本文分析了乙醇酸氧化酶分子量相差很大的原因,但目前认为乙醇酸氧化酶为单一亚基所组成的观点难于解释为何乙醇酸氧化酶具有多个不是等电点,对等电点的正确认识将有助于揭示乙醇酸氧化酶分子结构。  相似文献

菠菜中不同等电点的乙醇酸氧化酶的提取和特性   总被引:1,自引:1,他引:0  
从不同批次的菠菜绿叶中可纯化出5种不同活性和等电点(pI)的乙醇酸氧化酶(CO).SDSPAGE表明它们均有40kD亚基,它们的比活会随蛋白浓度的增加而上升.第一种CO的pI〈8.3,比活为0.47U/mg,PAGE测定其全酶相对分子量(Mr)为280kD,280kD较稳定.第二种CO的pI〈8.3,但比活为3.44U/mg,PACE测定其Mr为560kD,但很快解聚为280kD.解释了前人报告CO的Mr大多为280kD.第三种CO的pI≈8.3,比活为8.4U/mg,放置后pI会下降至〈8.3,此时用PAGE测出其Mr为560kD.第四种CO的pI〉8.3,比活为27.8U/mg,放置pI也会下降为8.3.第五种CO的pI〉10.2,活性高达113U/mg,放置后其pI也会下降至〈8.3,由于CO的pI会下降,暗示低pI的CO可能并不存在于植物体内,而是实验中的娇作物.  相似文献

通过缩短DEAE-celluloses柱长度,提高其洗脱液pH,可快速地从菠菜绿叶中纯化得到乙醇酸氧化酶同工酶,证实其具高活性113.0 U/mg·min和高等电点(pI)10.6.纯化后产率为0.14%,纯化倍数为66.5.经SDS-PAGE和毛细管电泳(CE-SDS)均只有40kD单带,说明已纯化至电泳纯.  相似文献

作者在之前研究中已证实提取的乙醇酸氧化酶(GO)实际上是GO和富phe碱性蛋白(BP)的混合物,BP因在SDS-PAGE中向负极泳动而容易被忽略.该文再次提取比活为113.4 U/(min.mg)的GO和BP的混合物.该混合物呈现phe的258 nm特征吸收峰,表明BP可能位于GO外部.类似的BP也可能存在于拟南芥、水稻等模式植物和大肠杆菌的粗蛋白中.  相似文献

通过PCR从甘蓝型油菜(Brassica napus)华双4号基因组DNA中扩增出乙醇酸氧化酶(GO)基因片段,DNA序列分析表明扩增GO基因片段的外显子部分与报道序列相同.以扩增出的GO基因片段作模板从一端设计引物扩增出一个相应的小片段.将GO基因大小两个片段反向连接,插入到植物表达载体2300-nap的napin启动子和nos终止子之间,植物表达载体2300-35S的35S启动子和nos终止子之间,分别构建成可转录表达出发夹RNA(hairpin RNA,hpRNA)结构的种子特异型和组成型油菜hpRNA干扰载体.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
