Effect of tea consumption on the levels ofα-ketoglutarate and pyruvate dehydrogenase in rat brain

Summary Administration of tea to rats fed on a normal diet results in a marked drop in brain levels of total thiamine as well as of -ketoglutarate and pyruvate dehydrogenase activities. The patterns of decrease in both enzyme activities are similar to that of total thiamine content; they drop to about 65% of the control at 14–20 weeks after continuous consumption of tea.Supported by a grant from the International Foundation for Science (IFS), Sweden.     

Interferon-β can induce the production of plasminogen activator by cultured human cancer cells

Summary Three cultured human cell lines, renal cancer cells (ACHN), bladder cancer cells (EJ), and fibroblasts transformed in culture by Co-60 gamma rays (KMST-6), when treated with interferon-, produced 1.5 to 4 times as much plasminogen activator as the untreated control cultures. This enhanced production of PA was inhibited by cycloheximide or actinomycin D.     

Sertoli cells of adult rats in vitro. II. Effect of different steroid precursors on estradiol 17β-synthesis

Summary The estradiol 17-synthesis by ⁴ pathway has been studied in homogenous cultures of Sertoli cells isolated from adult rat testes. The effect of αB-crystallin and Hsp27 on the availability of translation initiation factors in heat-shocked cells

The mechanism of the translational thermotolerance provided by the small heat shock proteins (sHsps) αB-crystallin or Hsp27 is unknown. We show here that Hsp27, but not αB-crystallin, increased the pool of mobile stress granule-associated enhanced green fluorescent protein (EGFP)-eukaryotic translation initiation factor (eIF)4E in heat-shocked cells, as determined by fluorescence recovery after photobleaching. Hsp27 also partially prevented the sharp decrease in the pool of mobile cytoplasmic EGFP-eIF4G. sHsps did not prevent the phosphorylation of eIF2α by a heat shock, but promoted dephosphorylation during recovery. Expression of the C-terminal fragment of GADD34, which causes constitutive dephosphorylation of eIF2α, fully compensated for the stimulatory effect of αB-crystallin on protein synthesis in heat-shocked cells, but only partially for that of Hsp27. Our data show that sHsps do not prevent the inhibition of protein synthesis upon heat shock, but restore translation more rapidly by promoting the dephosphorylation of eIF2α and, in the case of Hsp27, the availability of eIF4E and eIF4G. Effect of acute administration of Δ1-tetrahydrocannabinol on β-endorphin levels in plasma and brain tissue of the rat

Summary Acute treatment with ¹-tetrahydrocannabinol (¹-THC) elevated the concentration of -endorphin-like immunoreactivity (-ELIR) in plasma and in the hypothalamus, but not in the hippocampus of rats habituated to the injection procedure. These effects were not obtained with the psychotropically inert analog of (¹-THC), cannabidiol. In animals that had not been habituated to the injection procedure, placebo treatment induced a decrease in hippocampal -ELIR.The authors acknowledge the skillful technical assistance of Mrs Willeke Logtenberg.     

Effect of spike disease on the 5′-and 3′-nucleotidase activities in sandal plants

Summary In the sandal plant affected by spike disease, caused by mycoplasma-like organisms, the changes occurring in the 5-and 3-nucleotidase activities were studied and their possible significance discussed.     

The effect of ischæmia on nucleoproteins in liver cells

Résumé Chez la souris, 24 heures après la ligature du pédicule d'un lobe hépatique, la basophilie cytoplasmique disparaît alors que les noyaux conservent une colorabilité normale par la méthode deFeulgen. Les dosages chimiques établissent que le taux de l'acide ribonucléique s'abaisse considérablement, tandis que celui de l'acide désoxyribonucléique ne subit pas de modifications appréciables. Ces phénomènes sont liés à la destruction des granules et microsomes séparables par ultracentrifugation. Une partie de ces nucléoprotéines se retrouve, après ultracentrifugations répétées, dans le liquide surnageant.     

Effect of prior splenectomy on the growth of Sarcoma 180 in normal and Bacillus Calmette-Guérin infected mice

Zusammenfassung Die Regression des Sarkomas 180 in Albinomäusen wurde durch vorangehende Splenektomie gefördert. Die gegen Sarkoma 180 erhöhte Resistenz von Mäusen, die mit B.C.G. infiziert worden waren, wird durch die Abwesenheit der Milz nicht beeinflusst.

Effect of an ecdysteroid on DNA synthesis,DNA polymerase α and thymidine kinase activities ofDrosophila melanogaster cells in vitro

Summary 10^–7 M 20-OH-ecdysone treatment of a diploidDrosophila clone results in an inhibition of 60% of the DNA synthesis from 18 h of treatment on. After 48 h of hormone treatment the thymidine kinase activity is 70% inhibited; concomitantly a 60–70% lowering of the acid-soluble specific activity is observed. In the meantime the DNA polymerase activity is reduced.     

Effect of testosterone and 17-β estradiol on limb regeneration in the newt,Notophthalmus viridescens

Summary Gonadectomy, or injections of testosterone or 17- estradiol, had no apparent effect on the rate of regeneration or histological appearance of limb regenerates in the newt,Notophthalmus viridescens. Neither promotion, nor inhibition of limb regeneration was observed.This work was supported by a grant from the National Research Council of Canada to S.R.S.

Für das Auftreten einer ausgeprägten biologischen Aktivität scheint die Anwesenheit einerN-Acetyl-Gruppe an dem Aminoende der Peptidkette wichtiger zu sein als das Vorhandensein der zwei ersten amino-endständigen Aminosäurereste der-MSH-Kette.     

Dynamics of the interaction of γδ T cells with their neighbors in vivo

γδ T cells are a diverse component of the immune system in humans and mice with presumably important but still largely unknown functions. Understanding the dynamic interaction of γδ T cells with their neighbors should help to understand their physiological role. This review addresses recent advances and strategies to visualize the dynamic interactions of γδ T cells with their neighbors in vivo. Current knowledge regarding the dynamic contacts of tissue resident γδ T cells and epithelial cells, but also of the communication between circulating γδ T cells and DCs, monocytes and FoxP3⁺ regulatory T cells is revisited with emphasis on the role of γδ T cell motility.     

The influence of N-methyl-N-β-chloroethyl hydrazines on the mitotic index of Ehrlich ascites tumor cells and the leukopoiesis of the mouse

Summary The cytostatic activity of N-methyl-N--chloroethylbenzaldehyd hydrazone, (B1) is at least equal to that of procarbazine when its effect is tested with the Ehrlich ascites tumor cells of the mouse and the Yoshida sarcoma of the rat. B1 causes a slighter decrease of mitotic cells and no shift from prophase to metaphase. These results suggest that the cytostatic effect of B1 is due to interference with cell metabolism or an effect at the cell membrane and not to an effect on cell proliferation. This assumption is supported by a considerable depression, of lymphocytes and a minor effect on granulopoiesis, which is especially sensitive towards proliferation toxins. All these findings suggest a different mechanism of action of B1 and procarbazine.     

Effects of duration of gonadectomy,sex and age on adrenal steroid 5α-reductase activity in the rat

Summary Within 6–8 weeks, gonadectomy in male and female rats leads to an increase in adrenal 5-steroid reductase activity. However, long-term post-orchiectomy enzyme activity decreases to the control level and this effect is not related to the age of animals.Supported in part by grant No.564/VI from Polish Academy of Sciences. Involvement of xanthine oxidase and hypoxia-inducible factor 1 in Toll-like receptor 7/8-mediated activation of caspase 1 and interleukin-1β

Inflammatory reactions to ssRNA viruses are induced by the endosomal Toll-like receptors (TLRs) 7 and 8. TLR7/8-mediated inflammatory reaction results in activation of the Nalp3 inflammasome via an unknown mechanism. Here we report for the first time that TLR7/8 mediate activation of xanthine oxidase (XOD) in an HIF-1α-dependent manner. XOD produces uric acid and reactive oxygen species, which could activate Nalp3 and therefore induce activation of caspase 1, known to convert inactive pro-IL-1β into active IL-1β. Specific inhibition of the XOD activity attenuates TLR7/8-mediated activation of caspase 1 and IL-1β release. These results were obtained using human THP-1 myeloid macrophages. The findings were verified by conducting in vivo experiments on mice.     

Characterization of 17β-estradiol 3-(β-D-glucopyranoside) and 17-(α-D-glucopyranoside) as the metabolites of 17β-estradiol in the cultured ovaries of the silkworm,Bombyx mori

Summary The structures of the metabolites formed upon incubation of 17-estradiol with the ovaries of silkworm,Bombyx mori, have been determined as 17-estradiol 3-(-D-glucopyranoside) (1) and 17-(-D-glucopyranoside) (2) by spectroscopic means.     

Effect of dexamethasone on Fc γ receptor expression in foetal and neonatal rat gut

Summary When injected into 12-day-old suckling rats, dexamethasone caused a precocious disappearance of Fc receptors from enterocytes of the proximal small intestine. However, dexamethasone appeared to be necessary for the maintenance or production of such receptors in foetal rat gut cultured in vitro.Supported by an award from the Science and Engineering Research Council.     

Isotope effect studies on the dehydrogenation of Δ1-tetrahydrocannabinol in the rat

Summary Isotope effect studies on the metabolic dehydrogenation of ¹-tetrahydrocannabinol in rats are described and its is shown that this process is confined to a very short period following i.v. administration.
