethylheptyl homolog of (–)-(3R,4R)-7-hydroxy-delta-6-tetrahydrocannabinol (compoundII) is highly psychotropic in mice, rats and pigeons. The (+)-(3S,4S) enantiomer (III) was found to be psychotropically inactive at doses up to several thousand times those of the ED50 of (II).We thank Dr A. Breuer and Mrs H. Summary Hexoprenaline potentiated the14C-aminopyrine uptake (a reliable index of H+ generation) of isolated rat gastric cells stimulated by 10–6–10–4 mol/l carbachol, and inhibited that in response to 10–4 mol/l histamine without and in the presence of propranolol. It is concluded that hexoprenaline acts as a partial agonist on parietal cell H2-receptors and that -adrenoceptor activation may functionally modualte gastric acid secretion.Acknowledgments. S. Summary The incorporation of leucice-14C into protein in bovine mesenteric arteries was augmented by cyclic GMP (10–3 M) and decreased by cyclic AMP (10–3 M). There was no effect of 5 AMP (10–3 M). The phosphodiesterase inhibiting drugs theophylline (10–3 M) and papaverine (5×10–5 g/ml) both decreased the leucine-14C incorporation.We are indebted to Mrs.Lena Burlin for hear assistance. The mechanisms of HCO 3 and Cl transport across basolateral membranes from rat ileum were investigated in isolated vesicles by means of uptake experiments. Neither Cl/HCO 3 exchanger nor Na+–(HCO 3 )n cotransport seem to be present in ileal basolateral membranes. Moreover Cl uptake is unaffected bycis Na+ and/or K+ gradients, indicating the absence of Na+–Cl, K+–Cl and Na+–K+–2Cl symport activity. An electrically conductive pathway seems to be responsible for both HCO 3 and Cl fluxes. Evidence is also given for the presence of a Na+/H+ exchanger at the basolateral pole of ileal enterocytes.  相似文献

Summary The mechanism of catalase inhibition by CN, N 3 , and SH has been investigated by a polarographic method. CN has been found to inhibit reversibly by a coordinating mechanism, whereas N 3 and SH inhibits catalase irreversibly.  相似文献

Summary A procedure for the synthesis of acetyl phenylalanyl lysine chloromethyl ketone is described. The chloromethyl ketone derivative is a noncompetitive inhibitor of trypsin (K1=5.9×10–3 M).Acknowledgments. The authors are grateful to Dr Arthur L. Bacon for his constant support and encouragement. Summary The viscosity, apparent molar volume and conductivity of NaCl, KCl, NaBr, KBr, NaNO3 and KNO3 solutions at mass fraction of dioxane (10, 20 and 30%)-water mixtures at 30–45°C±0.01°C have been measured. Summary The hydrolysis of acetylcholine chloride (0.01M) by frog's rectus extracts, is inhibited by low concentrations of 3318 CT (CI-50 3.2×10–7) and high concentrations of D.F.P. (CI-50 1.3×10–5). Inversely, the hydrolysis of butyrylcholine perchlorate is inhibited by low concentrations of D.F.P. (CI-50 3×10–9) and high concentrations of 3318 CT (CI-50 3×10–4). Both are inhibited by similar concentrations of neostigmine (CI-50 1.1×10–7 and 1.5×10–7). Frog's rectus thus contains true and pseudo-cholinesterases. The inhibitions produced by D.F.P. added to the muscle itself (and not the extract) correlates well with the potentiation of the corresponding ester. Sensitization to AcCh and to BuCh appears to be specifically related to the inhibition of Ac ChE for the former ester, of XChE for the second one.  相似文献

Enterocytes isolated from rat jejunum were tested for the existence of a Cl/HCO 3 exchange, previously evidenced in basolateral membrane vesicles but not in brush border. Cells were found to retain functional integrity and transport capabilities long enough to allow Cl fluxes to be measured. Both efflux and uptake experiments indicate that a Cl/HCO 3 antiport, inhibited by 4,4-diisothiocyanostilbene-2-2-disulfonic acid (DIDS), is functional under resting conditions.  相似文献

Summary The time course of ATPase activity and force has been determined during contraction and relaxation in skinned (hyperpermeable) anterior byssus retractor muscle, ABRM, ofMytilus edulis and compared with corresponding measurements on skinned taenia coli of guinea-pigs. Following a calcium-induced contraction, lowering the [Ca++] to 10–8 M rapidly reduces ATPase activity within 2 min to resting levels while force declines only to about 30–50% of maximal tension within the same time. Thus slow relaxation is due to a catch-like-state which is common to different kinds of smooth muscles and can be reduced with cAMP in ABRM and by Pi in taenia coli.The support by the Deutsche Forschungsgemeinschaft and the excellent technical assistance of Mrs K. Summary Plasma membrane sheets prepared by zonal centrifugation of a premicrosomal pellet obtained from a rat liver homogenate are devoid of HCO 3 -ATPase activity. Since the microsomal fraction is also lacking in this ATPase activity, it can be concluded that the HCO 3 -ATPase is not involved in the secretion of HCO 3 into bile. Acknowledgments. Waste water purification is characterized by intensive mineralization and nitrification processes. Because of the high O2 demand, temporarily anaerobic conditions may be produced, and denitrification by nitrifying organisms as well as heterotropic denitrification may contribute to N2O release. In situ measurements (1993–1994) suggest that N2O is released from activated sludge in a domestic waste water treatment plant at an average rate of 1040 g m–2h–1 with a range between zero and 6198 g m–2h–1. The production of N2O seems to be related to the concentration of NO 2 and NO 3 as well as to the pH. In the waste water about 75–200 g N2O l–1 is dissolved. This N2O is released after discharge into the receiving waters. The N2O is produced essentially by nitrification rather than by heterotropic denitrification. On a long-term scale the increasing use of mechanical-biological waste water purification plants world-wide may add increasingly to the anthropogenic production of N2O, although the present amount of N2O produced is negligible compared to its global terrestrial production.  相似文献

Résumé Nous avons étudié l'effet de l'iodure 127 (10–3 et 10–4 M) sur la sécrétion de T4 *, T3 * et –I* par la thyroïde de rat, prémarquée à l'iode 131 et perfusée in situ avec du sang non radioactif contenant 12 mU de TSH/ml. Il n'a pas été noté de changement significatif dans la sécrétion de T4 * ou T3 * jusqu'à 45 min après le début de l'administration d'iodure 127, qu'il y ait ou non du méthimazole (10–3 M) dans le sang perfusé. Il y avait par contre une augmentation de la sécrétion d'-I* pendant la perfusion de 10–3 M d'ïodure, indiquant une décharge d'-I* pendant la perfusion de 10–3 M d'ïodure, indiquant une décharge d'-I* de provenance intrathyroïdienne. Cet effet est similaire à celui noté auparavant pendant le perfusion de 10–3 M de ClO 4 .  相似文献

Summary Vasoactive intestinal peptide (VIP, EC50=6.4×10–10 M) and histamine (EC50=3×10–6 M) activated the cyclic AMP generating system in gastric glands isolated from two human fetuses at 23 weeks gestation. Histamine antagonism by the H2 receptor blockers cimetidine (Ki=0.35×10–6 M) and ranitidine (ki=0.51×10–7 M) clearly characterized the histaminic activation as being of the H2 type. It is suggested that these two vasoactive hormones may operate as neurocrine/paracrine regulators of the differentiation and/or function of the human gastric mucosa in utero.  相似文献

Summary Maximal increments in adrenaline and dexamethasone (DXM) plasma concentrations were observed c15 (T50 40 min) and 30 (T50 210–240 min) minutes after an i.v. DXM dose (6 mg/m2 BSA) in man. There appears, however, to be no direct interaction between these agents in the development of induced neutrophilia, which occurs c240 min postinjection.Acknowledgments. Summary Mouse eggs (12–14%) and hamster eggs (87–94%) appear normal upon thawing after having been stored for 30 min to 25 h at –70°C and –196°C. While 89–98% of the normal appearing hamster eggs are capable of fertilization in vitro, only 4–13% of the mouse eggs can be fertilized. The ability of fertilized eggs to develop in culture and their viability upon transfer to pseudopregnant recipients is under investigation.This work was supported by grants from the U.S. Public Health Service and the Ford Foundation.
